A new method to assess phosphorus bioavailability in the sediments and soils was developed by using a homemade iron oxide/ cellulose acetate membrane(FeO/CAM). The results show that the amount of bioavailable phosphorus(FeO-P) desorbed from the sediment and soil increased with the shaking time,and FeO-P desorption would come to equilibrium after 16 ot 20h. With the increase of the ratio between soil mass and water volume, the quantity of the desorbed FeO-P would decrease and then went planar after 20 to 30g/L. The quantity of the desorbed FeO-P had no significant difference between the FeO/CAMs of 20 to 30cm2. The dynamics of FeO-P desorption from the sediments and soils would be accelerated by appropriately increasing the shaking frequency. In short, FeO/ CAMs overcome many disadvantages of iron oxide impregnated filter paper and are easy to be commercialized; therefore it may have a better future in the application to assessing phosphorus hioavailability in the sediments and soils.

Clostridium thermocellum JW20 (ATCC 31549), which was isolated from a Louisiana cotton bale, grew on cellulose, cellobiose, and xylooligomers and, after adaptation, on glucose, fructose, and xylose in the pH range of 7.5 to 6.1 with T(opt) of 60 degrees C, T(max) of 69 degrees C, and T(min) of above 28 degrees C. Doubling times during growth on cellulose and cellobiose were 6.5 and 2.5 h, respectively. The G+C content of the DNA was 40 mol% (chemical analysis). Growth on cellulose as substrate was totally inhibited in the presence of more than 125 mM sodium sulfate, 300 mM sodium chloride, 250 mM potassium chloride, 200 mM calcium chloride, 125 mM magnesium chloride, 40 mM lactate, or 250 mM acetate. The ratio of the fermentation products ethanol to acetate plus H(2) decreased when the culture was agitated. Agitation otherwise increased the rate of cellulose degradation in a growing culture but not under nongrowth conditions or with cell-free culture supernatant containing the extracellular cellulase. Shaking lowered the concentration of H(2) in the culture broth and thus minimized inhibition by the H(2) formed. Externally added H(2) caused an increased formation of ethanol during growth on cellulose or cellobiose. However, at an atmospheric pressure as high as 355 kPa (50 lb/in), H(2) did not cause significant growth inhibition beyond an increasing lag phase (up to 24 h). Several criteria to specifically prove the purity of C. thermocellum cultures were suggested.

A protocol for direct extraction of mRNA from soil samples was developed. Soil samples (10 g) were washed twice with 120 mM phosphate buffer (pH 5.2). The lysis of cells, fixation of RNA, and hydrolysis of DNA were achieved by vigorously shaking the washed soil in a 4 M guanidine thiocyanate solution containing 25 mM sodium citrate, 0.5% sarcosyl, and 0.1 M 2-mercaptoethanol. The pH of the homogenized mixture was adjusted with 2 M sodium acetate (pH 4.0); the mRNA was then extracted with phenol and chloroform. Total RNA was precipitated with isopropanol. This method extracts up to 17 mug of total RNA per g (wet weight) of soil containing 8.0 x 10 cells of Pseudomonas aeruginosa PU21, and mRNA has been detected in 160-ng total RNA fractions. This method has been used for the detection of mRNA transcribed from specific biodegradative genes, including the nah and mer operons, in contaminated soils. This extraction method can be completed within a few hours and has tremendous potential for ecological studies of in situ gene expression among soil microbiotas.

Methanobacterium espanolae, an acidiphilic methanogen, required acetate for maximal growth on H(2)-CO(2). In the presence of 5 to 15 mM acetate, at a growth pH of 5.5, the mu(max) was 0.05 h. M. espanolae consumed 12.3 mM acetate during 96 h of incubation at 35 degrees C with shaking at 100 rpm. At initial acetate levels of 2.5 to 10.0 mM, the amount of biomass produced was dependent on the amount of acetate in the medium. C nuclear magnetic resonance spectra of protein hydrolysates obtained from cultures grown on [1-C]- or [2-C]acetate indicated that an incomplete tricarboxylic acid pathway, operating in the reductive direction, was functional in this methanogen. The amino acids were labeled with a very high degree of specificity and at greater than 90% enrichment levels. Less than 2% label randomization occurred between positions primarily labeled from either the carboxyl or methyl group of acetate, and very little label was transferred to positions primarily labeled from CO(2). The labeling pattern of carbohydrates was typical for glucogenesis from pyruvate. This methanogen, by virtue of the properties described above and its ability to incorporate all of the available acetate (10 mM or lower) from the growth medium, has advantages over other microorganisms for use in the production of specifically labeled compounds.

The myxomycetes are called slime molds because of the synthesis of copious amounts of extracellular material (slime) during parts of the life cycle. In Physarum polycephalum, small amounts of slime are produced during exponential growth of microplasmodia in shake flasks, but the amount of this slime increased 10- to 20-fold at 16 to 34 hr after microplasmodia were induced to form spherules by transferring them to salt solution. The slime obtained during both periods is the same; an acidic polysaccharide consisting of galactose, sulfate, and trace amounts of rhamnose. Analysis of the galactose-to-sulfate ratio gave a value of about 4 to 1. Infrared spectroscopy showed increased absorbance at 820 cm(-1) characteristic of C-O-S vibrations. Electrophoresis on polyacrylamide gel revealed that the material moved as a single band which stained with Alcian Blue and periodic acid Shiff reagent. However, fractionation of identical material on Dowex columns and electrophoresis on cellulose acetate showed the slime to be made up of three major fractions. The polysaccharide appeared as an extracellular capsule closely adhering to the walls of the spherules. It could be separated from the wall by vigorous shaking. The increased synthesis of slime during spherulation was not blocked by cycloheximide, suggesting that new enzyme synthesis was not necessary for its formation.

