Separation and determination of sample constituents by capillary isotachophoresis are entirely based on physical phenomena. The method has therefore been proposed as a universal reference method for ionic constituents. The present paper shows that even neutral species can be adequately determined after suitable preceding reactions. Urea was completely hydrolysed by urease (EC 3.5.1.5) to ammonia and bicarbonate, followed by direct measurement of the ammonium ion concentration by capillary isotachophoresis. Standard Reference Material No. 912a urea (National Bureau of Standards) was used as a primary standard. The analytical linear range of the method extends to 64 mmol urea per litre. The precision of the method was in the range of 1.05-2.64% (CV) and the analytical recovery of added urea was excellent (99.4%, SD 1.13%). Further proof of accuracy was obtained by analysing the NBS human reference serum (standard reference material 909). The mean result by the capillary isotachophoretic method, 9.52 +/- 0.085 mmol/l, agrees well with the reference value, 9.64 mmol/l. The results obtained by capillary isotachophoresis showed good agreement with those obtained by the coupled-enzyme method (r = 0.995).

Ferritin is an iron-containing protein which is a normal component of serum. The levels of ferritin are increased in the sera of some children with neuroblastoma, and this increase appears to be a potent indicator of prognosis. To determine whether synthesis of ferritin by the tumor cells contributes to these increased serum levels, we examined incorporation of radiolabeled leucine by CHP 126, a neuroblastoma derived cell line, into ferritin. Using sequential immunoprecipitation and gel electrophoresis of sonicates from cells maintained in medium containing iron in amounts standard for tissue culture, incorporation of label into ferritin was 0.04% of that into total protein synthesized over the same time period. Addition of up to 40 micrograms of iron as ferric ammonium citrate increased ferritin synthesis to a maximum of 0.16% without altering synthesis of total protein. The pattern of iron-induced enhancement in the neuroblastoma cells was similar to that which was seen using Chang liver cells, a cell line well known to be capable of ferritin synthesis. These results confirm that neuroblastoma cells can synthesize ferritin and that synthesis is regulated by exogenous iron.

The sensitivity and selectivity of high-resolution gas chromatography/mass spectrometry in the negative ion chemical ionization mode with methane as reagent gas was evaluated for the characterization of polar substituted polycyclic aromatic compounds (PAC). The fragmentation patterns were affected by the nature of the substituent for polar substituted nitro-PAC that showed detection limits of 50 pg at full-scan acquisition. This technique has been applied to the characterization of polar high-performance liquid chromatographic fractions of diesel exhaust particulate (NBS Standard Reference Material 1650) and enabled the identification of 20 PAC of different chemical classes. Among them, hydroxynitro-, dinitro- and nitrosubstituted secondary amines were identified for the first time in diesel exhaust particulate. In addition, 'filament-on' thermospray (TSP) liquid chromatography/mass spectrometry (LC/MS) with positive and negative ions have been used for the characterization of similar polar compounds such as 2-nitroquinoline, 1,8-naphthalic anhydride, naphthalene sulphonic acid and 1,2-hydroxynitronaphthalene. LC analyses were performed on a reversed-phase system using either acetonitrile-water or methanol-water with 0.1 M ammonium acetate and 1% acetic acid as eluent. With negative ion TSP LC/MS a four- to fivefold loss in sensitivity was observed for naphthalene sulphonic acid compared with nitrohydroxy-PAC, that showed a minimum detectable amount of 50 ng in the reconstructed ion chromatogram.

DNA Polymerase-alpha/primase complexes have been isolated from human neuroblastoma IMR-32, embryonic chicken brains (ECB) and rat prostate tumor PA-3 cells. In the presence of (NH4)2SO4 the major part (90%) of primase activity is released from the Pol-alpha/primase complex. A novel hydrophobic interaction column was used for purification of the primase from PA-3 cells. A nuclear protein factor (NPF-1) that stimulates DNA pol-alpha/primase activity has been purified from rat liver of various ages (3-6 months). The nuclear protein factor which only stimulates the primase activity is under investigation. The monoclonal antibodies (SJK 132-20 and 237-71) were used to detect DNA pol-alpha polypeptides from 11- to 19-day-old embryonic chicken brains.

A modification of a Dionex System 12 ion chromatograph is described which enables organic anions (acetate and formate), inorganic cations (ammonium, sodium and potassium) and inorganic anions (chloride, nitrate and sulphate) to be determined sequentially in one measuring procedure. The modified instrument consists of a programmable controller unit, a conductimetric meter, two conductimetric detectors of the Dionex System 12 ion chromatograph, the HPIC-AS4A and HPIC-CS3 modern separation units, AMMS-1 and CMMS-1 micro-membrane suppressor columns, a unique system of valves from Dionex and two dual pumps from Biotronik. The limits of detection are between about 1 and 3 micrograms/l for chloride, nitrate and sulphate and between about 2 and 10 micrograms/l for acetate, formate, ammonium, sodium and potassium. The reliability of the method was demonstrated by analysing two NBS simulated rain water Standard Reference Materials. Some examples are given of the application of the method to the sequential determination of the main precipitation components in typical samples from urban and rural regions of the F.R.G. The ion concentrations varied between about 0.02 and 300 mg/l.

