Freeze dried and finely ground leaves of two plants with known antimicrobial activity, Anthocleista grandiflora and Combretum erythrophyllum were extracted with acetone, ethanol, methanol, methylenedichloride, methanol/
chloroform
/water and water at a 1 to 10 ratio in each case. The quantity and diversity of compounds extracted, number of inhibitors extracted, rate of extraction, toxicity in a bioassay, ease of removal of solvent and biological hazard were evaluated for each extractant. An arbitrary scoring system was developed to evaluate the above parameters for the different extractants. Acetone gave the best results with these plants with an arbitrary value of 102 followed by methanol/
chloroform
/water (81), methylene dichloride (79), methanol (71), ethanol (58) and water (47). Four five minute sequential extractions of very finely ground A. grandiflora
shaking
at a high rate extracted 97% of the total antimicrobial activity.

A flow injection Fourier transform infrared spectrometric procedure has been developed for the determination the dithiocarbamate pesticides Ziram and Thiram in solid samples. All the operations involved, such as extraction, filtration and measurement, were integrated in the experimental set-up in order to avoid excessive manipulation of samples and standards. Ultrasonic assisted and mechanical extraction were evaluated for the solubilization of the analytes and, additionally, the effect of carrier flow rate, sample loop volume and the ratio between sample mass and volume of solvent employed were studied. Quantitative extractions with
chloroform
were obtained for both Ziram and Thiram, after 5 and 2 min, respectively, of mechanical
shaking
of sample slurries. Absorbance measurement, in the wavenumber range of 1600-1460 cm-1 for Ziram and 1400-1315 cm-1 for Thiram, was carried out, and the area values of the peaks obtained, as a function of time, were interpolated in external calibration lines prepared from standard solutions of Ziram and Thiram in
chloroform
. Analyses of commercial formulations and spiked soil samples incubated two weeks were in a good agreement with values found by other methodologies. Absolute detection limits of 400 micrograms for Ziram and 785 micrograms for Thiram and variation coefficients of 6.4% and 2.5% were obtained by use of the aforementioned methodology.

The synthesis of novel acetal thia-cage compounds has been accomplished by the direct substitution for the oxygen atom by the sulfur atom in the reaction of the acetal groups of oxa-cages with Lawesson reagent (LR). Reaction of the tetraoxa-cage compound 2 with LR in dichloromethane at 25 degrees C sequentially gave the monothia-, dithia-, trithia-, and tetrathia-cage compounds 3, 6, 7, and 9. The reaction mechanism for the conversion from oxa-cages into thia-cages was proposed. The diacetal trioxa-cages 18-20 and 24-26 were also transformed into the thia-cages 21-23 and 27-29, respectively. Reaction of the trioxa-cages 34 and 35 with LR under the same reaction conditions gave the thia-cages 36 and 37 with the carbonyl group intact. Treatment of the pentaoxa[5]peristylane 40 with LR in
chloroform
under supersonic

at refluxing temperature gave the monothia[5]peristylane 41 and the dithia[5]peristylane 42. A simple and rapid liquid-liquid extraction of palladium has been studied involving ion-pairing of bromocomplexes of palladium(II) with hexadecylpyridinium bromide (HDPB) dissolved in
chloroform
. The stoichiometry and distribution of (HDP)2PdBr4 between the aqueous and organic phase was investigated by spectrophotometric mole ratio method. The extraction efficiency of palladium(II) by HDPB was studied as a function of several variables: acid, salt, surfactant concentration and equilibrium time. The results showed that PdBr4(2-) extraction could be explained by assuming the formation of (HDP)2PdBr4 complexes in the aqueous solution and transfer to organic phase. time was only a few min. The average recovery of palladium(II) from an aqueous solution containing 10 microg/ml of analyte was 99% with an RSD% of 0.95. The percentage recovery of 0.2 microg/ml palladium(II) was 96%.

Experimental infection of BALB/c mice with the Gram-positive bacterium Nocardia asteroides (strain GUH-2) results in life-long movement abnormalities including head
shaking
and spinning when held by the tail. The head
shaking
is temporarily inhibited by treatment with dopamine's precursor levodopa, suggesting that abnormalities in dopaminergic neurotransmission may be involved in these movement abnormalities. Cell-free filtrates from N. asteroides cultures induce > 70% dopamine depletion in rat pheochromocytoma PC12 cells, suggesting that Nocardia's effects on dopamine neurons may result in part from secreted factors. The nature of this dopamine-depleting activity was examined in the present study. Dopamine-depleting activity in N. asteroides culture filtrate was resistant to heat (100 degrees C x 30 min), proteases, and
chloroform
extraction, and was present in a low molecular mass (< 3 kDa) fraction. It was partially inhibited by decreasing (to 4.0) or increasing (to 10.0) the filtrate pH. GUH-2 filtrate increased cellular lactate dehydrogenase release by only 2%, and induced apoptotic morphology in only 11% of PC12 cells, suggesting that dopamine-depleting activity was not due to either cell injury or induction of apoptosis. These results suggest that a protease-resistant, low molecular mass substance secreted by N. asteroides may be responsible for its dopamine-depleting effects.
