Anomalous
DNA
synthesis was seen in the stomach mucosa of mice with experimental
stomach ulcer
during different phases of the ulcerous process, using histoautoradiography. At the early stage of ulcer formation a decrease in the label index (LI) is seen. Formation of the ulcer, morphologically similar to the shronic one, is accompanied by growth of the number of
DNA
-synthesizing cells in its margins. Over a period of ulcer healing proliferative activity of epithelium decreases approximately to an initial level. Histoautoradiographic studies of bioptates of the stomach mucosa obtained under spot gastroscopy in patients suffering from the ulcerous disease allowed to reveal intensifying proliferative activity of epithelium in the ulcer margins. Similar changes in LI were found in gastritis, followed by the gland affection, and in atrophic gastritis.

The study investigated a stimulating effect of tris-[2-hydroxyethyl] ammonium salt of iron-containing polyacryl acid on proliferative processes in the connective tissue in development of experimental
gastric ulcer
. The drug was found to exhibit an ulcerostatic effect, to change the levels of collagen, noncollagen proteins, glycosaminoglycan fractions,
DNA
and RNA.

DNA
was extracted from paraffin-embedded gastric cancer tissues, and the possibility of G-->T mutations present at codon 12 of H-ras oncogenes was detected in Chinese gastric cancer patients with polymerase chain reaction (PCR) and oligodeoxynucleotide (ODN) probe hybridization. Point mutations were identified in 5 of 17 gastric cancer cases (29.4%), and in none of 7
gastric ulcer
cases. These results indicate that G-->T mutations at codon 12 of H-ras oncogene may play a role in activating the oncogenes in gastric cancers.

There is increasing evidence that the herpes simplex virus may account for some
gastric ulcer
disease. To examine this possibility, 62 tissue biopsies from 21 patients were obtained during esophagogastroduodenoscopy for gastroduodenal ulcer disease and from one operative specimen during the procedure for perforation of a
gastric ulcer
. The samples were collected from the base and rim of the ulcer, as well as from apparently healthy tissue adjacent to the lesion. When the
DNA
was extracted from these tissues and hybridized to a herpes simplex virus-specific
DNA
probe, positive results were obtained with 9.5 per cent (2 out of 21) of the patients with benign ulcers. Positive signals were obtained only with ulcer-associated tissues and never with healthy tissue. Hybridization also occurred with
DNA
from one ulcerative carcinoma in the study. These data suggest that a subset of ulcer disease may be caused by herpes simplex virus or that this virus may be secondarily associating with these lesions.

A 75-year-old man initially complained of pollakiuria and low abdominal pain, and died of massive bleeding from an exacerbated
gastric ulcer
. The diagnosis of primary cardiac lymphoma was made postmortem. The tumor involved only the epicardium and myocardium, which met the criteria of primary cardiac lymphoma as defined by the Armed Forces Institute of Pathology. The lymphoma consisted of large cells and expressed the B cell marker, CD20. Although chronic inflammation due to chronic renal failure was observed in the pericardium around the lymphoma, polymerase chain reaction (PCR) was conducted to detect monoclonality at the
DNA
level in lymphoma cells, which were shown to comprise a monoclonal population.

The influence of autotransfusions of hemosorbent-treated blood on parameters of
DNA
-synthetic activity of blood lymphocytes was studied in 114 patients with
gastric ulcer
. A statistically significant increase in parameters of
DNA
-synthetic activity of lymphocytes was recorded simultaneously with the appearance of morphological signs of blast transformation on electrograms. A conclusion has been made on association of lymphocyte blastogenesis under the influence of autotransfusions with the process of ulcerous sanagenesis acceleration.

We studied the healing promoting action of Z-103 on the chronic
gastric ulcer
induced by acetic acid (AAU) or Fe-ascorbic acid (FAU) in rats. The area of the gastric ulcers, hydroxyproline (Hyp) and
DNA
contents in the ulcer region were measured as an index of ulcer healing. The area of gastric ulcers was the largest on day 4 and thereafter gradually decreased, but the ulcers still remained at the 14th day. Hyp contents in the ulcer region decreased until the 7th day in both models, and then this level increased. Significant decrease in
DNA
contents in the ulcer region was observed on the 7th day only in FAU. In AAU and FAU, administration of Z-103 (3 mg/kg/day x 2, p.o.) resulted in a significant decrease in the area of gastric ulcers on the 14th day and a significant increase in Hyp contents in the ulcer region on the 7th day as compared with the control group. Z-103 increased the
DNA
contents in the ulcer region on the 4th day in AAU and on the 7th day in FAU. These results suggest that tissue destruction surrounding the ulcer region in AAU and FAU models might occur until the 4th or 7th day after operation, and that the acceleration of ulcer healing by Z-103 on these models may be facilitated by the wound healing action of this drug.

The changes in gastric (fundic and antral) mucosal protein,
DNA
and RNA content, have been investigated in different experimental
gastric ulcer
models of rat. It seems that indomethacin and stress (restraint) ulcer formation were followed by a predominant fundic (oxyntic cell area) mucosal
DNA
increase, while in the antral mucosa this phenomenon was connected to a significant RNA increase. The observed changes of the RNA/
DNA
ratio in the fundic mucosa were interpreted as a sign of accelerated cell renewal. The authors conclude that during ulceration the regenerative processes concomitantly take place.

The healing of acetic acid-induced gastric and duodenal ulcers was examined together with the biochemical indices of growth in gastric and duodenal mucosa in the following three groups of rats: (a) chow-fed, (b) fed an isocaloric liquid diet, (c) fed the liquid diet plus pentagastrin injections (250 micrograms/kg, 3 times/day). Animals received the diet regimen for 10 days from 1 day after induction of ulcer (day 0). Following the feeding regimens, serum gastrin levels, oxyntic gland mucosal
DNA
synthesis, and gastric secretory function were significantly lowered in the rats fed liquid diets.
DNA
synthesis in the duodenal mucosa was not different from the pre-ulcer levels. Pentagastrin significantly restored the
DNA
synthetic and gastric secretory activity of the liquid diet-fed rats toward the levels in the chow-fed group. In the latter group, a significant increase in
DNA
synthesis and levels of serum gastrin was found at day 6 (after 5 days feeding), which corresponded with a rapid, spontaneous healing of ulcers. Feeding rats liquid diet significantly delayed the healing of gastric, but not duodenal ulcers. Repeated administration of pentagastrin accelerated
gastric ulcer
healing in the liquid diet group toward the rate observed in the chow-fed group, but had no effect on the healing of duodenal ulcers. These results indicate that cell proliferation is an important factor in the healing of gastric ulcers.

Pyrrole aldehydephenyl semicarbazone was shown to be an effective anti-ulcer agent in five experimental models in rats, namely, the indomethacin-induced, acetic acid-induced, pyloric ligation-induced and 0.6 mol HCl, absolute alcohol-induced ulcers, at doses of 40-100 mg/kg. Its anti-ulcer activity and characteristics are similar to those of furazolidone. Its oral acute toxicity in mouse is much lower than furazolidone. This compound exhibited mild inhibitory effects on gastric pepsin secretion, caused increases in hexosamine level and decreases of
DNA
content in gastric juice. It showed no influence on gastric acid secretion and was considered to have "cytoprotective action" on the gastric mucosa. However, this compound was found to be ineffective against the stress-restraint
gastric ulcer
model.
