Concerning the effect of the perfusion of ischemic areas of the myocardium by a stroma-free hemoglobin solution was reported after the starvation of the interventricular branch of the left coronary artery (RIVA) in minipigs. The effect of the perfusion is dependent on the kind of the vascular course. The perfusion of the distal part of the starvated coronary artery shows the better reaction in face of the artemalization of the concomitant cardiac vein with regard to the resaturation of the myocardium with oxygen and several hemodynamic control parameters. Histologic examinations of the ischemic myocardium after the arterialization of the cardiac veins showed however that the symplasm preserved its vitality and showed only reversible changes. The applied stroma-free hemoglobin solution needs a further development. However, the use is possible on principle according to the bibliography. Whether a new clinical therapy could be developed by a transitory arterialization of the cardiac veins in acute myocardial ischemia, should be discussed.

The erythropoietic response of snakes was examined after injecting human urinary erythropoietin (Ep), testosterone propionate (TP), and L-thyroxine (T4), separately and in combinations, into starved ophids. The effect of starvation was reflected by a decrease in the number of erythrocytes, a fall in hemoglobin concentration, and a decline in hematocrit. Statistically significant elevation of erythrocyte number, hemoglobin concentration, and hematocrit was observed at 24 hr following the administration of Ep + T4, and Ep + TP + T4 into starved ophids. The erythrocyte number was also increased by T4 treatment at 24 hr. Furthermore, while T4 and Ep individually increased the red blood cell number at 168 hr, T4, TP + T4, and Ep + TP + T4 elevated the hemoglobin concentration and Ep + T4 and Ep + TP + T4 increased the hematocrit value. It is suggested that the influence of any one of the hormones utilized in the present study on blood morphology of fasted snakes depends to a greater extent on the presence or absence of the other hormone(s).

The human leukemia K562 cell line can be induced by 20 micro M hemin to reversibly accumulate embryonic and fetal hemoglobins without any change in the rate of cell division. When we reduced the rate of cell division by glutamine starvation or addition of hydroxyurea, the cells increased by tenfold the basal hemoglobin level of 0.3-0.5 pg Hb/cell. The combined effects of hemin and inhibitors of cell division permitted K562 cells to attain levels of hemoglobin (26-34 pg Hb/cell) close to that found in normal red cells. This superinduction was reversible and cells could be recycled indefinitely. Furthermore, electrofocusing experiments show that the three primary hemoglobin species produced by these cells (Hb Gower 1, Hb Portland, and fetal Hb), were induced, or reinduced, synchronously by inhibitors of cell division but asynchronously by hemin. Differing effects of hemin and inhibitors of cell division were observed in the absence of irreversible differentiation and suggest different molecular mechanisms controlling globin gene expression.

In several pathophysiologic states, i.e., cirrhosis of liver, protein calorie malnutrition, starvation, carbohydrate deprivation, etc., thyroid hormone metabolism is reported to be altered with a decrease in serum T3 and a reciprocal increase in TR3. Uncontrolled diabetes mellitus is a similar state in which glucose does not enter the cells causing cellular starvation and hyperglycemia ensues. Therefore, serum T4, T3, RT3, T3-resin uptake, TSH, and glucose were determined after an overnight fast in 94 male diabetics (aged 28 to 85 years) during a routine follow-up visit to the outpatient clinic and 24 healthy male adults (aged 24 to 81 years). Glycosylated hemoglobin concentrations were measured as well in normal subjects and 16 newly discovered diabetics. In normal subjects, no significant relationships between fasting plasma glucose and T3 and RRT3 levels were observed. In diabetics there was a significant positive (r = 0.611; p less than 0.001) correlation between glucose and RT3. Similarly, a significant negative relationship was observed between glucose and T3 (r = 0.491; p less than 0.001). T4, free T4, T3-resin uptake, and TSH were normal in diabetics. In 16 newly discovered diabetics, with fasting plasma glucose greater than 200 mg/dl, serum T3 rose (96 +/- 5 to 128 +/- 5 ng/dl) and RT3 declined (26.3 +/- 10.4 +/- 1.4 ng/dl) on improvement of hyperglycemia (fasting plasma glucose less than 140 mg/dl) after intensive therapy for 6 to 8 weeks. Glycosylated hemoglobin levels declined as well (14.6 +/- 0.9% to 9.3 +/- 0.7%). These data indicate: (1) thyroid hormone metabolism may be altered in diabetes mellitus with a fall in serum T3 and a reciprocal rise in RT3; and (2) T3 and RT3 concentrations may serve as indicators of metabolic control in diabetes mellitus.

Plasma amino acid profiles along with hemoglobin, hematocrit, albumin, protein, blood urea nitrogen and serum creatinine values for ten patients undergoing abdominal operations were studied before operation and for 16 days there-after at different intervals. Six patients in the control group were studied in a similar manner. From the observations obtained, we concluded that total amino acid valued are a more sensitive reflection of patient nutrition in both the preoperative and postoperative periods. In future, total amino acid levels may become part of the nutritional assessment of a patient undergoing an operation. The histidine levels in plasma remain low for the longest period of time, an indication of a great need for histidine. Hence, greater attention should be paid to the histidine content of a diet or solution administered parenterally, or both. In addition, branched chain amino acids, alanine, glycine, cystine, arginine, lysine, tryptophan and threonine are required in greater quantity than the other amino acids as a result of the increased catabolism and partial starvation of the patients postoperatively. In formulation hyperalimentation solutions, an increased need for these amino acids should be kept in mind.

