About one-third of the more than 300 C2H2 zinc finger proteins of Drosophila contain a conserved sequence motif, the zinc finger-associated domain (ZAD). Genes that encode ZAD proteins are specific for and expanded in the genomes of insects. Only three ZAD-encoding gene functions are established, and the role of ZAD is unknown. Here we present the crystal structure of the ZAD of Grauzone (ZAD(Grau)), a Drosophila transcription factor that specifically controls the maternal Cdc20-like APC subunit Cortex. ZAD forms an atypical treble-clef-like zinc-coordinating fold. Head-to-tail arrangement of two ZAD(Grau) molecules in the crystals suggests dimer formation, an observation supported by crosslinking and dynamic light scattering. The results indicate that ZAD provides a novel protein-protein interaction module that characterizes a large family of insect transcription factors.

To investigate the role of phenyl radical in the growth of PAHs (polycyclic aromatic hydrocarbons), pyrolysis of toluene with and without benzene has been studied by using a heatable tubular reactor couple with an in-situ sampling vacuum ultraviolet (VUV) single photon ionization (SPI) time-of-flight mass spectrometer (TOFMS) at temperatures 1155-1467 K and a pressure of 10.02 Torr with 0.56 s residence time. When benzene was added, a significant increase of phenyl addition products (biphenyl, terphenyl, and triphenylene) was observed and the mass spectra showed a clear regular sequence with an interval of approximately 74 mass number, corresponding to the phenyl addition (+C6H5) followed by H-elimination (-H) and cyclization (-H2). The analysis showed that the PAC (phenyl addition/cylization) mechanism is efficient for the growth of PAHs without a triple fusing site, for which the HACA (hydrogen abstraction/C2H2 addition) step is inefficient, and produces PAHs with five-membered rings. The PAC process was also suggested to be efficient in the subsequent growth of PAHs with five-membered rings. The role of the PAC mechanism in combustion conditions is discussed in relation to the importance of disordered five-membered ring structure in fullerene or soot core.

Glandular trichome is specialized multicellular structures that have capability to synthesize and secrete secondary metabolites and protect plants from biotic and abiotic stresses. Our previous results revealed that the C2H2 zinc-finger transcription factors (GIS) acts upstream of GL3/EGL3-GL1-TTG1transcriptional activator complex to regulate trichome initiation through GA signal in Arabidopsis. In the present study, we are reporting that ectopic expression of AtGIS could regulate glandular trichome development through GA signaling in tobacco. X-gluc staining of various organs from transgenic plants showed that AtGIS expressed mainly in the glandular trichomes. Statistical analysis demonstrated that over expression of GIS increased significantly glandular trichome production on the leaf, stem, branch, and sepal in tobacco. After PAC treatment, reduction of glandular trichome production in transgenic plants was more severe with compared to wild type plants. Furthermore, GA treatment could induce expression of AtGIS. More importantly, our results also demonstrated that overexpressed AtGIS significantly affect the main components of trichome exudates, such as significantly increase the content of nicotine, Cembratriene-4, 6-diol. Taken together, these results suggest that ectopic expression of AtGIS regulates glandular trichome development and may play a key role in compounds secretion in tobacco.
