Two murine IgG2Ak monoclonal antibodies (703D4, 704A 1) were produced and characterized after immunization with a human large cell lung cancer line (NCI-H 157). These antibodies detect different epitopes on 31 kilodalton [35S]methionine incorporating protein(s). Radiobinding and immunohistochemical studies show these antibodies bind to most (11/13) human non-small cell lung cancer (adenocarcinoma, epidermoid, and large cell), but not to small cell lung cancer (0/11) tumors tested. The epitopes these antibodies recognized are also expressed on human melanomas (7/8), two other tumors (osteogenic sarcoma, renal cell carcinoma), but not on many other human tumors (breast, colon, neuroblastoma, lymphoid), and not on a panel of normal adult human tissues. Because the antigen(s) are preserved after fixation and because of their ability to distinguish lung cancer types from each other and normal tissues, they should be of clinical, as well as of biologic interest.

A retrospective review of 112 cases treated by one author (J.C.) over the 21 year period 1953-1973 is performed. In 62% of the cases squamous cell carcinoma was the histologic diagnosis, with adenoid cystic carcinoma accounting for a further 16%. The remaining histologic types were few in number and included fibrosarcoma, muco-epidermoid carcinoma, melanoma, chondrosarcoma, adenocarcinoma, malignant Schwannoma, lymphosarcoma, malignant mixed tumor, rhabdomyosarcoma, angiosarcoma, and osteogenic sarcoma. Three years following diagnosis more than 1/3 of the squamous cell carcinomas were alive but by 10 years the survival rate had fallen to 16%. Orbital involvement at the time of presentation or noted at surgery had a poor prognosis with a 17% five year cure rate and only 2% alive at 10 years. Erosion of the pterygoids or pterygopalatine fossa invasion as diagnosed radiologically or intra-operatively revealed all patients dead of disease within five years despite radical surgery and irradiation. Cervical nodal metastases at primary presentation was a particularly ominous finding with all patients dead of disease within three years. Metastases developing later had a better prognosis with 39% surviving five years when treated aggressively.

Wistar rats treated with cyclophosphamide, total lymphoid irradiation (TLI), and/or cyclosporin A (CSA) develop a state of immune suppression permitting the growth of tumor xenografts. Experiments were carried out on this newly developed model to investigate the growth patterns of a mouse osteosarcoma and a human colon adenocarcinoma. The combination of cyclophosphamide and CSA permitted a limited period of growth of the mouse osteosarcoma with a tumor take rate of 66%. No takes were observed with the human adenocarcinoma. The combination of cyclophosphamide and TLI resulted in a period of immunosuppression followed by recovery of the immune status. During the period of immunosuppression, tumor xenografts showed a 100% take rate. The most efficient immunosuppression was achieved by a combination of cyclophosphamide, TLI and CSA administered on alternate days. Wistar rats subjected to this treatment showed prolonged tolerance to mouse osteosarcoma and human adenocarcinoma xenografts. There was no alteration in the tumor doubling time or histological morphology of the xenografts in the adapted host as compared with those in the donor tumors. The tumor growth curve showed a pattern of initial growth, a period of stagnation, followed by a steady but slower growth phase. The significance of the results and the advantages of the rat model described in this paper for human tumor xenotransplantation are discussed.

Laser nephelometry was used to measure the lectin-mediated agglutination of cultured human tumor cells (of osteosarcoma and adenocarcinoma of the lungs). The rates of a decrease in the value of light dispersion (V delta VLD) and the length of time (to) necessary for complete cell sedimentation (at V delta VLD-O) can be used as quantitative indices of cell agglutination. V delta VLD and to were found to correlate with the concentrations of tumor cells and lectins (PHA and Con A).

