This is a clinicopathologic study of 6 cases of alveolar soft-part sarcoma. The patients age ranges from 8 to 47 years. All patients are women. In 5 cases the localization of the primary tumor is the lower extremities. Histologically the tumors have a distinctive organoid pattern outlined by thin-walled capillaries and are composed of nests of large polyhedral cells with abundant finely granular, acidophilic cytoplasm. Two tumors, among the 4 tested cases, have cytoplasmic

positive diastase-resistant crystalline structures. Electron microscopic study of one
tumor
shows intracytoplasmic glycogen, small membrane-bound electron-dense granules, Golgi lamellae and crystalline structures. Immunoperoxidase study performed in one case reveals a positivity with antikeratin and anti-enolase (NSE) antibodies. The prognosis of this
tumor
is poor. The main metastasis are in the lung and in the brain. Often the patients develop metastasis before detection of the primary tumor. The histogenesis of alveolar soft part sarcoma and the identity of the characteristic crystalloids remain open for discussion.

The correlation between increased serum alpha-fetoprotein (AFP) and a human vitelline component of 5 testicular embryonal carcinomas (1 was one histological type, 4 were more than one histological type) with no histological features specific for yolk sac
tumor
(YST) or endodermal sinus
tumor
(EST) was presented. In all 5 cases, distinct cells simulating the human yolk sac endodermal cell (HYSEC) with eosinophilic-granular or clear-vacuolated cytoplasm were found. These cells contained

positive, diastase resistant eosinophilic hyaline globules (EHG) which were positive for AFP. In the embryoid body, the same cells with positive AFP as the HYSEC were also seen. Electron microscopic investigation of these cells revealed basement membrane-like materials which were observed in organoid structure of YST. Positive AFP in these cells showed no binding to Concanavalin A (Con A), which was similar to AFP in YST. It was suggested that testicular embryonal carcinoma with higher serum AFP showing no histological features specific for YST had a vitelline component simulating the HYSEC and produced an increased serum AFP. Immunohistochemical staining for AFP of embryonal carcinoma with higher serum AFP is useful for detecting a human vitelline component and is important to guess the survival for the patient.

is reported. A 31-year-old male complained of severe abdominal pain and had a laparotomy following a diagnosis of acute panperitonitis. During the operation, a large retroperitoneal

attached to the abdominal aorta was found, accompanied with remarkable hemorrhage and necrosis. Histologically, the

showed immature large cells with a solid pattern, focally forming reticular, tubular and glomeruloid structures.

positive hyaline bodies, multinuclear giant cells and mitoses were noted in the
tumor
cells. Immunohistochemically, the

cells in the reticular pattern were positive for alpha-fetoprotein. No abnormality was found in the testes on palpation and ultrasound sonographic examination.

In order to clarify the biological characteristics of rat mammary tumors induced by 7,12-dimethylbenz-[a]-anthracene (DMBA), histochemical and immunohistochemical studies were performed. Two types of luminal spaces were observed within the
tumor
. In one type, the lumen was surrounded by eosinophilic columnar cells which were strongly reactive for soybean agglutinin (SBA) but weakly stained with keratin antibodies. In the luminal spaces, substances positive for
PAS
, dialyzed iron ferrocyanide or alcian blue and resistant to mucopolysaccharidase were occasionally observed. Ultrastructurally, the luminal surface was characterized by the presence of microvilli and tight junctions. In the other type, the lumen was often found in highly cellular foci and surrounded by pale, polygonal or elongated cells which were weakly stained with keratin antibodies but not SBA. The luminal spaces presented a peculiar structure filled mainly with mucoid substances sensitive to hyaluronidase, chondroitinase ABC and heparitinase, and the inner surface of the spaces was surrounded by basement membrane components: laminin, fibronectin and type IV collagen. The results of the present study therefore showed that DMBA-induced mammary tumor consists, partly, of a structure resembling human adenoid cystic carcinoma.

