Revistin, a substance that strongly inhibits the reverse transcriptase activity of murine leukemia virus in our screening system, was obtained from a cultured broth of a soil streptomyces which was closely related to Streptomyces filipinensis. The assay method for the activity was based on the inhibition by a test material of the incorporation of 3H-dTMP into DNA synthesized by the reverse transcriptase of an oncogenic RNA virus. Crude revistin was isolated by serial procedures of salting out with ammonium
sulfate
and precipitation with cetylpyridinium chloride. The crude material showed neither antibacterial nor antifungal activity. It exhibited against
splenomegaly
in mice caused by Rauscher leukemia virus infection.

The polypeptides of serologically related viruses of hemorrhagic enteritis (HE) in turkeys, marble spleen disease (MSD) in pheasants, and
splenomegaly
in chickens (SMC) were separated by sodium dodecyl
sulfate
-polyacrylamide gel electrophoresis (SDS-PAGE) and analyzed by protein immunoblotting with polyclonal antibodies to HE virus (HEV). The viral polypeptides II, III, IV, V, VI, and VII were detected on SDS-PAGE with the size range from 18 to 97 kDa in HEV. Viral polypeptides II, III, V, VI, and VII were detected in MSD virus and virus of SMC. Protein immunoblotting of viral proteins with anti-HEV serum revealed antigenic differences between the 3 viruses of avian adenovirus type-II examined. The differences were that the polypeptides II, III, IV, V, VI, and VII were identified in HEV and the polypeptides II, V, VI, and VII were identified in MSD virus and virus of SMC. The bands of penton base (polypeptide III) and fiber (polypeptide IV) were seen in HEV only by protein immunoblotting.

Lipopolysaccharide (LPS) fractions were obtained from smooth cultures of Brucella abortus strains 2308 and S-19 by butanol extraction procedures. The LPS from the initial butanol extraction contained 10 to 15% protein and was reduced to less than 1% protein by treatment with proteinase K. Mouse C127I cells were transformed with a chimeric plasmid consisting of bovine papillomavirus DNA and human granulocyte-colony-stimulating factor (G-CSF) cDNA placed under the control of the SV40 early promoter. Protein and lipid analyses were conducted on isolated erythrocyte and lymphocyte plasma membranes from 7-wk-old male C57BL copper-deficient and copper-supplemented mice to investigate mechanisms for the altered immunity that accompanies dietary copper deficiency. Beginning at parturition, dams were fed a diet low in copper (0.5 mg/kg) and the offspring were weaned to this diet. on the developing chick embryos became lower when the treatment was done at later developmental stage. When AG was injected on the 5th day of incubation, the frequency of retardation of development of the body and liver, aplasia of gallbladder and
enlarged spleen
was highest without high mortality compared with the other experiments. Embryos with liver damage could not hatch. 14C-AG hydrochloride injected on the 5th day of incubation easily distributed into the embryo through the yolk from the thin albumen of the injection site, but when injected on the 9th day, the agent transferred very little into the yolk and embryo. The change of transport in the eggs with development may elucidate the difference of lethality of AG. Additionally, it was confirmed using thin layer chromatography that a large part of the radioactivity in the embryo might be an unknown metabolite of AG and the substance was accumulated with extremely high concentration in the liver at 24-48 hr after the injection of AG on the 5th day of incubation. This is probably the main cause of the peculier abnormalities in the liver of chick embryos.

showed low lethality at the intraperitoneal administration against young mice and rats. When the maximum tolerated dose of AG
sulfate
(750 mg/kg in mice and 500 mg/kg in rats) was intraperitoneally injected into pregnant mice and rats on day 0 to 6 or day 7 to 13 of gestation, the incidence of resorption was comparatively high in the day 7 to 13 group of both species, but severe abnormalities as observed in the chick embryos and other external abnormalities were not induced in rodent fetuses. 14C-AG hydrochloride intraperitoneally injected to pregnant mice on day 12 was widely distributed in the maternal body and fetuses within 1 hr and rapidly excreted. Furthermore, AG might be hardly metabolized in mice, while a metabolite clearly appeared in chick embryos. Therefore, the lower toxicity of AG on rodent animals as compared with chick embryos may be due to the fact that the agent is not metabolized and rapidly excreted from maternal body and fetuses.

We surveyed the apoA-I gene of the patient and five of his family members by direct sequencing after amplification by polymerase chain reaction and found a codon 8 nonsense mutation (TGG --> TAG, Trp --> stop) in exon 3 of the apoA-I gene. The results of a pedigree analysis by DNA sequencing and restricted fragment length polymorphism (Sty I) were consistent with an autosomal codominant trait. Coronary angiography was performed to evaluate coronary atherosclerosis, but no significant luminal narrowing was detected. An intracoronary ultrasound study showed mild intimal hyperplasia in segment 6. In summary, this is a case of apoA-I deficiency without evidence of coronary heart disease.
