A simplified and accurate method for determination of naturally occurring vitamin D3 in bovine milk was established by high-performance liquid chromatography (HPLC) using successively reversed-phase and straight-phase columns. Exactly 25.0 ml of a sample of bovine milk was taken and the lipid was extracted with a solvent mixture of petroleum ether and ethyl ether (1:1) with small amounts of ethanol and Triton X-100, present. The extracted lipid was subjected to the first preparative HPLC using a Nucleosil 5C18 column (reversed-phase type) with acetonitrile-methanol (1:1) as the mobile phase, and a fraction containing vitamin D3 was isolated. The fraction was subsequently subjected to the second analytical HPLC using a Zorbax SIL column (straight-phase type) with 0.4% isopropanol in n-hexane as the mobile phase. Vitamin D3 was assayed by estimating the peak height on the chromatogram. The overall recovery and CV values were 92.1 +/- 8.7% and 9.4%, respectively, which were satisfactory. The proposed method was applied to several kinds of colostrum, early and later bovine milk in pairs. The assayed values in the colostrum of the group with large amounts of vitamin D3 administered before delivery to prevent parturient paresis were higher than those in the group with no administration. However, the values of the former group generally decreased in the respective early and later milk in step and there were few differences among those in the later milk of the two groups. Sixty-four cows with parturient paresis were given a standard treatment of 500 ml calcium gluconate (10.7 g Ca++) infusion solution. Twelve of these cows (experimental group = EG 1) additionally received 420 micrograms 1-alpha-hydroxycholecalciferol (1-alpha-OHD3) with 4 mg 25-hydroxycholecalciferol (25-OHD3) by the intramuscular route. A further 9 and 10 cows were given 210 micrograms 1-alpha-OHD3 and 2 mg 25-OHD3 (EG 3) with the calcium infusion, while 10 cows received either 420 micrograms 1-alpha-OHD3 (EG 4) or 210 micrograms 1-alpha-OHD3 (EG 5). Twenty-three cows (EG 2 and 4) which were given a calcium infusion alone acted as controls. The incidence of recurrence could not be reduced by the administration of Vitamin D3 metabolites concurrently with a calcium infusion, although calcium and phosphorus levels were higher between 24 and 48 hours after calcium infusion alone. The combination of 1-alpha-OHD3/25-OHD3 at the higher dosage resulted in the highest calcium levels.

The Blank Park Zoo began suffering mortalities in the spring of 2012 within a flock of 229 captive budgerigars (Melopsittacus undulatus) housed in an interactive public-feeding aviary. Clinical signs in affected birds included weakness, posterior paresis, inability to fly, or acute death. Gross and microscopic lesions were not initially apparent in acutely affected deceased birds. Many birds had evidence of trauma, which is now hypothesized to have been related to the birds' weakness. Investigation into the cause(s) of morbidity and mortality were complicated by the opening of a new interactive enclosure. For this reason, environmental conditions and husbandry protocols were heavily scrutinized. Microscopic examination of dead budgies later in the course of the investigation revealed mineralization of soft tissues consistent with hypervitaminosis D. Pooled serum analysis of deceased birds identified elevated vitamin D3 levels. Vitamin D3 analysis was performed on the feed sticks offered by the public and the formulated maintenance diet fed to the flock. This analysis detected elevated levels of vitamin D3 that were 22.5-times the manufacturer's labeled content in the formulated diet. These findings contributed to a manufacturer recall of more than 100 formulated diets fed to a wide variety of domestic and captive wild animal species throughout the United States and internationally. This case report discusses the complexities of determining the etiology of a toxic event in a zoologic institution.
