Layer hens (310 days old) affected with subcutaneous tumours were investigated pathologically. Basopholic intracytoplasmic viral matrix inclusions (MIs) were widely distributed in the chickens affected with subcutaneous myxoma rhabdomyosarcoma, perineuroma, glioma, intra-abdominal adenocarcinoma, and nephroblastoma. MIs were observed in the myocardial cells and the impulse-conducting-system cells. They were also present in the smooth muscle cells of the arteries in the spleen and lungs, in the smooth muscle of the digestive tract muscular layer (crop, oesophagus, proventriculus, gizzard, duodenum, jejunum, ileum, caecum, and large intestine), and in the smooth muscle of the capsule in the ovary and pancreas. They were also observed in the podocytes of glomeruli and renal epithelium in the kidneys, tumour cells of nephroblastoma, chondrocytes of the trachea, squamous-epithelial cells of the pharynx, and nerve cells of the cerebrum and tumour cells of the glioma in the cerebellum. Histochemically, MIs were stained with RNA, but not with DNA. MIs in the various tissues were strongly positive for the avian leukosis virus (ALV) antigen. Ultrastracturally, MIs were found in the cytoplasm of myocardial cells and podocytes of renal glomeruli. They consisted of electron-dense small granules and ring-shaped particles. Viral particles were observed in the vesicles of the cytoplasm of myocardial cells and glomerular podocytes. The gene product specific for subgroup A of ALV was detected in the livers or tumours by reverse transcription-polymerase chain reaction. This result suggests that MIs can be formed in organs rather than muscular systems in the chickens naturally affected with ALV-associated tumours.

An adult free-living European robin (Erithacus rubecula) with a large, firm, subcutaneous mass on the pectoral muscle was examined. The bird was unable to fly and died spontaneously. Necropsy revealed a yellowish, bilobate mass almost completely replacing the pectoral muscles with extensive osteolysis of the keel bone. Histopathology revealed a poorly demarcated, highly cellular sarcomatous tumour with metastases to the lungs, pulmonary blood vessels and heart. Immunohistochemistry was negative for neuron-specific enolase, S-100 protein and the p-27 major capsid protein of avian leukosis viruses. The homogeneously positive immunolabelling for vimentin and scattered positivity for myoglobin and desmin suggested a diagnosis of rhabdomyosarcoma. A retrospective examination of the records for 194 birds of the thrush family, including 64 robins submitted over a 20-year period, showed no diagnoses of neoplasia.

In the 10 years to November 1987, 33 different types of naturally occurring neoplasms were diagnosed in 2281 domestic fowls submitted for necropsy examination. The most prevalent type was lymphoma (1825/2281 = 80%) including two cases in chickens from a flock free from Marek's disease virus, exogenous avian leukosis viruses and reticuloendotheliosis virus; the remaining lymphomas were diagnosed on gross and/or histological criteria as Marek's disease (1069) or lymphoid leukosis (754). Of the non-lymphoid neoplasms, leiomyomas of the ventral ligament of the oviduct (99) were the most common followed closely by metastatic abdominal adenocarcinomas (93) and haemangiomas or haemangiosarcomas (91). A large number of fibromas and fibrosarcomas (40), and a smaller number of myxomas and myxosarcomas (17) were observed. Three fibrosarcomas and two myxosarcomas were presented as metastatic abdominal neoplasms, and two fibromas and seven myxomas were found on the rostral extremity of the upper beak of hens, including five cases of myxoma from one flock. Other less prevalent neoplasms described were myelocytomas or myeloblastomas (12), avian keratoacanthomas (11), granulosa-theca cell tumours (11), oviduct adenomatosis and adenocarcinomas (10) and ovarian adenocarcinomas (8). Small numbers of some neoplasms or well-characterized variants were encountered, namely histiocytic sarcomas (5), lipomas (4), liposarcomas (2), reticulum cell sarcomas (3), plasmacytomas (3), intracutaneous keratinizing epitheliomas (3), feather folliculomas (2), proventricular adenomas (2), hepatocellular adenocarcinomas (2), cholangiocellular adenomas and adenocarcinomas (6), pancreatic adenocarcinomas (3), nephroblastomas (7), astrocytomas (5), pinealomas (3), Schwannomas (3) and multifocal melanomas (3). Single examples were found of osteoma, osteosarcoma, rhabdomyosarcoma, leiomyoma of the gizzard wall, intestinal adenocarcinoma, renal adenocarcinoma, adrenocortical adenoma and ultimobranchial cyst.

ABSTRACT The prototype fowl glioma-inducing virus (FGVp) causes fowl glioma and cerebellar hypoplasia in chickens. In this study, we investigated whether a strain of avian leukosis virus (ALV), associated with avian osteopetrosis and mesenchymal neoplasms, is able to induce fowl glioma. We encountered avian osteopetrosis and mesenchymal neoplasms, including myxosarcoma and rhabdomyosarcoma, in Japanese native chickens used for both egg-laying and meat production. These birds were also affected by non-suppurative encephalitis and glioma in their brains. Four ALV strains (GifN_001, GifN_002, GifN_004, GifN_005) were isolated, and a phylogenic analysis of envSU showed that these isolates were classified into different clusters from FGVp and the variants previously reported. Whereas the envSU shared a high identity (94.7%) with that of Rous sarcoma virus (strain Schmidt-Ruppin B) (RSV-SRB), the identity between envTM of GifN_001 and that of FGVp was high (94.5%), indicating that GifN_strains may emerge by recombination between FGVp and other exogenous ALVs. Specific-pathogen-free chickens inoculated in ovo with GifN_001 revealed fowl glioma and cerebellar hypoplasia. These results suggest that the newly isolated strains have acquired neuropathogenicity to chickens.