The activation process of vacuolar proteinases in the yeast Saccharomyces cerevisiae via precursor maturation is initiated by the PRA1/PEP4 gene product, proteinase yscA. Chromosomal deletion of the PRA1/PEP4 locus leads to accumulation of inactive pro-proteinases in the vacuole. Nine active-site mutations of proteinase yscA have been constructed in vitro. All these mutations lead to the expression of proteinase yscA species in vivo that are inactive against the in vitro substrate hemoglobin and the in vivo substrates pro-proteinase yscB and pro-carboxypeptidase yscY. However, three active-site mutations in proteinase yscA sustained the precursor maturation of proteinase yscB and carboxypeptidase yscY after exchange of the genomic wild-type allele with the respective proteinase yscA mutant alleles. In contrast to yeast strains deleted in proteinase yscA, the respective mutants carry out all cellular functions that rely on a proteolytically active vacuole. This wild-type behaviour of proteinase yscA mutant cells is dependent on the presence of active proteinase yscB. Proteinase yscA and proteinase yscB are equally able to fulfil essential cellular functions. For instance, either proteinase is able to maintain viability under starvation. However, mature proteinase yscB is not stable in the absence of proteinase yscA. The wild-type-like conformation of proteolytically inactive mutant proteinase yscA proteins stabilizes mature proteinase yscB and thus enables continuous maturation of pro-proteinase yscB by active proteinase yscB. After inhibition of the proteolytic activity of proteinase yscB in these proteinase yscA mutants with phenylmethysulfonyl fluoride or deletion of the PRB1 gene, maturation of all zymogens investigated in the vacuole, including the proteinase yscA mutant proteins, is blocked. The proteolytic activities of the vacuole in such a strain can be regained, however, by introduction of a wild-type proteinase yscA gene allowing subsequent autocatalytic maturation of wild-type pro-proteinase yscA. This indicates that an initial self-activation process of proteinase yscA is necessary for the activation of vacuolar zymogens.

Twenty patients with anorexia nervosa and a body weight below 60% of the standard weight were studied. One died of starvation; the others survived. Four patients, including the deceased, had such severe weakness that they could not sit up without support, and another five could sit up only from a lateral position. Serum albumin or hemoglobin levels at the beginning of therapy could not be used for nutritional assessment because of dehydration, while increased blood urea nitrogen was associated with acute illness. The present results together with data from previous studies of fatal anorexia indicate that the risk of mortality may be quite low when body weight is above 60% of the standard. We suggest that gross muscle weakness in addition to body weight for height can be a valuable indicator to assess the criticalness in anorexia nervosa.

Ketone bodies are derived from the accelerated beta-oxidation of fatty acids during prolonged starvation or severely impaired carbohydrate metabolism. D-3-hydroxybutyrate (3OHB) is the major ketone circulating in the blood. Fully automated assay of 3OHB using a centrifugal analyzer was developed. The within-run and between-run levels of precision were acceptable, with coefficients of variation of from 0.75% to 4.35%. The recovery was 101.00 +/- 3.71%. The linearity was up to 5 mmol/L. Delayed serum separation even after 24 hours had no effect. The stability of 3OHB at -20 degrees C was greater than that at 4 degrees C when the serum was stored. No significant interference was observed with hemoglobin, bilirubin or triacylglycerol. NaF-treated plasma gave a significant underestimation of 3OHB. There was no significant difference in blood 3OHB between normal (blood glucose < or = 110 mg/dL, n = 87) and hyperglycemic subjects (blood glucose 110-200 mg/dL, n = 42), but when the blood glucose concentration was greater than 200 mg/dL, the difference in blood 3OHB between normal subjects and hyperglycemic patients became significant. The blood 3OHB concentrations increased according to the degree of hyperglycemia. There were sensitive changes in blood 3OHB during the treatment of a patient with diabetic ketoacidosis. The monitoring of blood 3OHB can be used clinically as an index of ketosis and as a signal of metabolic control in diabetes.

Labial salivary glands are found in the majority of insects. They are relatively large, extend back into the thorax, and in Rhodnius, they are cherry red in color due to a pigment derived from traces of hemoglobin absorbed form the gut. In most insects they are acinous shaped, with long excretion channels that present differentiated regions which from salivary reservoirs. The glands may be relatively simple or complexly branched and convoluted. In Rhodnius they are described as being unilobed with no traces of division. The main duct leaves the gland at its anterior extremity. The acini have different kinds of cells but all of them are seen as sources of secretion. Our material has a different shape due to the fact that the animals spent 20 days under starvation conditions. New data are also obtained through treatment with collagenase and HCl. The importance of the study of these glands lies in the fact that it will further understanding of the transmission of Chagas' disease.

Ketones can reactivate the production of fetal hemoglobin (HbF) in vitro and in vivo. A reactivation of HbF by ketones, which are generated during starvation, remains largely speculative. Therefore, we investigated HbF in 31 women with anorexia nervosa or bulimia, using both of these as models of intermittent starvation ketosis. For comparison, we also studied 42 female control subjects matched for age. beta-Hydroxybutyrate levels were higher in patients than in controls (460 +/- 90 v 110 +/- 20 mumol/L; P < .0001). We correlated beta-hydroxybutyrate, metabolic, and hematologic parameters with HbF. HbF was measured with high pressure liquid chromatography. The data were analyzed with logistic regression analysis. An elevated HbF fraction (> 0.87%) was observed four times as often in patients than in controls (29% v 7%, P = .01). After adjustment for age, we found HbF elevations associated with beta-hydroxybutyrate levels (P = .005). No other correlations between the various metabolic/ hematologic parameters and HbF were significant. In conclusion, beta-hydroxybutyrate generated in starvation is associated with increased levels of HbF. Thus, unrestrained lipolysis can produce beta-hydroxybutyrate in sufficient quantities to induce a clinically measurable amount of HbF. These findings suggest that intermittent ketosis might also explain some increases of HbF in type 1 diabetes and pregnancy.
