Mannan
sulfate, a novel sulfated polysaccharide, was prepared and investigated for its activity against human immunodeficiency virus type 1 (HIV-1) in vitro.
Mannan
sulfate completely inhibited HIV-1-induced cell destruction and viral antigen expression in HIV-1-infected Molt-4 (clone 8) cells at a concentration of 4 micrograms/ml. The 50% antiviral effective doses obtained with mannan sulfate in Molt-4 (clone 8) cells and in MT-4 cells were 1.5 and 9.3 micrograms/ml, respectively. No toxicity for Molt-4 (clone 8) cells or MT-4 cells was observed at a concentration of 4,000 and 2,500 micrograms/ml, respectively.
Mannan
sulfate was also inhibitory to other enveloped viruses, i.e. herpes simplex virus types 1 and 2, vaccinia virus and vesicular
stomatitis
virus. These results suggest that mannan sulfate may be useful for the chemotherapy of various viral infections, including those causing and associated with AIDS.

Dendritic cells (DC) are the most potent antigen-presenting cells that regulate immune responses. One of the mechanisms for hepatitis C virus (HCV) persistence is the ability of HCV to suppress DC function. Direct HCV infection to blood DC has been implicated for DC dysfunction. To clarify the susceptibility of each DC subset to HCV, we used pseudotype vesicular
stomatitis
virus (VSV) coated with chimeric HCV envelope glycoproteins (E1 and E2). We demonstrate that pseudotype VSV enters myeloid DC (MDC) but not plasmacytoid DC (PDC). The highest efficiency of pseudotype VSV entry to MDC was observed when MDC were cultured with GM-CSF. Such efficiency decreased when MDC are matured with the treatment of IL-4, CpG oligodeoxynucleotide, or CD40 ligand.
Mannan
inhibited pseudotype VSV entry to MDC, but Ca(2+) chelators failed to do so. These results show that pseudotype VSV possessing HCV-E1 and E2 enters immature MDC through the interaction with lectins in a Ca(2+)-independent manner.

The adherence of Candida on dentures is related to diseases such as denture
stomatitis
and aspiration pneumonia.
Mannan
is a major component of the Candida cell surface, and contributes to the cell adherence. A previous report indicated that the adherence of C. albicans to culture dishes was inhibited by the coating them with mannan. The purpose of this study was to examine the adhesion inhibitory effect of mannan coating on acrylic denture surfaces against C. albicans and C. glabrata. The amount of Candida attached on the acrylic surfaces coated with mannan was calibrated by culture methods.
Mannan
showed significant inhibitory effects on Candida adhesion in both the yeast and hyphal form in a concentration-dependent manner, and the durability of the inhibitory effect continued for three days. These results suggest that mannan coating on the denture base acrylic can prevent Candida adhesion on the denture.
