Temporal changes of plasma erythropoietin (Epo) in mice exposed to hypobaric hypoxia were studied by a fetal mouse liver cell culture method. Since a colony formation inhibitory activity was found in the mouse plasma, thirteen pretreatment procedures for bioassay were compared and the procedure of shaking with chloroform followed by dialysis was concluded to be the best. When normal mice (P50 = 40.4 +/- 2.2 Torr) were exposed to hypoxia of 350 Torr, the plasma Epo level was elevated, with peak at the 2nd to 3rd day, and afterwards declined gradually. On the contrary, cyanated mice (P50 = 30.1 +/- 1.5 Torr) showed much less of the Epo response when exposed to 350 Torr. Under 200 Torr hypoxia, both mice exhibited a similar and remarkable extent of the response. These results suggest that the renal Epo-producing tissue or its oxygen-sensing system is less hypoxic in cyanated mice than in normal mice under 350 Torr, and that the physiologically optimal oxygen affinity of blood is variable depending on hypoxic degrees. The fact that the inhibitory activity showed an inverse temporal change to that of Epo, suggested a possible important role of this activity in the regulation of erythropoiesis under hypoxia.

The molecular stability and function of hemoglobin (Hb) Hasharon (alpha 2 H beta 2) and Hb Hasharon2 (alpha 2 H delta 2) were studied and compared to Hbs A, A2 and S. Hb Hasharon and Hb Hasharon2 had slightly lower P50 values than Hb A and Hb A2 but had normal responses to organic phosphates. The molecular stability of Hb Hasharon and Hb Hasharon2 (as measured by mechanical shaking and heat denaturation at 60 degrees C) were less than Hb A and Hb A2 but greater than Hb S in the oxy- and carbonmonoxy-forms. In the met-form, however, Hb Hasharon and Hb Hasharon2 were less stable than hemoglobins S, A and A2. The oxy-form of Hb Hasharon forms methemoglobin at a faster rate than Hb A and Hb S. The mechanical and heat stabilities and the rate of methemoglobin formation of oxy-Hb Hasharon were studied in the presence of sulfisoxazole. This drug increased the rate of methemoglobin formation, thus causing a further decrease in the stability of Hb Hasharon. The relationship between these laboratory findings and previously observed clinical findings associated with Hb Hasharon are discussed.

A second case of Hb York (beta 146 His leads to Pro), was discovered in a patient with polycythemia. The oxygen equilibrium curves (OEC) of red cell suspensions in a buffer (pH 7.4) at 37 degrees C revealed a biphasic curve with a P50 of only 12.5 mm Hg (normal value: 26.5 +/- 1.0 mm Hg). The purified Hb York had an extremely high affinity for oxygen with diminished cooperativity and decreased Bohr effect. The oxygen affinity was significantly reduced by inositol hexaphosphate. Molecular stability studies by mechanical shaking of various liganded forms of Hb York revealed stabilities between those of Hb A and Hb S. Isolated beta Y-subunits were more unstable than beta A-subunits at every pH examined. Hb York was 1.4 times more unstable than Hb A in 18.9% isopropanol.

Amyotrophic lateral sclerosis (ALS) is characterized by progressive degeneration of motoneurons. One potential mechanism is excitotoxicity. We studied the behaviors of spinal neurons using an in vitro preparation of the sacral cord from the G93A SOD1 mouse model of ALS. Measurements were conducted at presymptomatic [approximately postnatal day 50 (approximately P50)], early (approximately P90), and late (>P120) stages of the disease. Short-latency reflexes (SRs) in ventral roots, presumably monosynaptic, were evoked by electrical stimulation of a dorsal root. The fraction of motoneurons capable of responding to this activation was evaluated by measuring the compound action potential [total motor activity (TMA)] evoked by antidromic stimulation of the distal ventral root. In mutant SOD1 (mSOD1) mice, both the SR and the TMA decreased with age compared with nontransgenic littermates, ruling out the SR as a source of increasing excitotoxicity. Spinal interneuron activity was assessed using the synchronized ventral root bursts generated by both bath application of blockers of inhibitory neurotransmitters (glycine, GABA(A)) and agonists of glutamate receptors (especially NMDA receptors). After symptom onset, a higher percentage of preparations from mSOD1 mice exhibited bursting, and these bursts exhibited more sub-bursts and a more disorganized pattern. In mSOD1 mice with clear muscle tremor, the ventral roots exhibited spontaneous synchronized bursts, which were highly sensitive to the blockade of NMDA receptors. These data suggest that although short-latency sensory input does not increase as symptoms develop, interneuron activity does increase and may contribute to excitotoxicity.

Suspension of the pristine and COOH-substituted multi-walled carbon nanotubes (P- and C-MWCNTs) with different outer diameters (ODs) by humic acids (HAs) from a peat soil was examined. Under shaking condition, MWCNTs were not suspended within 5 d. Without HAs, C-MWCNTs were slightly suspended by sonication within 16 h, but no suspension was observed for the pristine ones (P-MWCNTs). HAs greatly enhanced suspension of both P- and C-MWCNTs. The suspension enhancement was attributed to HA sorption, which increased electrostatic repulsion and steric hindrance between individual MWCNTs. Introduction of O-containing hydrophilic moieties to MWCNTs via HA sorption enhanced the interactions of their surfaces with water through H-bonding. Suspending capability of various MWCNTs on suspended mass concentration basis by four HAs showed inconsistent orders with the increasing or decreasing trend of their ODs. However, the suspended surface area concentrations of both P- and C-MWCNTs by individual HAs consistently followed an order of P8 > P30 > P50, and C8 > C30 > C50 (P and C, respectively, refer to P- and C-MWCNTs, and the numbers represent their ODs). These data implied that MWCNTs with smaller OD could be more strongly suspended by a given HA relative to those with larger OD under sonication condition.
