A cell line established from human embryonic lung, HEL-R66, was demonstrated to be highly susceptible to herpes simplex virus types 1 and 2, vaccinia virus, Newcastle disease virus (NDV), Japanese encephalitis virus (JEV), western equine encephalitis (WEE) virus, Sindbis virus, vesicular
stomatitis
virus (VSV), and
rabies
virus. The maximal yields of NDV, JEV, WEE virus, and
rabies
virus in this cell line exceeded by 2--4 logs those in control human embryonic lung cells. Inability of this cell line to produce interferon upon treatment with native and UV-irradiated forms of virogenic and lentogenic strains of NDV and with poly I:C was revealed. A refractory state to challenging VSV did not develop in HEL-R66 cells treated with the inducers. Furthermore, pretreatment of HEL-R66 cells with interferon did not potentiate the capacity to produce interferon in response to the addition of poly I:C, whereas the same treatment enhanced the production of interferon in normal human embryonic lung cells.

Different patterns of
rabies
virus infection were observed in BHK-21/13S and HEp-2 cell cultures at late stages of persistent infections. The infection of BHK-21/13S cells was characterized by periodical increases and declines in the portion of the antigen-containing cells (from 100% to less than 1%) and low infectivity titers which did not correspond to fluctuations in the antigen production and occasional resistance to challenge with vesicular
stomatitis
virus. In contrast, persistently infected HEp-2 cell culture exhibited a more constant course of antigen production which never reached extreme points; the infectivity titers generally correlated with the portion of the antigen-containing cells. Cultivation in the presence of antirabies serum did not "cure" either of the cultures.

Three cell cultures chronically infected with fixed
rabies
virus, strain MNIIVP-74, have been obtained: HEp-2/2, BHK/13S, and RK-13. In the former two cultures, the infectious virus titers were 2.0 to 5.25 Ig LD50/ml. In RK-13 cells, traces of the infectious virus were found. In the chronically infected HEp-2/2 culture the maximum amount of the antigen-containing cells determined by the fluorescent antibody procedure was 60% and in BHK/13S 80%. Chronically infected cultures had a reduced growth rate and were as sensitive as the controls of superinfection with vesicular
stomatitis
virus. The virus recovered from chronically infected culture produced a disease in mice at later intervals than the original virus used in the same doses.

This study of the physico-chemical properties of bovine ephemeral fever virus was initiated to establish whether or not it should be classified as a rhabdovirus. In contrast to the regular bullet-shaped morphology of some rhabdoviruses the virus particles are often cone-shaped or slight variants from bullet-shaped. The virion contains single-stranded RNA sedimenting at 42S and six proteins with mol. wt. of 164, 101, 64, 53, 43 and 29 x 10(3). The protein P101 is located on the surface of the virus and is glycosylated. It is removed by treatment of the virus particles with trypsin. Protein P64, the nucleoprotein, was found to be a phosphoprotein, like the N protein of
rabies
virus, whereas in vesicular
stomatitis
virus NS is the phosphorylated protein. Virus harvests contain defective-interfering particles. The particles are short cone-shaped forms about one-third the length of the infectious virion and similar in morphology to defective-interfering particles of vesicular
stomatitis
virus. These particles interfere with the replication of bovine ephemeral fever virus but not with the Indiana serotype of vesicular
stomatitis
virus. They contain single-stranded RNA sedimenting at 18 to 20S. The particles appear to have a protein composition identical to that found in the virus particle. The physico-chemical properties of bovine ephemeral fever virus justify its inclusion in the family Rhabdoviridae. The protein composition differs in detail from that found for vesicular
stomatitis
and
rabies
viruses, but is similar to that found for Obodhiang and kotonkan, two
rabies
serogroup viruses isolated from insects in Africa.

RNA-dependent RNA polymerases were found to be associated with two salmonid rhabdoviruses: infectious hematopoietic necrosis (IHN) virus and the virus of hemorrhagic septicemia (VHS). The protein composition of these rhabdoviruses closely resembles that of
rabies
virus rather than that of vesicular
stomatitis
virus (McAllister and Wagner, 1975). The optimal temperature for in vitro transcription was found to be approximately 18 degrees C for IHN virus and approximately 15 degrees for VHS,, closely approximating optimal temperatures for growth of these viruses in salmonid cells. Unlike vesicular
stomatitis
virus, manganese ion (1 mM) could be used as a divalent cation substitute for magnesium ion (5 mM). The in vitro transcription products of IHN and VHS viruses hybridized completely to the homologous genome but not at all to the heterologous genome.

