Immunochemically active fractions were obtained using Separon Hema-300-glc(R) from serum of
lactic dehydrogenase
virus (LDV) infected mice and from homogenates of human tumors. The mixture of proteins of tumorous origin from the cytosol giving a positive reaction in the leukocyte adherence inhibition (LAI) test was found in the same fractions showing maximum of absorbance at 340 nm in the spectrophotometer and a corresponding peak in the refractometer. Analogous peaks were not proved in material obtained from healthy controls, but they were found in some human placentas and fetal organs. The LDV fraction obtained from mouse serum served as a "control" antigen in LAI test for human
tumor
testing, and results corresponded with those obtained using cytosol specific for the
tumor
under study.

Treatment of NMRI mice i.p. with dehydrodipeptides [acetyldehydro-3-(2-thienyl)alanyltyrosine (SI); acetyldehydro-3-(2-furyl)alanyltyrosine (SII)] rendered macrophages cytolytic for several
tumor
cells in vitro. Normal peritoneal mouse macrophages from untreated mice not given injections of the peptides or from control mice given injections of phosphate-buffered saline were not cytotoxic. Moreover, supernatants from these in vivo-activated mouse peritoneal macrophages significantly increased the release of the cytoplasmic enzyme

from freshly added target cells, showing that these cells had been killed. The macrophage activation to lyse
tumor
cells was sharply dose dependent and appeared about 48 hr after injection of the peptides. Although dehydrodipeptide SI was active in vivo at concentrations as low as 500 microgram/mouse, the same substance lacked activity in vitro at all concentrations tested up to 800 microgram/ml. Dehydrodipeptides activate macrophages through a T-cell-independent process to lyse
tumor
target cells. Macrophages from athymic nude (nu/nu) mice were less cytotoxic, but they still were stimulated; and the culture supernatants could kill about 50% of the
tumor
cells used. There are indications for a relative specific structure-activity relationship of dehydrodipeptides for inducing cytotoxic macrophages.

From the original Dunning R-3327 rat prostatic adenocarcinoma, several distinct sublines have been obtained. These sublines include a well-differentiated, slow-growing, androgen-sensitive
tumor
(R-3327-H); a well-differentiated, slow-growing, androgen-insensitive
tumor
(R-3327-HI); and a fast-growing, androgen-insensitive, anaplastic
tumor
(R-3327-AT). These three sublines were compared in order to develop new model methods for the prediction of the androgen sensitivity and the degree of differentiation of prostatic adenocarcinomas. The R-3327-AT was very distinct in all parameters examined except the tissue protein electrophoretic patterns which contained a uniform pattern in all tumors. The significant differences between R-3327-H and -HI sublines were (a) the inability of testosterone to stimulate DNA synthesis in the R-3327-HI
tumor
and (b) the difference in the enzymatic profiles of these sublines. The specific activity of three enzymes (3 alpha-hydroxysteroid dehydrogenase, leucine aminopeptidase,
lactic dehydrogenase
) increased while the activity of another three enzymes (6 alpha,7 alpha-hydroxylase, 5 alpha-reductase, alkaline phosphatase) decreased in the sublines which are androgen insensitive and less differentiated. An arbitrary index was constructed, based upon these enzyme differences, which clearly discriminates the degree of androgen sensitivity and differentiation of these R-3327 rat prostatic adenocarcinomas.

Urinary
lactic dehydrogenase

LDH
) activity and isozyme pattern were investigated in 38 patients with bladder tumors of various stages and grades and in 10 healthy persons. Urinary
LDH
activity was markedly elevated and directly correlated with histologic stage and grade of tumors. The urinary
LDH
isozyme pattern differed significantly from that of normal urine even in early and well-differentiated
tumor
cases. LDH4 and LDH5 became more prominent as the degree of the histologic stage and grade of the
tumor
increased. In the patients with tumors more advanced than stage T2, LDH4 and LDH5 were greater than LDH1 plus LDH2, the reverse of what is found in normal urine. As analogous result was obtained in patients with tumors of grade II or higher. No appreciable difference was found in the
LDH
isozyme pattern of patients with single or multiple tumors although the total activity was greater with multiple tumors. A comparison between
LDH
isozyme pattern in urine and in
tumor
tissue was evaluated in 26 patients with a good correlation (r = 0.40, P less than 0.05). Comparison of the results with urine
LDH
and urine cytology was attempted for each stage and grade. In 5 of 16 cases having negative urine cytology, a positive isozyme index was present and 10 of 13 cases having positive urine cytology had a positive isozyme index. These data suggest that evaluation of urinary
LDH
combined with urinary cytology might be of value in the diagnosis of bladder tumors and for following patients who have had bladder tumors.

