The yeast-mycelium dimorphism of the genus Benjaminiella poitrasii has been investigated. To understand the mechanism of dimorphism two stable yeast-phase mutants (Y-1 & Y-2) and one slow growing mycelial mutant (M-1) of B. poitrasii were isolated after NTG treatment of parent strain spores and studied for their biochemical characteristics. Effects of (i) kind and concentration of carbon source, (ii) presence of complex organic nitrogen and (iii) C:N ratio in the growth medium on the morphology of parent and mutant strains were carried out at 28 degrees C under shaking conditions. Ethanol induced morphological change and its reversal were studied in all the strains in order to elucidate the possible mechanism of morphogenesis.

In order to develop a process for production of dodecanedioic acid (DC-12) as starting materials for the chemical industry, a n-dodecane assimilating and DC-12-producing yeast was isolated from Taiwan soil. The taxonomical characteristics of this newly isolated yeast were examined and it was identified as Candida tropicalis NTU-512. The newly isolated strain was improved successively by NTG mutagenesis. A high potency DC-12-producing mutant 91 which showed slight growth both in DC-12 and n-dodecane enrichment media was isolated. The DC-12 productivity of mutant 91 reached 3,326 mg/I by shaking culture at 30 degrees C for 72 h. This figure is 2.5-fold that of the parent strain (1,310 mg/I).

Three types of colony, powdery gray, white and bald, were isolated from Streptomyces avermitilis ATCC31272. Among them, only the powdery grey one produces avermectins. Sa-76 strain was selected from the powdery grey strain by mutation with high energy electric flow, and its avermectins titer attainde 100 micrograms/mL in shaking flask. Avermectin B1 was extracted and purified from the mycelia of Sa-76, and identified by UV, IR, 1H-NMR, 13C-NMR and mass spectra. After Sa-76 strain was treated twice with NTG, a strain named Sa-76-8 was selected with avermectins titer over 2000 micrograms/mL. The Sa-76-8 strain was treated with NTG once again, and a high avermectins producing strain named as Sa-76-9 with avermectins titer up to 3500-4000 micrograms/mL was selected.

A xylanase producer WXULI-11 was isolated and identified as Pseudomonas sp. By combination treatment of WXULI-11 with UV and NTG, a mutant WLUN024 was obtained and its enzyme activity reached 354 IU/mL in 36 h by shaking flask. The mechanism of its xylanase production was primarily studied. The results showed that this enzyme was induced by xylan, xylose and some other xylosic material, while xylose had "two-way effect" on xylanase synthesis. Effects of nitrogen and other factors on enzyme production were also investigated.

NTG was used to make chemical mutation for Bacillus sphaericus, RifR and SmR labelled strains were selected, which could resist drug as much as 100 u/ml. The resistance to drug was stably inherited. The RifR strain was used as recipient and the plasmid containing the lysostaphin gene was transfered into it by protoplasts. Results showed that the lysostaphin gene could be expressed stably at high level in Bacillus sphaericus and the lysostaphin activity was about 122 u/ml medium after shaking culture.

A strain designated MK107-39, producing kojic acid with a high yield, was obtained by a new screening method using a 96-well microtiter plate after NTG treatment of Aspergillus oryze ATCC 22788. The amount of kojic acid produced by strain MK107-39 in a shaking flask was 28 g/l from 100 g/l of glucose, which was 7.7-times higher than that produced by parent strain. The kojic acid yields per cell and the amount of glucose consumed were 9.8 and 6.0-times higher than those of the parent strain. Based on differences in the use of carbohydrates and organic acids, it seems that strain MK107-39 has some mutation regarding carbohydrate metabolism. By customizing the medium and culture conditions such as glucose concentration, dissolved oxygen concentration and pH of the fermentation broth, more than 110 g/l of kojic acid was produced in a 3-l jar fermentor. Upon scale up to a 600-l pilot fermentor, enhanced production of kojic acid was successfully achieved. The kojic acid yield from glucose consumed, Y(P/S), was 0.43 (g/g) in this pilot plant-scale fermentation.
