Powdered, sterilized zinc polycarboxylate was implanted subcutaneously in 56 young hamsters and 54 mice, using equal numbers of each sex, with 24 sham operated hamsters and 26 mice as controls. The hamsters were sacrificed after 15 months and the mice after 12. Implant material was recovered from one of 42 surviving mice and from 16 of 43 surviving hamsters. Benign adenomas of the gut, thyroid, sebaceous glands and ovary were found in 3 experimental mice and in one control. Benign adrenal adenomas were found in 3 experimental hamsters and in one control. One control hamster developed a rhabdomyosarcoma in a limb. One experimental hamster developed a leiomysarcoma in close relation to an implant.

Previously we showed for coxsackievirus A9 (CAV-9) that specific interactions between the RGD motif of capsid protein VP1 and the alpha v beta 3 integrin are involved in virus binding and entry into green monkey kidney cells (GMK) and some other cell lines. The RGD-recognizing alpha v beta 3 integrin is known as the vitronectin receptor (VNR). During replication in the gut, CAV-9 like all other enteroviruses are exposed to host proteolytic enzymes, and we showed previously that the RGD-containing 15 amino acids long carboxy terminal extension of VP1 is cleaved off by trypsin. The trypsin-treated CAV-9 was still infectious, although at an apparently reduced level as assessed in GMK cells. This indicated that the virus was able to bypass the RGD-dependent entry and possibly use an alternative receptor. We have now found that in RD cells, a human rhabdomyosarcoma cell line, neither RGD-containing oligopeptides nor polyclonal antiserum to VNR are able to protect the cells from CAV-9 infection suggesting that the RGD motif is not involved in binding or entry of the virus into these cells. This result was further confirmed by demonstrating that, in RD cells, the trypsin-treated CAV-9 lacking the RGD-containing insert appeared to be as infectious as the untreated virus. The most striking difference between the virus receptors in the RD and the GMK cells was seen when the rate of virus uncoating was studied. For virus particles bound to the RD cells, the uncoating step started already at 18-20 degrees C and the process went on rapidly at 36 degrees C resulting in complete disintegration of cell-bound virions. In contrast, alpha v beta 3-bound virus particles in the GMK cells appeared to uncoat slowly even at 36 degrees C and during the 90 min observation period only a small, hardly visible fraction was found to be disintegrated. Trypsin-cleaved CAV-9 showed the rapid disintegration kinetics in GMK cells as well suggesting that these cells contain both types of receptor specificity. These results indicate that CAV-9 is able to use two different entry routes into host cells depending on the target cells and on phenotypic properties of the virus regulated by host proteases.

The purpose of this study was to assess the diagnostic value of magnetic resonance cholangiopancreatography (MRCP) in studying the anatomy, sites, and causes of obstructive jaundice. From September 1994 to May 1996 three-dimensional MRCP was performed on 31 patients with abdominal pain and obstructive jaundice with a fast spin-echo T2-weighted pulse sequence. The images were reconstructed using maximal intensity projection, AVERAGE and SURFACE algorithm processing techniques at a graphics workstation. All the reconstructed images were compared with those obtained using conventional cholangiographic techniques, such as endoscopic retrograde cholangiopancreatography, percutaneous transhepatic cholangiography and drainage, and intraoperative cholangiography. The patients' diagnoses included choledochal cyst (13), cholangiocarcinoma (five), choledocholithiasis (four), pancreatic head carcinoma (three), rhabdomyosarcoma (one), papillary Vater carcinoma (one), recurrent gastric carcinoma (one), ascaris (one), and biliary atresia (two). Extrahepatic biliary dilatation was present in all 13 patients with choledochal cyst; the pancreatic ducts and their entrance level to the common bile duct were observed in eight of these patients. The level of obstruction in patients with cholangiocarcinoma was well documented but the biliary tract of one patient with biliary atresia was not identified by MRCP. In one patient with biliary rhabdomyosarcoma, MRCP clearly delineated the extrabiliary extension of the tumor. In a patient with ascaris in the common bile duct an increase in signal intensity inside the digestive tract of the worm denoted fluid in its gut. Lithiasis was shown in all of the four patients with choledocholithiasis. Thus, MRCP is a useful tool in the assessment of biliary tract obstruction and its causes, and is a valuable addition to ultrasonography.

A 69-year-old woman was diagnosed with alveolar rhabdomyosarcoma (ARMS) of the nasopharynx. She has a history of catastrophic thromboembolic event in the abdomen that caused short-gut syndrome and dependence on total parenteral nutrition (TPN) twelve hours per day. She was treated for short-gut syndrome with teduglutide, a glucagon-like peptide-2 (GLP-2) analog, which led to reduction of TPN requirements. However, a few months later, she developed metastatic alveolar rhabdomyosarcoma. Though a causative relationship is unlikely between the peptide and ARMS due to the brief time course between teduglutide therapy and sarcoma diagnosis, neoplastic growth may have been accelerated by the GLP-2 analog, causing release of IGF-1. The transmembrane receptor for IGF-1 is frequently overexpressed in ARMS and is implicated in cell proliferation and metastatic behavior. This case describes a rare incidence of metastatic alveolar rhabdomyosarcoma in a sexagenarian and possibly the first case reported associated with the use of teduglutide. Teduglutide was discontinued due to a potential theoretical risk of acceleration of sarcoma growth, and the patient's rhabdomyosarcoma is in remission following sarcoma chemotherapy.

The incidence of pediatric cancer is lower than that of adult cancer worldwide. However, the former has detrimental side effects on the health of individuals, even after the cancer is cured, due to the impact of treatment on development. Recently, correlations have been made between the gut microbiome and cancer in several studies but only on adult participants. There is always a complication of dealing with pediatric cancer treatment protocols because they usually include a combination of chemotherapy, radiotherapy, and intensive prophylactic antibiotics. In the current study, a pilot study was conducted to analyze ten fecal samples from three pediatric cancer patients, suffering from rhabdomyosarcoma near their pelvic region, and two healthy individuals. A correlation between microbial composition and response to treatment was reported, in which the responders had generally a lower microbial diversity compared to non-responders. In addition, nucleotide changes and deletions in the tested 16S rRNA sequences post radiotherapy were detected. Despite the small sample size used in the experiments due to the uncommon rhabdomyosarcoma in children, the results can help in understanding the influence of radiotherapy on the gut microbiome in pediatric cancer patients. More work with larger sample size and different cancer types need to be conducted to understand the influence of radiotherapy on gut microbiome to mitigate the deleterious impact of radiation on treated children.
