Mouse neuroblastoma cells derived from cholinergic clone NS 20 were synchronized by isoleucine plus glutamine starvation. Basal adenylate cyclase activity increased linearly during the different phases of the cell cycle. Pharmacological data are presented indicating that adenosine, dopamine and prostaglandin E1 control through distinct receptors the same adenylate cyclase activity. The demonstration that basal enzyme activity and its responsiveness to the three agonists tested followed different evolution patterns during the cell cycle suggests that enzyme activity (or content) and activity (or number) of enzyme coupled receptors can be independently modulated.

We present a quantitative gas-chromatographic method for determining urinary 3-methoxy-4-hydroxymandelic acid (vanillylmandelic acid) and 3-methoxy-4-hydroxyphenylacetic acid (homovanillic acid). In this rapid technique an internal standard is used and the procedure involves ether extraction and silyl ether formation. Analytical recovery of vanillylmandelic acid averages 87.5% (CV, 0.95%), of homovanillic acid 102.3% (CV, 9.95%). Our data on 34 samples from 13 neuroblastoma patients show that homovanillic acid is more consistently elevated than is vanillylmandelic acid.

Eleven neonates with bladder obstruction, causing retention of urine, and a large, palpable lower abdominal mass were treated at the Yale-New Haven Hospital during a five year period. Intrinsic and extrinsic lesions, including urethral atresia, posterior urethral valves, ectopic ureteroceles, hydrocolpos and a neuroblastoma of the pelvis, were diagnosed by cystography, vaginography, pyelography and barium enema examination or a combination of these reoentgenographic methods. The multiplicity of underlying lesions make the obstructed bladder syndrome in the neonate a unique condition.

Retroperitoneal teratoma is the third most common primary retroperitoneal neoplasm of childhood ranking behind neuroblastoma and nephroblastoma. Ten per cent of these tumors are malignant. Two cases of benign retroperitoneal teratoma are reported, and the first description of the angiographic appearance of such a tumor is presented.

Glycopeptides suggesting a complex oligosaccharide composition are present on the surface of cells from human neuroblastoma tumors and several cell lines derived from the tumors. The glycopeptides, labeled with radioactive L-fucose, were removed from the cell surface with trypsin, digested with Pronase, and examined by chromatography on Sephadex G-50. Human skin fibroblasts, brain cells, and a fibroblast line derived from neuroblastoma tumor tissue show less complex glycopeptides. Although some differences exist between the cell lines and the primary tumor cells, the similarities between these human tumors and animal tumors examined previously are striking.

High-speed liquid chromatographic determinations of vanilmandelic acid and homovanillic acid in human urine have been made. The effects of taking refreshments on the assay were examined. HVA conjugates in urine also were hydrolyzed and determined. Urinary excretion of vanilmandelic acid, free and conjugated homovanillic acid in normal subjects and patients with neuroblastoma, phaeochromosytoma and Sipple's syndrome were determined and discussed. Urinary excretion values (mean +/- 1 S.D.) of vanilmandelic acid and homovanillic acid in 14 normal subjects were 2.0 +/- 0.7 mg/day and 4.0 +/- 1.0 mg/day, respectively.

A series of 35 primary cerebral neuroblastoma is reported. These rare tumours occur most often in children in the first half of the first decade. Grossly the tumors are often massive, discrete, lobular, firm and cystic. Histologically three variants, largely determined by the extent and distribution of the fibrous connective tissue stroma, are recognized: (1) a classical variant, which most resembles the peripheral neuroblastoma and is characterized by a high frequency of Homer Wright rosettes and a relatively high frequency of ganglionic differentiation; (2) a desmoplastic variant, which is characterized by an intense connective tissue stroma; and (3) a transitional variant, in which both the classical and the desmoplastic features may be present within the same case, either concurrently or consecutively. Both the desmoplastic and the transitional forms are less likely to exhibit differentiation to mature ganglion cells, but the importance of identifying the primitive cell elements as neuroblasts is emphasized. With rare exceptions, this can be established only by specific silver impregnations on frozen material. Occasionally the direction of growth may be largely leptomeningeal. Seven illustrative clinical histories with pathological correlations are described. The over-all clinical behaviour of these tumours is that of malignant neuroepithelial neoplasms, characterized by a high recurrence rate. Recurrence may, however, be a late development, in some cases occurring five or seven years after apparently successful surgical removal. The tumour shows shows a high incidence of metastatic spread, almost 40 per cent of the cases examined at autopsy having disseminated in the cerebrospinal pathways. Exceptionally, extraneural metastases may also develop. However, long post-operative survival occasionally occurs, and the subsequent clinical course is not always predictable in the individual case. The differential diagnosis is briefly discussed. The cellular nature of the tumour and its biological behaviour recall those of the cerebellar medulloblastoma. Post-operative radiation to the entire neuraxis should be considered for these neoplasms.

The reaction sequence for the biosynthesis of gangliosides by mouse neuroblastoma cells has been investigated by studying the pattern of incorporation of labeled precursors into sialoglycosphingolipids. Cultured NB41A cells incorporated N-[3H]acetylmannosamine into the sialic acid moiety of GM3 in less than 10 min. Labeled GM2 was not detected in cells incubated for less than 30 min, while measurable radioactivity did not appear in GM1 until after 60 to 90 min. Analogous experiments were carried out using [14C]galactose. No significant amount of labeled hexose was incorporated into asialo-GM2 during 60 min of culture. These studies are in accord with results of previous studies on glycosyltransferases of NB41A cells (Kemp, S. F., and Stoolmiller, A. C. (1976), J. Neurochem. 26, 723-732), and further support the concept that the pathway of synthesis of gangliosides proceeds via GM3 leads to GM2 leads to GM1.

Hematogenous spread of malignant tumors to the dental pulp is very rare. A case of adrenal neuroblastoma in a 71/2-year-old boy which metastasized to the mandible and dental pulp is described. Tumor cells were found within the pulpal blood vessels of a deciduous molar tooth.

Thirteen cell lines of tissue culture originated from human malignant neoplasms were transplanted into cheek pouches of adult golden hamsters treated with anti-hamster thymocyte serum. In eleven cell lines, transplantation tests with 3.5 X 10(6) to 6.8 X 10(6) cells resulted in neoplastic growth within 4 weeks. In other two cell lines including one from neuroblastoma and another from Burkitt's lymphoma, the inoculum size had to be raised to 11.8 X 10(6) and 12.2 X 10(6) cells to get tumor growth, respectively. The histological appearance of those tumors was consistent with that of the individual primary neoplasms despite of their long in vitro histories. Anaplasia grading of the tumors appeared not to be correlated with the frequency of take.