The lectin (PHA and Con A) induced agglutination of a human tumour cell line (lung adenocarcinoma and osteosarcoma) was estimated by the spectrophotometric method. A decrease in the optic density for 1 min (delta D) of cell suspension and the time (t0) necessary for a complete sedimentation of cells (at delta D = 0) were used as quantitative indicators of agglutination. An increase in the concentration of tumour cells and lectins resulted in an increase of delta D and a decrease of t0. The results of spectrophotometry were correlated with the microscopic study data for tumour cells agglutination.

On in vitro passaging of the rat mammary adenocarcinoma R3230AC cell line, phenotypic changes occurred that were expressed in vivo. The histology of this mammary adenocarcinoma changed to a fibrosarcoma and then to an osteosarcoma. The overall population of early passaged cells consisted of a mixture of predominantly epithelial-like cells and few fibroblast-like cells; however, later passaged cells consisted more of the latter type. Along with the changes in histology, the tumor line also became highly metastatic in animals after in vitro passaging. The etiology of these phenotypic changes was not determined. Cytogenetic studies revealed chromosome changes in cells of later passages. Cells of 90 or greater passages that produced bone tumors were found to have fewer chromosomes and a metacentric marker isochromosome. In this report a correlation is made between the aberrant change in histology, increased metastatic ability and the presence of a marker chromosome of the R3230AC tumor.

"Crohn's Carcinoma" of a 34-year-old patient is presented in this paper. The fine structure of the tumour is discussed in detail, because there have been no previous reports of electron microscopic studies of this tumour. The adenocarcinoma cells were electron microscopically less differentiated than expected on the basis of light microscopic examinations. In the tumor cells, several round dense granules were observed. Similar granules were also described in the cells of intestinal type gastric and colon cancers, being considered a sign of pathologic mucus secretion. In a part of the tumour cells, intracisternal parallel tubular inclusions were seen like those described in malignant melanoma and osteosarcoma. In the dysplastic small-bowel mucosa adjacent to the tumour, the maturation disorder of the epithelial cells was similar to changes described in the precancerous states of the colon.

Uptake studies of the potential endoradiotherapeutic agent, 6-125I-iodo-2-methyl-1,4-naphthoquinol bis(diammonium phosphate) have been carried out in vitro on a wide range of normal and malignant human cells. In general, for a standardized dose of 0.1 microCi/ml, the uptake of the compound into normal cells was 0.0015-0.135 pCi/cell. Uptake into malignant cells was significantly higher than normal cells; uptakes of 0.89-11.3 pCi/cell were noted for melanoma, teratoma of testis, osteosarcoma and adenocarcinoma of colon and pancreas. Comparative uptake ratios for melanoma: Chang liver cells and testicular teratoma:normal testis were 29 23, respectively. Larger uptake ratios are usually observed with higher doses.

A simple combine treatment with immunosuppressive agents permitting the growth of human tumor xenografts in conventional mice is presented. It consists of two simultaneous applications of 90 mg of procarbazine (PCH) per kg and 30 mg of cyclophosphamide (CY) per kg, alternating with two doses of 0.15 ml/10 g of antilymphocyte serum (ALS) for 4 days before grafting. No postgraft treatment is used. With a take rate of 100%, at 60 days after subcutaneous transplantation into male C3H mice, fragments of a human colon adenocarcinoma had grown (on the average) into cherry-sized tumors. Two osteosarcomas, an Ewing sarcoma, and a bronchogenic carcinoma were also studied and grew similarly. Tumors could be established in the three tested mouse strains but grew better in male animals. Synergy of PCH and CY and ALS depends upon the alternating sequence of both drugs and ALS. The histology of the colon carcinoma and the Ewing sarcoma was unchanged as compared to the tumors growing in nude mice. In contrast, the osteosarcoma developed in a more differentiated fashion in the immunosuppressed mice. The presented model may serve as a screening system for anticancer drugs.

We report the third case of carcinosarcoma of the prostate. The epithelial element consisted of a highly malignant adenocarcinoma. The mesenchymal component was composed of malignant osteosarcoma and chondrosarcoma.
