To investigate the
NMR
relaxation times for irradiated rat lung tissue, we measured T1 and T2 at 11 different times during the injury's 1-year time course. A biexponential analysis of T2 was used to determine T2 fast (T2f) and T2 slow (T2s). In addition, we measured water content and correlated changes in the relaxation times with pathological changes. The correlation indicates the following: (1) Shortly after irradiation, the biexponential T2 decay for 1/3 of the samples became monoexponential and there were no noticeable pathological changes observed using light microscopy. (2) During radiation
pneumonitis
, T2f and T2s were prolonged. This accompanied acute edematous changes and inflammatory cell infiltration. (3) Finally, during radiation fibrosis T1 shortened and collagen increased. We observed no significant correlation between relaxation time changes and water content changes throughout the 1-year time course.

The purpose of this study was to establish the proton
NMR
relaxation times of collapsed but otherwise normal lung tissue and to determine whether an inflammatory process within a collapsed lung can be detected by alterations in relaxation times. The lungs of three groups of rabbits were studied: group A (n = 7) had a sterile collapse of one lung for two days. The two other groups also had one lung collapsed, but with bacterial (group B, n = 6) or chemically induced (group C; n = 6)
pneumonitis
superimposed. The contralateral lung, which was acutely deflated at the time of thoracotomy, served as a control in each animal. T1, T2 and the total water content were measured on freshly excised lung samples. In group A, there was no significant difference in T1 (606 +/- 14* ms vs. 595 +/- 18 ms;* = SEM) or T2 (80.6 +/- 1.7 ms vs. 78.4 +/- 2.6 ms) between the collapsed and the control lung tissue. In each animal in groups B and C, T2 was longer in the collapsed lung with superimposed
pneumonitis
than in the control lung tissue (group B: 116.8 +/- 6.9 ms vs. 82.9 +/- 1.8 ms, P less than .001; group C: 120.5 +/- 5.9 ms vs. 86.0 +/- 1.5 ms, P less than .001). T1 changes were similar, but less marked. There was a linear relationship between the relaxation times and the total water content of the lung samples (T1:r = 0.87; T2:r = 0.91). In the present communication we present the results of surgical treatment of tumours of the adrenal glands, basing on the material from our clinical department. During 1983-1997 at the Department of Clinical Urology Central Clinical Hospital of the Military University School of Medicine (CSK WAM) 27 patients (15 men, 12 women) with tumors of the adrenal glands were treated. Among the operated patients 3 were diagnosed with Cushing's syndrome, 3 with pheochromocytoma, 1 with Cohn's syndrome, while in the remaining 20 the tumours were hormonally inactive. Surgical treatment included 15 right, 10 left and 2 bilateral adrenalectomies. There was no intraoperational mortality. In all patients with hypersecretion, the symptoms disappeared after adrenalectomy. The size of tumours ranged 4-20 cm. In 3 patients there appeared a need for broadening of the size of surgical intervention--2 splenectomies and 2 nephrectomies and 1 resection of pancreatic tail--all due to regional infiltrations with tumorous tissues. During surgery there occurred 2 cases of iatrogenic pleuric injury. Postoperative complications included 1 case of
pneumonia
and 1 case of wound infection. In case of unilateral tumours of the adrenal glands the best method for surgery appears to be the lateral (lumbal) resection and in case of large tumours and/or bilateral tumours the most convenient appears to be the anterior (transperitoneal) resection. Streptococcus pneumoniae is a major human pathogen that causes high mortality and morbidity rates and has developed resistance to many antibiotics. The genome of S. pneumoniae has recently been completely sequenced revealing many genes encoding hypothetical proteins of unknown function. We have found that the gene encoding one such conserved protein, SP14.3, is essential for growth of S.
pneumonia
. Since it is essential, SP14.3 represents a potential target for drug discovery. Here, we describe the three-dimensional solution structure of SP14.3 as determined by
NMR
spectroscopy. The structure consists of two domains each with an alpha/beta-fold. The N-terminal domain contains two alpha-helices and a three-stranded beta-sheet, while the C-terminal domain is composed of one alpha-helix and a five-stranded beta-sheet. The N-terminal domain of the protein contains a highly negatively charged surface and resembles the fold of the N-terminal domain of Thermus thermophilus ribosomal protein S3. The C-terminal domain has a protein fold similar to human small nuclear ribonucleoprotein Sm D3 and Haloarcula marismortui ribosomal protein L21E. The two domains of the protein tumble in solution overall as a whole with an overall molecular rotational correlation time (tau(m)) of 12.9 ns at 25 degrees C. The relative orientation of the two domains is not defined by the nuclear Overhauser effect data. Indeed, residual dipolar couplings and the structure calculations indicate that the relative orientation of the two domains is not rigidly oriented with respect to one another in solution.

