One of the major clinical findings in Alzheimer's disease (AD) is the formation of deposits of beta-amyloid protein in amyloid plaques, derived from the beta-amyloid precursor protein (beta-APP). To determine the possible use of beta-APP as a diagnostic marker for AD in

from an Ommaya reservoir implanted in the lateral ventricle several months earlier at the time of transplantation of adrenal medulla to caudate nucleus and could detect dopamine only in its sulfconjugated form. Peak
CSF
levodopa and dopamine sulfate levels occurred 1 to 1.5 hours after peak plasma concentration of levodopa. The time course of clinical improvement and worsening correlated precisely with the appearance and disappearance of both levodopa and dopamine sulfate in the
CSF
. The precise correlation between
CSF
dopamine sulfate and levodopa indicates that in patients with advanced PD the brain retains some capacity to convert levodopa to dopamine. The transient nature of the correlation between motor fluctuations,
CSF
levodopa, and
CSF
dopamine sulfate is consistent with suggestions that in patients with advanced PD there is a diminished capacity to store dopamine synthesized from exogenous levodopa. Dopamine sulfate appears to be a useful index of the availability of dopamine in the parkinsonian brain.

We have measured somatostatin-like immunoreactivity SLI in cerebroventricular fluid of patients with
Parkinson's disease
(PD) and other extrapyramidal disorders with hyperkinesia. Patients with PD showed a significantly lower concentration of SLI when compared with levels in control patients with chronic stable multiple sclerosis or temporal lobe epilepsy. Less markedly decreased levels of SLI were also noted in patients with torsion dystonia. Of two patients with Huntington's disease one showed a high and one a medium concentration of SLI. According to the site of the stereotactic cannula, verified by ventriculopathy, SLI concentrations in
CSF
specimen obtained from the foramen Monro tended to be higher than in specimen from a supraforaminal level. Of 5 other patients with lateral and third ventricle being accessible during the passage of the stereotactic cannula, 4 showed higher SLI concentrations in the third ventricle compared to the lateral ventricle. , 10 with Alzheimer's disease, 7 with Huntington's chorea). We applied a battery of neuropsychological tests to determine the degree of dementia and to exclude the presence of mood alterations.

Ferritin levels were measured in postmortem brain tissue from patients dying with
Parkinson's disease
[treated with L-3,4-dihydroxyphenylalanine (L-DOPA)] and from control patients. Ferritin levels were decreased in the substantia nigra, caudate-putamen, globus pallidus, cerebral cortex, and cerebellum when compared with age-matched control tissues. However, in

from L-DOPA-treated patients and in serum from L-DOPA-treated and untreated parkinsonian patients, ferritin levels were normal. Previous studies have suggested an increased total iron content in substantia nigra of parkinsonian brain. The failure of substantia nigra ferritin formation to be stimulated by increased iron levels suggests some defect in iron handling in this critical brain region in

. The reason for decreased ferritin levels throughout the parkinsonian brain is not clear but does not seem to reflect a general system deficit in ferritin.

was removed 7 d, 5 months and 1 year after operation. When added to 18 d neuronal cultures for 3 d, the 7 d

caused neuronal cell and a glial reaction. The 4 months

caused cell death, but markedly less than the 7 d

. One year after transplantation the

had no toxic effects; these cultures were similar to control cultures. It is concluded that

from PD patients may contain aggressive IgG-species specific for DA neurons, and that the amount of such antibodies decrease after adrenal transplant operations. It is suggested that neurodegenerative diseases may become aggravated by autoimmune reactions.