A protein has been isolated from bovine brains by using a modification of the procedure used to purify glia maturation factor. The method consists of ammonium sulfate precipitation, chromatography with DEAE-Sephacel, Sephadex G-75, and hydroxylapatite columns, passage through a heparin-Sepharose column, and finally fractionation by reverse-phase HPLC with a C4 column. The isolated protein reacts strongly with the mouse monoclonal antibody G2-09 and has a molecular weight of approximately 17,000 and an isoelectric point of pH 4.9. The N terminus is blocked, but tryptic digestion releases 28 peptides, 8 of which have been sequenced. The total known residues add up to more than two-thirds of the entire 140-residue protein, estimated from amino acid composition, and show no sequence homology with any known protein. Reversible thermal renaturation greatly enhances its biological activity. The purified protein stimulates differentiation of normal neurons as well as glial cells. It inhibits the proliferation of the N-18 neuroblastoma line and the C6 glioma line while promoting their phenotypic expression. We designate this protein glia maturation factor beta.

Murine neuroblastoma cells (clone N1E-115) possess both high- and low-affinity muscarinic receptors. The low-affinity muscarinic receptor, when stimulated, initiates the formation of cyclic GMP by activating the enzyme guanylate cyclase; whereas stimulation of the high-affinity receptor inhibits prostaglanding E1-mediated cyclic AMP formation by inhibiting the enzyme adenylate cyclase. We have reported that lithium ion (Li+) inhibits cyclic GMP formation mediated by the muscarinic receptor agonist, carbachol, in a concentration-dependent manner and that neither ammonium nor sodium ions have such an effect. We extended this study to show that Li+ was an apparently noncompetitive inhibitor of the low-affinity muscarinic receptor with an IC50(+/- SEM) = 13.6 +/- 0.8 mM. In addition, Li+ with a similar IC50 inhibited the cyclic GMP response in intact cells to sodium azide, which is thought to stimulate guanylate cyclase directly. Moreover, though Li+ was found to have a slight inhibitory effect on prostaglandin E1-stimulated cyclic AMP formation (15% inhibition at 10 mM), it had no effect on the function of the high-affinity muscarinic receptor in intact murine neuroblastoma cells.

Rubrophilin, a unique brain specific polypeptide, was purified to apparent homogeneity from microsomal fractions of bovine brains. The peptide stains pink with Coomassie Brilliant Blue R-250 (C.I. No. 42660) under specific conditions, has an apparent Mr of 53,000, and is acidic with an apparent pI of 4.9. The purification involves initial solubilization of delipidated microsomes in sodium dodecyl sulfate, followed by ammonium sulfate fractionation, reversed ammonium sulfate gradient elution from diatomaceous earth, gel filtration on polyacrylamide (Biogel P-200), gradient elution chromatography from hydroxylapatite, and reverse-phase chromatography from phenyl-Sepharose. A yield of about 5 mg of rubrophilin was obtained from 9 g of microsomal proteins. Amino acid analysis shows that rubrophilin contains only nine amino acids with residues/mol as follows: alanine (102), glutamic acid (97), lysine (65), proline (55), aspartic acid (48), glycine (44), serine (37), threonine (35), and valine (10). Cysteine, methionine, tryptophan, tyrosine, isoleucine, phenylalanine, histidine, and arginine could not be detected. Relative rubrophilin content of vertebrate brains was as follows: mammals greater than birds greater than reptiles greater than fishes. It is present in mouse retina and human neuroblastoma cell cultures but could not be detected in octopus optic lobe or in cultured C-6 rat glioma cells.

Four monoclonal antibodies were obtained to rat brain choline acetyltransferase (CAT). The enzyme was purified 95,000-fold from rat brain by precipitation with acetic acid at pH 4.5, fractionation with 40 to 60% (NH4)2SO4, CM-Sephadex chromatography, and affinity column chromatography on agarose-hexane-coenzyme A. The enzyme preparation was applied to the affinity column in the presence of 10 mM acetylcholine to increase the affinity of CAT for coenzyme A; the enzyme then was eluted with 10 mM acetyl coenzyme A. Fusion of P3X63 Ag8 myeloma cells with spleen cells isolated from a BALB/c mouse that had been immunized with affinity-purified CAT with a specific activity of 29.4 mumol of ACh synthesized/min/mg of protein resulted in the isolation of four hybridomas synthesizing antibodies to CAT that inhibit the activity of the enzyme. Anti-CAT 1 or 2 inhibits CAT activity 100%. At the highest antibody concentration tested, anti-CAT 3 inhibited acetylcholine synthesis 80%. Hybridoma antibody-dependent inhibition of CAT activity was reversed by dissociation of immune complexes via dilution, demonstrating that antibody binding does not irreversibly alter the structure of the enzyme. When bound to [rabbit anti-mouse IgG . protein A Staphylococcus aureus] complexes, anti-CAT 1, 3, and 4 each were effective reagents for the precipitation of CAT activity from solution. Thirty-one to 53% of the precipitated enzyme was recovered following the dissociation of immune complexes. Anti-CAT 1, 2, and 3 inhibit CAT from 18-day chick embryo brain, NS20-Y mouse neuroblastoma cells, and rat brain.
