The presence of 1.0mm-dibutyryl cyclic AMP (N(6),O(2')-dibutyryladenosine 3':5'-cyclic monophosphate) and 1.5mm-theophylline completely inhibits the growth of mouse neuroblastoma N2a cells by 24-36h. When compared with N2a cultures without inhibitors (controls), the proportion of cells in S phase, measured by radioautography with [(3)H]-thymidine, was decreased from 55 to 12%. In addition, the presence of the inhibitors decreased apparent [(3)H]fucose incorporation into glycoproteins by 50%, and removing the inhibitors resulted in a rapid recovery of both DNA synthesis and glycoprotein metabolism. Measurement of intracellular acid-soluble radioactive fucose revealed that decreased fucose uptake could account for the apparent change in incorporation. Removing dibutyryl cyclic AMP and theophylline from the medium resulted in a rapid uptake of radioactive fucose to within control values, which illustrated that the inhibitors decreased transport of the carbohydrate, although the cells remained viable. Treatment with dibutyryl cyclic AMP and theophylline also reversibly inhibited glycoprotein degradation. Plasma membranes isolated from growing cells and from growth-inhibited cells labelled with [(14)C]fucose and [(3)H]fucose respectively were co-electrophoresed on sodium dodecyl sulphate/polyacrylamide gels. These displayed no apparent differences in synthesis of specific membrane glycoproteins. Electrophoresis of plasma membranes isolated from cultures pulse-chased with [(14)C]fucose and [(3)H]fucose was used to discern turnover patterns of specific plasma-membrane glycoproteins. High-molecular-weight glycoproteins exhibited rapid rates of turnover in membranes from growing cells, but moderate turnover rates in growth-inhibited cells and cells reversed from growth inhibition. These data indicate that growth arrest of N2a cells results in alterations in the metabolic turnover of plasma-membrane glycoproteins.

The distribution of prostaglandin D synthetase activity was determined in various tissues of rat by using the supernatant fraction (10,000 x g, 20 min) of the homogenates. The highest activity was found in brain, spinal cord, and alimentary tract. The activity was uniquitously distributed in all parts of brain, and the highest specific activity was found in hypothalamus and thalamus. Homogenates of two neuroblastoma cell lines were found to produce prostaglandin D2, whereas a glioma cell line was almost inactive. Prostaglandin D2 is a potent and specific activator of the adenylate cyclase system of cultured neuroblastoma cells, suggesting the possibility that it may act as a neuromodulator in the central nervous system.

Phosphorus nuclear magnetic resonance is used to study changes in the levels of the major phosphate-containing intermediary metabolites concomitant with induced cellular differentiation in the N-18 and C-46 neuroblastoma clonal lines. The study reveals differences between the 31P-NMR profiles of the two clonal lines and also striking differences attendant to dibutyryl cAMP-mediated morphological differentiation in the N-18 clone. Phosphorus-31 NMR would appear to provide a new technique with which to study genetic differentiation.

Eleven cases of Pneumocystis carinii pneumonia were diagnosed during a 3 1/2-year period at a pediatric hospital where this infection had never been identified previously despite appropriate studies. The incidence of infection was 3.0, 7.4, and 4.2 cases per 1,000 patient months in children being treated for acute leukemia, neuroblastoma, and rhabdomyosarcoma, respectively. The outbreak coincided with increased intensity of chemotherapy for these malignancies. Ten of the patients had received four or more chemotherapeutic agents within three months of the onset of infection. Because no exogenous source of the epidemic was found, latent endogenous infection activated by immunosuppression was presumed to be the ultimate cause of the outbreak. Increased intensity of chemotherapy may result in P carinii outbreaks and may be an indication for anti-Pneumocystis prophylaxis with trimethoprim/sulfamethoxazole in patients at risk.

To help clarify the role, in cytoplasmic microtubule function, of the proteins of high molecular weight that coassemble with tubulin in vitro, a monospecific antibody against the proteins of high molecular weight was prepared from the serum of immunized rabbits by affinity chromatography. With indirect immunofluorescence we found that the protein in both cultured neuroblastoma cells and 3T3 cells in distributed in an extensive filamentous array that originates at sites near the nucleus and extends to the cell periphery or, in neuroblastoma cells, gathers into bundles which enter neurites. No filaments were seen with nonspecific antibodies from serum taken before immunization, and prior incubation of the specific antibody with purified protein of high molecular weight (but not tubulin) prevented filament visualization. The filamentous pattern in 3T3 cells was disrupted by colchicine. This evidence indicates that the proteins of high molecular weight are found in cells in association with cytoplasmic microtubules.

Ni-Cr castings produced in manufacturers' laboratories were consistently undersize, while precious metal castings were consistently oversize. Ni-Cr castings, produced in NBS laboratories using a modified investing technique, were routinely oversize.

Adrenal neuroblastomas are usually diagnosed by intravenous urography because of the displacement of the kidney or distortion of its contour. This report describes five children with left adrenal neuroblastoma where the mass was purely anterior to the kidney and produced displacement vectors similar to tumors in the body or tail of the pancreas.

Lateral diffusion of membrane lipids and proteins was determined in differentiating C1300 mouse neuroblastoma cells by fluorescence photo-bleaching recovery measurements. It is demonstrated that upon differentiation the lateral diffusion of membrane lipids and proteins is increased specifically in the extending neurites. This indicates the appearance of a topographical heterogeneity in the cell membrane, whereby more fluid domains become located in the membrane of the neurites.

We describe a method for the assay of urinary 4-hydroxy-3-methoxyphenylacetic acid (homovanillic acid), based on its separation on a microparticulate reversed-phase column and direct electrochemical detection. Patients with neuroblastoma, pheochromocytoma, and Parkinson's disease have increased amounts of this compound in their urine.

An intravenous urogram in a child with left adrenal neuroblastoma and bone metastases deomonstrated malrotation of the left kidney but no visible mass. A bone scan showed uptake of 99mTc by the tumor which suggested the diagnosis, confirmed by an abdominal echogram.
