A comparative study of various factors of humoral immunity in human chronic herpes stomatitis (CHS) in periods of relapses and remission of the infection showed the exacerbations of the disease to occur in the presence of complement-fixing (CF) and virus-neutralizing (VN) antibodies in the blood serum the titres of which moderately rose with the progress of the infection. A relapse of the disease occurs during marked decline in the content of secretory IgA and true secretory Sc-IgA, serum and secretory IgG, low levels of serum interferon, and a small rise in titres of 19S specific CF antibodies. Remission comes as these values become normal. Although specific immunological changes do occur in CHS, they are not very marked, whereas the factors of nonspecific host resistance change considerably in close correlation with the periods of relapse and remission of the infection.

Job' syndrome and IgA immunodeficiency are a rare dysfunction of the immune system. In this work, we reported a case of a young woman who had recurrent episodes of bacterial infections in the urinary tract and genital, generalized erythematous eczematous patches and stomatitis of oral mucosa and fever. During the hospitalization, laboratory data showed high immunoglobulin IgE and low IgA levels. The T-lymphocyte presented a reduction of CD8+ cells. Tests of granulocyte function have showed a global deficit in the in vitro and in vivo chemotaxis. The correlation between these two clinic conditions is not completely clarified but it is possible to hypothesize that CD8+ lymphocytes produce an inhibition factor of chemotaxis. Job' syndrome is characterized by a selective reduction of CD8+ cells subpopulation which have an immunoregulatory function on the production of IgE by plasmacells. In the ipoIgA, an intrinsic inability of B-IgA cells to proliferate and to differentiate produce a defect in the IgA production. In these two clinic disorders there is an effective dysfunction of immune system. It is possible to hypothesize that an effective defect of CD8+ cells and an immaturity of B-cells may coexist in our patient. That justifies an abnormal production of Ig and a defect in granulocyte chemotaxis.

In phylogenetically diverse species with the help of T lymphocytes or soluble factors, viral infections induce the Ag-specific B lymphocytes to proliferate and terminally differentiate into IgM, IgG, IgA, IgD, or IgE Ab-secreting cells. Based on previous studies searching for IgD, it was inferred that serum IgD in the mouse is nearly undetectable, although in other species, e.g., humans, IgD is a measurable component of serum Ig. More recently, new information has been obtained indicating that IgD is secreted in minute quantities during normal B cell differentiation. We observed that IgD is secreted in significantly increased quantities in mice undergoing an acute infection with lymphocytic choriomeningitis virus or vesicular stomatitis virus compared with uninfected animals. A substantial fraction of the observed IgD was found to be virus specific. Using a solid-phase immunoenzymatic technique, virus-specific IgD Ab-forming cells were detected in the spleen; their numerical increase correlated with the level of secreted antiviral IgD. In addition, immunohistochemical staining revealed IgD+ plasma cells that occurred with a similar kinetic profile as the virus-specific IgD Ab-forming cells. These findings provide direct evidence that synthesis of IgD is a physiologic event in the mouse. Its precise function in the immune response to pathogens, however, remains to be determined.

Desquamative gingivitis is a fairly common complaint. Typically seen in females who are middle-aged or older, it is predominantly a manifestation of a range of vesiculobullous disorders. The main complaint is of persistent soreness of the gingiva. Most cases are related to lichen planus or pemphigoid, but it is also important to exclude pemphigus, dermatitis herpetiformis, linear IgA disease, chronic ulcerative stomatitis, and other conditions. Biopsy is invariably required to confirm the diagnosis after a full history, general, and oral examination. Apart from improving the oral hygiene, immunosuppressive therapy is typically required to control the condition.

However, less common conditions such as pemphigus, dermatitis herpetiformis, linear IgA disease, erythema multiforme, chronic ulcerative stomatitis and other conditions may need to be excluded. Diagnosis is initially clinical, based on a full history, general, and oral examination but precise diagnosis can be difficult without further data and therefore biopsy and immunological studies are invariably required to confirm the specific diagnosis. Apart from improving oral hygiene, immunosuppressive therapy is typically required to control many of these conditions.