The sterol composition of Pneumocystis carinii, an opportunistic pathogen responsible for life-threatening
pneumonia
in immunocompromised patients, was determined. Our purpose was to identify pathway-specific enzymes to impair using sterol biosynthesis inhibitors. Prior to this study, cholesterol 15 (ca. 80% of total sterols), lanosterol 1, and several phytosterols common to plants (sitosterol 31, 24alpha-ethyl and campesterol, 24alpha-methyl 30) were demonstrated in the fungus. In this investigation, we isolated all the previous sterols and many new compounds from P. carinii by culturing the microorganism in steroid-immunosuppressed rats. Thirty-one sterols were identified from the fungus (total sterol = 100 fg/cell), and seven sterols were identified from rat chow. Unusual sterols in the fungus not present in the diet included, 24(28)-methylenelanosterol 2; 24(28)E-ethylidene lanosterol 3; 24(28)Z-ethylidene lanosterol 4; 24beta-ethyllanosta-25(27)-dienol 5; 24beta-ethylcholest-7-enol 6; 24beta-ethylcholesterol 7; 24beta,-ethylcholesta-5,25(27)-dienol 8; 24-methyllanosta-7-enol 9; 24-methyldesmosterol 10; 24(28)-methylenecholest-7-enol 11; 24beta-methylcholest-7-enol 12; and 24beta-methylcholesterol 13. The hypothetical order of these compounds poses multiple phytosterol pathways that diverge from a common intermediate to generate 24beta-methyl sterols: route 1, 1 --> 2 --> 11 --> 12 --> 13; route 2, 1 --> 2 --> 9 --> 10 --> 13; or 24beta-ethyl sterols: route 3, 1 --> 2 --> 4 --> 6 --> 7; route 4, 1 --> 2 --> 5 --> 8 --> 7. Formation of 3 is considered to form an interrupted sterol pathway. Taken together, operation of distinct sterol methyl transferase (SMT) pathways that generate 24beta-alkyl sterols in P. carinii with no counterpart in human biochemistry suggests a close taxonomic affinity with fungi and provides a basis for mechanism-based inactivation of SMT enzyme to treat Pneumocystis pneumonia.

Continued investigations into the phenolic content of the leaves and stems of C. intermedia yielded tyrosol and a methoxy analogue, 2-[4-[O-alpha-apiofuranosyl-(1' '-->6')-beta-d-glucopyranosyloxy]phenyl]ethanol, 4-[O-alpha-apiofuranosyl-(1' '-->2')-beta-d-glucopyranosyloxy]benzaldehyde, five glycosylated flavonols, two isoflavones, four flavanones, two isoflavones, and two flavones. , CD, and MS methods. Because flavonoids are presumed to contribute significantly toward the scavenging effects of active oxygen species, our results indicate that the tentative claimed health-promoting properties may be attributed to the presence of these and other phenolics in C. intermedia.

Other major sterols were (24S)-ergost-7-en-3beta-ol, (24S)-stigmast-7-en-3beta-ol, and (24S)-stigmasta-5,7-dien-3beta-ol. The sterols of the hosts were found to be very different from those of the fungi. The 24-alkyl sterols of the fungi had the 24alpha-configuration, whereas those of the hosts had the 24beta-configuration. Similarities to the sterol composition of the AIDS
