Parkinson's disease, a major neurodegenerative disorder in humans whose etiology is unknown, may be associated with some environmental factors. Nocardia otitidiscaviarum (GAM-5) isolated from a patient with an actinomycetoma produced signs similar to Parkinson's disease following iv injection into NMRI mice. NMRI mice were infected intravenously with a non-lethal dose of 5 x 10(6) colony forming units of N. otitidiscaviarum (GAM-5). Fourteen days after

, most of the 60 mice injected exhibited parkinsonian features characterized by vertical head
tremor
, akinesia/bradykinesia, flexed posture and postural instability. There was a peak of nocardial growth in the brain during the first 24 h followed by a decrease, so that by 14 days nocardiae could no longer be cultured. At 24 h after infection, Gram staining showed nocardiae in neurons in the substantia nigra and occasionally in the brain parenchyma in the frontal and parietal cortex. At 21 days post-infection, tyrosine hydroxylase immunolabeling showed a 58% reduction of tyrosine hydroxylase in the substantia nigra, and a 35% reduction of tyrosine hydroxylase in the ventral tegmental region. Dopamine levels were reduced from 110 +/- 32.5 to 58 +/- 16.5 ng/mg protein (47.2% reduction) in brain from infected mice exhibiting impaired movements, whereas serotonin levels were unchanged (191 +/- 44 protein in control and 175 +/- 39 ng/mg protein in injected mice). At later times, intraneuronal inclusion bodies were observed in the substantia nigra. Our observations emphasize the need for further studies of the potential association between Parkinson's disease or parkinsonism-like disease and exposure to various nocardial species.

Strong covalent immobilization of amikacin on Uni-Graft((R)) DV straight vascular prostheses made of gelatine-sealed poly(ethylene terephthalate) fibres was performed according to procedure described in the Polish Patent No. P-358934. The concentrations of amikacin in sample solutions were estimated either by HPLC or by UV spectroscopy method previously optimized for amikacin measurements. A high correlation was found between these two methods. It was found that the antibiotic was bound in mixed-type way via three types of interactions: strong covalent bonds (dominating amount: 81.84%) and weak interactions: physical adsorption and ionic bonds (18.19%). Even when total amount of physically and ionically attached drug has been released, the remaining covalently bound amount still locally protected the prostheses in vitro against bacteria. The release test was conducted in PBS at pH 7.4 at 37 degrees C and showed that about 15% of total drug amount was eluted from the matrix during the first 7 days of
shaking
, then no more antibiotic was released. It suggested that about 85% of amikacin attached to prosthesis modified in mixed-type mode was bound via covalent interactions. A bacterial inhibition test on Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923 and Pseudomonas aeruginosa ATCC 27853 showed inhibition of growth for all strains at low inoculum concentrations up to 30 days as well as high inoculum concentration for E. coli. At high concentrations of S. aureus and P. aeruginosa, the modified prostheses showed slight bacteriostatic effect since 10th day of experiment. Amikacin-modified vascular prostheses might therefore be protected against
bacterial infection
locally, without long-lasting drug release to human system.

Shaking
is a nonantibiotic management technique for the
bacterial disease
American foulbrood (AFB) (Paenibacillus larvae sensu Genersch et al.), in which infected nesting comb is destroyed and the adult honey bees, Apis mellifera L. (Hymenoptera: Apidae), are transferred onto uncontaminated nesting material. We hypothesized that colonies shaken onto frames of uninfected drawn comb would have similar reductions in AFB symptoms and bacterial spore loads than those shaken onto frames of foundation, but they would attain higher levels of production. We observed that colonies shaken onto drawn comb, or a combination of foundation and drawn comb, exhibited light transitory AFB infections, whereas colonies shaken onto frames containing only foundation failed to exhibit clinical symptoms. Furthermore, concentrations of P. larvae spores in honey and adult worker bees sampled from colonies shaken onto all comb and foundation treatments declined over time and were undetectable in adult bee samples 3 mo after
shaking
. In contrast, colonies that were reestablished on the original infected comb remained heavily infected resulting in consistently high levels of spores, and eventually, their death. In a subsequent experiment, production of colonies shaken onto foundation was compared with that of colonies established from package (bulk) bees or that of overwintered colonies. Economic analysis proved
shaking
to be 24% more profitable than using package bees. These results suggest that
shaking
bees onto frames of foundation in the spring is a feasible option for managing AFB in commercial beekeeping operations where antibiotic use is undesirable or prohibited.

Bacterial attachment and growth on material surfaces are considered to be the primary steps leading to the formation of biofilm. Biofilms in hospital and food processing settings can result in
bacterial infection
and food contamination, respectively. Prevention of bacterial attachment, therefore, is considered to be the best strategy for abating these menaces and therefore the development of antibacterial metals becomes important. In this study, nine pure metals, viz. titanium, cobalt, nickel, copper, zinc, zirconium, molybdenum, tin, and lead have been tested for their antibacterial properties against two bacterial strains, Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli. This was accomplished using two assay methods, the film contact method and the
shaking
flask method. The results show that the antibacterial properties varied significantly with different metals and the effectiveness of metals to resist bacterial attachment varied with the bacterial strain. Among the metals tested, titanium and tin did not exhibit antibacterial properties. TEM images showed that metal accumulation resulted in the disruption of the bacterial cell wall and other cellular components.

Although few antibacterial treatments have been proposed for pacemakers, the rate of cardiac pacemaker-related inflammation still exhibits an abnormal increase. By analyzing the statistical reports of the inflammation at different time stages caused by pacemakers, herein, we proposed a new hypothesis to explain this unusual phenomenon.