Attachment of Candida albicans to plastic materials of dental prostheses or to salivary macromolecules adsorbed on their surface is believed to be a critical event in the development of denture stomatitis. In an earlier study, it was shown that adhesion of C. albicans to polystyrene, a model system to study the adhesion of C. albicans to plastic materials, can be partially inhibited with an mAb directed against cell wall polysaccharides of C. albicans. In the present study, the role of whole saliva in the adhesion of C. albicans to polystyrene has been investigated, and three mAbs directed against epitopes of cell wall mannoproteins have been used to mimic the inhibitory effect observed with salivary secretory IgA (sIgA) on the adhesion of C. albicans to polystyrene. In the absence of whole saliva, adherence of C. albicans 3153 increased with germination. However, the presence of whole saliva enhanced the adhesion to polystyrene of C. albicans 3153 yeast cells but decreased the adhesion of germinated cells. The enhancement of adhesion of yeast cells to polystyrene mediated by saliva was confirmed with an agerminative mutant of C. albicans 3153. The inhibition of the adhesion of C. albicans 3153 germ tubes to polystyrene was due to the salivary sIgA since sIgA-depleted saliva enhanced the adhesion of C. albicans 3153 to polystyrene. The inhibitory effect mediated by sIgA was not related to the inhibition of germination but to the blockage of adhesins expressed on the cell wall surface of the germ tubes. The three mAbs studied reduced the adhesion of C. albicans 3153 to polystyrene at levels equivalent to those for purified sIgA. The highest reduction in the adhesion was obtained with the IgA mAb N3B. The best results were obtained when the three mAbs were combined. The results suggest that whole saliva plays a different role in the adhesion of C. albicans to polystyrene depending on the morphological phase of C. albicans. These results may give new insights into the conflicting role of saliva in the adhesion of C. albicans to plastic materials of dental prostheses.

The efficiency of licopide in combination with traditional treatment was evaluated in 59 children aged 1-14 years suffering from allergic diseases and relapsing herpetic stomatitis. Immunological studies were carried out before and after a course of therapy: local oral immunity, cellular immunity (in the blast transformation test with phytohemagglutinin), lymphocyte populations and subpopulations, humoral immunity values (IgA, IgM, IgG, and IgE), and C3 complement component were evaluated. The results indicate that licopide in complex with traditional methods normalized the immunological values, had a favorable impact on the course of relapsing herpetic stomatitis, and led to a stable remission of allergic diseases (p<0.001).

Recombinant rhabdovirus vectors expressing human immunodeficiency virus (HIV) and/or simian immunodeficiency virus (SIV) proteins have been shown to induce strong immune responses in mice and rhesus macaques. However, the finding that such responses protect rhesus macaques from AIDS-like disease but not from infection indicates that further improvements for these vectors are needed. Here, we designed a prime-boost schedule consisting of a rabies virus (RV) vaccine strain and a recombinant vesicular stomatitis virus (VSV) both expressing HIV Envelope (Env). Mice were primed and boosted with the two vaccine vehicles by different routes and in different combinations. Mucosal and systemic humoral responses were assessed using enzyme linked immunosorbent assay (ELISA) while the cellular immune response was determined by an IFN-gamma ELISPOT assay. We found that an immunization combination of RV and VSV elicited the highest titers of anti-Env antibodies and the greatest amount of Env-specific IFN-gamma secreting cells pre- and post-challenge with a recombinant vaccinia virus expressing HIV(89.6) Env. Furthermore, intramuscular immunization did not induce antigen-specific mucosal antibodies while intranasal inoculation stimulated vector-specific IgA antibodies in vaginal washings and serum. Our results show that it is feasible to elicit robust cellular and humoral anti-HIV responses using two different live attenuated Rhabdovirus vectors to sequentially prime and boost.

Denture stomatitis is an inflammatory condition affecting palatal mucosa covered by a denture. Aetiology is multifactorial involving poor hygiene, constant denture wearing, trauma and colonisation by Candida species. In inflamed tissues raised levels of albumin, IgG isotypes and secretory IgA (sIgA) would be expected. SIgA is a major component of the protective immunological response to mucosal injection. Palatal secretions of 21 patients with denture stomatitis and 22 denture wearing patients with clinically healthy palates were analysed for albumin, IgG isotypes and sIgA levels. Albumin and IgG isotype levels were found to be higher in denture stomatitis patients compared with healthy palate patients. Unexpectedly, SIgA levels were found to be significantly (p<0.05) lower in denture stomatitis patients compared with healthy palate patients.
