The purpose of this study was to elucidate the role of extracellular matrix components such as aggrecan,
fibronectin
, and laminin in the extraocular muscle of patients with
strabismus
. Resected tissues of the medial rectus muscle of 47 patients with intermittent exotropia obtained during recession-resection surgery were frozen under liquid nitrogen and pulverized by a Freezer/Mill to solubilize the tissue for enzyme immunoassay. The total amounts of aggrecan,
fibronectin
, and laminin in the resected tissue were correlated with clinical data of patients such as age, exodeviation, and refractive error. The amount of aggrecan decreased significantly with the advance of age (P < 0.0001, Spearman rank correlation test), while the amount of laminin or
fibronectin
had no correlation with age. Patients with basic type intermittent exotropia showed larger, although not significantly, amounts of aggrecan than those with convergence insufficiency type (P = 0.0538, Mann-Whitney U-test). The amount of aggrecan may be related to motor aspects of intermittent exotropia.

The noncanonical wnt/planar cell polarity (PCP) pathway [1] regulates the mediolaterally (planarly) polarized cell protrusive activity and intercalation that drives the convergent extension movements of vertebrate gastrulation [2], yet the underlying mechanism is unknown. (Xstbm), Frizzled (Xfz7), and Prickle (Xpk), disrupts radially polarized
fibronectin
fibril assembly on mesodermal tissue surfaces, mediolaterally polarized motility, and intercalation. Polarized motility is restored in Xpk-perturbed explants but not in Xstbm- or Xfz7-perturbed explants cultured on
fibronectin
surfaces. The PCP complex, including Xpk, first regulates polarized surface assembly of the
fibronectin
matrix, which is necessary for mediolaterally polarized motility, and then, without Xpk, has an additional and necessary function in polarizing motility. These results show that the PCP complex regulates several cell polarities (radial, planar) and several processes (matrix deposition, motility), by indirect and direct mechanisms, and acts in several modes, either with all or a subset of its components, during vertebrate morphogenesis.

This paper aims to identify key biological processes triggered by resection surgery in the extraocular muscles (EOMs) of a rabbit model of
strabismus
surgery by studying changes in gene expression. Resection surgery was performed in the superior rectus of 16 rabbits and a group of non-operated rabbits served as control. Muscle samples were collected from groups of four animals 1, 2, 4 and 6 weeks after surgery and processed for RNA-sequencing and immunohistochemistry. We identified a total of 164; 136; 64 and 12 differentially expressed genes 1, 2, 4 and 6 weeks after surgery. Gene Ontology enrichment analysis revealed that differentially expressed genes were involved in biological pathways related to metabolism, response to stimulus mainly related with regulation of immune response, cell cycle and extracellular matrix. A complementary pathway analysis and network analysis performed with Ingenuity Pathway Analysis tool corroborated and completed these findings. Collagen I,

and versican, evaluated by immunofluorescence, showed that changes at the gene expression level resulted in variation at the protein level. Tenascin-C staining in resected muscles demonstrated the formation of new tendon and myotendinous junctions.