Oxygen supply is a key parameter in aerobic fermentation processes like the industrial production of amino acids. Although the oxygen transfer rate (OTR; or the volumetric oxygen transfer coefficient k(L)a) is routinely analyzed by engineers during stirred tank fermentations, it is often not taken into account by biologists conducting screening experiments in shake flasks. To show the importance of knowing how to avoid oxygen transfer limitations during primary screenings, Corynebacterium glutamicum ATCC 13032 (wild-type strain) and DSM 12866 (lysine-producing strain) were cultivated in shake flasks with different culture liquid volumes and under different shaking conditions. With the Respiration Activity Monitoring System, the OTR was determined quasi-continuously. Optical density as well as concentrations of lysine and byproducts (lactate, acetate, succinate) were determined off-line and correlated with the OTR signal. From the results, design criteria for improved screening in shaken bioreactors that help to avoid selection of suboptimal strains during early process development steps can be derived. Finally, the suitability of DSM 12866 as a strain for industrial processes with a high space-time yield is discussed.

Predictors of gastric emptying (GE) in patients with idiopathic Parkinson's disease (PD) of a solid and liquid meal are not well defined. For measurement of GE 80 patients with PD were randomly assigned to receive either a solid meal (250 kcal) containing 13C-octanoate (n = 40) or a liquid meal (315 kcal) with 13C-acetate (n = 40). All patient groups were off medication affecting motility and were matched for age, gender, body mass index, disease duration and severity, using Unified Parkinson's Disease Rating Scale (UPDRS). Gastric emptying was compared with a healthy control group (n = 40). Multiple regression analysis was used to determine predictors of gastric emptying. Exactly 88% and 38% of PD patients had delayed GE of solids and liquids respectively. Solid and liquid emptying was similar in women and men. There were no differences in GE in PD patients < 65 years of age when compared with patients > or = 65 years. Multiple regression analysis showed that motor handicaps such as rigour and action tremor are independent predictors of solid GE (r = 0.68, P < 0.001). The severity of motor impairment, but not any other neurological symptom, as assessed by UPDRS is associated with gastroparesis in PD and solid emptying is more likely to be delayed.

The population of filamentous acetate-utilizing methanogens in paddy field soils was 2.0 x 10(4) MPN/g dry soil in the submerged condition. They were able to form colonies in a deep agar medium, but not in a roll tube. Filamentous acetate-utilizing methanogens isolated from Kanagi, Japan (strain K-5) and Tsukuba, Japan (strain T-3) were divided into two types based on length of filaments. One type, strain K-5, formed a short chain which was dispersed easily by weak shaking. The other type, strain T-3, formed a long chain, which formed cotton-like flocs and was not dispersed by weak shaking. They had sheaths composed of a pair of adjacent membranes on the outside of the cell membranes. The 16S rRNA gene similarities of strain T-3 and K-5 to Methanosaeta concilii strain Opfikon were 100% and 99.5% respectively. Filamentous acetate-utilizing methanogens were also isolated from paddy field soils in various other regions of Japan. Our results suggest that Methanosaeta is universal in paddy soils and that it plays an important role in methane production from acetate.

Mutations in the gene for aspartoacylase (ASPA), which catalyzes deacetylation of N-acetyl-L-aspartate in the central nervous system (CNS), result in Canavan Disease, a fatal dysmyelinating disease. Consistent with its role in supplying acetate for myelin lipid synthesis, ASPA is thought to be cytoplasmic. Here we describe the occurrence of ASPA within nuclei of rat brain and kidney, and in cultured rodent oligodendrocytes. Immunohistochemistry showed cytoplasmic and nuclear ASPA staining, the specificity of which was demonstrated by its absence from tissues of the Tremor rat, an ASPA-null mutant. Subcellular fractionation analysis revealed low enzyme activity against NAA in nuclear fractions from normal rats. Whereas two recent reports have indicated that ASPA exists as a dimer, size-exclusion chromatography of subcellular fractions showed ASPA is an active monomer in both subcellular fractions. Western blotting detected ASPA as a single 38 kD band. Because ASPA is small enough to passively diffuse into the nucleus, we constructed, expressed, and detected in COS-7 cells a green fluorescent protein-human ASPA (GFP-hASPA) fusion protein larger than the permissible size for the nuclear pore complex. GFP-hASPA was enzymatically active and showed mixed nuclear-cytoplasmic distribution. We conclude that ASPA is a regulated nuclear-cytoplasmic protein that may have distinct functional roles in the two cellular compartments.

The effects of demineralized water (DEMI H(2)O) and 0.5 M ammonium acetate (0.5 M AAc) on losses of L-glutamic acid and L-arginine in the course of shaking and filtration at low temperature (6 degrees C) were tested. The concentration of L-glutamic acid decreased by 6.3% in DEMI H(2)O and by 4.9% in 0.5 M AAc, whereas the L-arginine concentration decreased by 6.0% (DEMI H(2)O) and 10.7% (0.5 M AAc). We found a significantly (P < 0.05) higher degradation of L-arginine in 0.5 M AAc compared with that of DEMI H(2)O.