Purified infectious hematopoietic necrosis (IHN) virus and the virus of haemorrhagic septicaemia (VHS) (Egtved virus) each contain five structural proteins which were designated L, G, N, M-1, and M-2. The IHN viral polypeptides have molecular weights estimated to be 157,000, 72,000, 40,000, 25,000 and 20,000, respectively, whereas those of VHS viral polypeptides are estimated to be 157,000 74,000, 41,000, 21,500, and 19,000, respectively. The carbohydrate composition of the glycoprotein (G) was confirmed by demonstrating selective incorporation of [3H]glucosamine into the designated G protein of both viruses. Phosphoproteins were identified by incorporation of [32P]orthophosphate into the N and M-1 proteins of IHN virus and into the N protein of VHS virus. The glycoprotein of each virus was selectively solubilized by treatment with Triton X-100 in low salt buffer, whereas the M-1, and M-2 proteins along with the G protein were solubilized by Ttition X-100 in 0.43 M NaCl. The protein composition of the salmonid rhabdoviruses resembles that of the
rabies
virus group more closely than the vesicular
stomatitis
virus group.

Viruses isolated from fish with viral haemorrhagic septicaemia (VHS), infectious haematopoietic necrosis (IHN), spring viraemia of carp (SVC), swim-bladder inflammation (SBI) and pike fry disease (PFD) have been grown to high titre in fathead minnow cells. While our preparations of the IHN, SVC, SBI and PFD viruses showed typical rhabdovirus morphology with bullet-shaped particles and distinct surface projections, the VHS virus preparations had a less typical rhabdovirus morphology but were pleomorphic with a preponderance of flexuous rods. Using virus labelled with [-3H]-uridine, it was shown that each virus contained RNA which sedimented at 38 to 40 S and was hydrolysed by very low concentrations of ribonuclease. The viruses of SVC, PFD and SBI had a polypeptide composition similar to that of vesicular
stomatitis
virus, the prototype rhabdovirus, but the IHN and VHS viruses gave a pattern similar to that of
rabies
virus. In serum neutralization tests the SVC and SBI viruses were indistinguishable. VHS virus showed no serological relationship with the other four viruses but there was a low level of cross-reaction between the PFD, IHN and SVC-SBI viruses.

Proteins from four fish rhabdoviruses have been studied by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The viruses were: trout viral hemorrhagic septicemia (VHS), infectious hematopoietic necrosis virus (IHN), spring viremia virus of carp (SVC), and the pike fry rhabdovirus (PFR). For the two salmonid viruses (VHS-IHN), gel electrophoresis indicated the proteins, with molecular weights estimated to be 190,000, 80,000, 38,000, 25,000, and 19,000, respectively. The electrophoretic profile of the two other viruses (SVC-PFR) revealed four major proteins with molecular weights of 190,000 80,000 42,000 and 21,000, respectively. In this case a minor component with 50,000 daltons was found. For each virus only one protein was found to be glycosylated, i.e., the one with a molecular weight of 80,000. A major protein (molecular weight between 38,000 and 42,000) was found to be associated with the nucleocapsid. All these results revealed marked similarities in protein structure between the four fish rhabdoviruses and the previously well-characterized members of rhabdovirus group. However, one can distinguish two groups of viruses: the first one is composed of salmonid viruses (VHS and IHN) with a protein structure comparable to that of
rabies
virus and potato yellow dwarf virus; the second one is composed of carp and pike viruses, having a protein structure very similar to that of vesicular
stomatitis
virus.

We investigated a 73-kDa polypeptide (p73), a minor component of the
rabies
virion (HEP-Flury and ERA strains), accounting for as much as 1% of total virion proteins. Two-dimensional gel electrophoresis and immunoblotting with the antiserum against the heat shock protein 70 (hsp70) demonstrated that p73 was identical to a constitutive type of cellular hsp70. The antiserum also detected p73/hsp70 in the purified virions of other negative-stranded RNA viruses, such as vesicular
stomatitis
virus (New Jersey serotype), Newcastle disease virus (Miyadera strain), and influenza A virus (PR8 strain), among which, however, the contents were variable.

The nucleotide sequence of the L gene of Marburg virus, strain Musoke, has been determined. The L gene has a single long open reading frame encoding a polypeptide of 2330 amino acids (MW 267,175) that represents the viral RNA-dependent RNA polymerase. The putative transcription start signal (3'CUACCUAUAAUU 5') and the termination signal (3' UAAUUCUUUUU 5') of the gene could be identified. Computer-assisted comparison of the L protein with L proteins of other nonsegmented negative-stranded RNA viruses (Paramyxoviridae: Sendai virus, Newcastle disease virus, human parainfluenza 3 virus, measles virus, human respiratory syncytial virus; Rhabdoviridae: vesicular

virus) revealed significant homologies primarily in the N-terminal half of the proteins. We have identified three common conserved boxes (A, B, and C) among filo-, paramyxo-, and rhabdovirus L proteins, which are probably involved in the polymerase function. The L proteins can be divided into an N-terminal half, which seems to accommodate the common enzymatic sites, and a C-terminal half carrying virus specific peculiarities. The data presented here suggest a common evolutionary history for all nonsegmented negative-stranded RNA viruses and show that filoviruses are more closely related to paramyxo- than to rhabdoviruses.