The effect of differentiation induction by a
tumor
-promoting phorbol diester, 12-O-tetradecanoylphorbol-13-acetate (TPA) on a clonal human megakaryoblastic cell line, MEG-O1s, was investigated, and a prominent response was demonstrated. The cells became weakly adherent, developed conspicuous cytoplasmic blebs, and displayed mature megakaryocytic characteristics by light microscopy such as the development of azurophilic cytoplasmic granules and a mosaic pattern of oxyphilic patches, multiplication of nuclei, and enhancement of the
PAS
reaction and alpha-naphthyl acetate esterase staining. Ultrastructural studies demonstrated the development of prominent cytoplasmic blebs, budding of blebs, and multiplication of nuclei. Numerous granules with central nucleoids that are similar to alpha granules had developed as well as granules with high electron density and clearly demarcated zones. Surface marker analysis revealed a moderate increase in IgG Fc receptor levels and a profound decrease in C3 receptor sites. By an immunofluorescent technique using monoclonal and polyclonal antibodies, a dramatic change in the expression of several megakaryocyte-platelet-specific proteins was demonstrated. All the proteins that had been expressed before induction such as platelet glycoprotein (GP) IIb/IIIa, fibrinogen, von Willebrand factor, factor XIIIA, beta-thromboglobulin (beta-TG), and HLA class 1 antigen were profoundly enhanced after induction by TPA. Induction by TPA led to the expression of fibronectin and factor V, which were not detected on nontreated cells. An ultrastructural immunoperoxidase study demonstrated platelet GPIb and GPIIb/IIIa in both plasma membranes and protein synthesis areas such as perinuclear cisternae and endoplasmic reticulum after TPA induction. beta-TG was also observed in some cytoplasmic granules of TPA-treated cells. TPA remarkably increased the secretion of beta-TG into the culture medium of MEG-01s. Ploidy was also increased from 2C to 4C to 4C to 8C. Similar maturation of MEG-01s was induced by other phorbol diester analogues such as phorbol-12,13-dibutyrate, but not by phorbol itself. These results indicate that phorbol diester, TPA, can bring about differentiation and maturation of a human megakaryoblastic cell line (MEG-01s) and that MEG-01s cells will provide a useful model for studying megakaryocytic differentiation and numerous megakaryocyte-platelet-specific proteins.

In a study on lung adenomas induced by the administration of a single intragastric dose of sodium thiocyanate, diethylnitrosamine, or its precursors to 15-day-old C57BLxC3H F1 mice the occurrence of crystalline inclusions in pulmonary parenchyma was noted. A correlation between crystalline deposits and number and type of adenomas seen between 46 and 110 weeks was observed. In 45/61 adenomas, eosinophilic cells containing inclusions were seen scattered in the pulmonary tissue, adjacent to

Prostate-specific antigen (PSA) and prostate acid phosphatase (PAP) were assayed using a radioimmunologic method in 306 patients from November 1986 through April 1987. Study patients included 10 women, 10 men under forty years of age, 25 patients with malignancies involving structures other than the prostate, and 280 patients with diseases of the prostate ie. benign hypertrophy of the prostate (BHP) (n = 170), or histologically-proved carcinoma of the prostate (CaP) (n = 110). Serum PSA levels were undetectable in women and following total prostatectomy; levels of 3 ng/ml were found in young men, with no circadian variations. Non-prostatic carcinomas had no influence on PSA levels. PSA levels in BHP patients were 6.9 +/- 8.4 ng/ml and correlated positively with the weight of the gland. In patients with carcinoma of the prostate, PSA levels were 24.4 +/- 19.3 ng/ml, correlated positively with
tumor
spread, and returned to normal following successful palliative hormone treatment, with new increases reflecting recurrences. PSA assays are of little value for screening for carcinoma of the prostate; however carcinoma of the prostate is found in 70% of patients with inconsiderable BHP and PSA levels above 15 ng/ml. PSA is mainly useful for monitoring patients with carcinoma of the prostate. No patient with BHP had marked elevations of PAP, whereas high PAP levels were found in 26% of patients with carcinoma of the prostate. Eighty-eight per cent of patients with carcinoma of the prostate had increased