) activity of the aqueous humour has been estimated in both eyes of 7 patients having uniocular retinoblastoma. In 1 patient the aqueous humour
LDH
activity in the healthy eye was above normal, but there was no clinical evidence of malignancy.
Tumour
tissue was detected in this eye 9 months later, and the aqueous humour showed a rise in
LDH
activity. A high
LDH
activity persisted even after irradiation, though no tumour tissue was visible ophthalmoscopically. It is suggested that the estimation of the
LDH
activity in the aqueous humour of the healthy eye in cases of uniocular retinoblastoma might be of value in the early detection of a bilateral retinoblastoma.

isozyme patterns were measured in homogenates of 12 autochthonous ethylnitrosourea-induced rat gliomas and 5 cloned rat astrocytoma cell lines maintained in culture and transplanted to brain or flank sites in syngeneic hosts. The total
LDH
values in the autochthonous gliomas did not differ appreciably from normal brain controls, but the proportions of the cathodal isozymes, LDH4 and LDH5, were increased to a degree similar to that reported by others in spontaneous human malignant astrocytic gliomas. The cloned astrocytoma lines, both in vitro and in transplants at intracerebral or subcutaneous sites, commonly demonstrated elevated total
LDH
values and, without exception, showed a preponderance of isozymes, LDH4 and LDH5, that was distinctly more marked than in autochthonous tumors. Especially in cultured and transplanted rat gliomas, these findings suggest that astrocytic
tumor
cells maintain energy supplies by utilizing anaerobic glycolysis in relatively hypoxic environments. These data further underscore the need to develop laboratory brain tumor models for use in therapy trials that not only retain the convenience and predictability of transplantable gliomas but also approximate closely the metabolic properties of human spontaneous gliomas.

In most of the cases studied, histo- and cytoenzymochemical values of metabolism in cancer of the mammary gland correlate with the histological type, differentiation level and the degree of the

malignancy. As the histological degree of malignancy increases and the level of differentiation of the mammary cancer decreases, the activity of glycolysis and pentous shunt enzymes,
lactate dehydrogenase
and glucose-6-phosphate dehydrogenase, increases and that of the Krebs cycle enzymes, succinate and malate dehydrogenase, as well as the enzyme of glycerophosphate shuttle mechanism and oxidation of glycerol-L-glycerophosphate dehydrogenase decreases simultaneously. These data suggest that histo- and cytochemical methods of the enzyme activity determinations may be used for the specification of the prognosis and choosing of the auxiliary methods for the therapy of the mammary gland cancer based both on morphology and metabolism of the tumour.

The distribution of
lactate dehydrogenase

LDH
) isoenzymes in healthy blood donors and in patients suffering histological identified
tumor
(neoplasms) laryngis was investigated. The values of 110 healthy persons (controls) proved to be comparable with the average data described in literature. The
LDH
distribution pattern of 90 tumour affected persons showed significant differences comparing with controls. The alteration appears as absolute increase in
LDH
-1 (H4) isoenzyme parallel with progressive decrease in the hybrid enzyme rations containing M subunits (
LDH
-2,
LDH
-3,
LDH
-4), and the
LDH
-5 consisting of four M sununits was undetectable. The conclusions drawn from our investigations seem to be in agreement with Warburg's conception, supposing that in malignant tumours the aerob glycolysis increases. The determination of
LDH
isoenzyme distributions for diagnostic purposes in clinical practice is recommended.

lactate dehydrogenase
were studied by disc-electrophoresis in polyacrylamide gel, and in the clinic--in 1% agar gel. Oncovirus A12 invasion of the culture of rat embryo fibroblasts (REF) was found to result in the increased percentage of the cathode fractions activity (LDG-4 and LD-5) and in the disappearance of LDG-1 yet during the first day of the experiment prior to hypoxia and enhanced proliferation, i. e. it is most likely to be primary. In the homogenates of cancerous
tumor
and large intestine polyps of man also a reliable increase of the cathode and a decrease or disappearance of the anode fractions accur. A correlation of the experimental and clinical data allowed a suggestion to be made that LDG isoenzymes changes are genetically conditioned and play an important role in the process of oncogenesis, providing conditions for the increased intensity of glycolysis and proliferation.

A 58-year-old white male with a history of bronchogenic carcinoma presented with a total retinal detachment overlying a choroidal metastasis. His main
tumor
burden had been extirpated by pneumonectomy followed by radiation therapy four months prior to admission. With the development of a painful, glaucomatous eye, unresponsive to conventional therapy, enucleation was performed. Histologic examination of the enucleated globe revealed a metastatic
tumor
to the choroid, consistent with primary bronchogenic carcinoma. Aqueous humor and plasma examination revealed elevated ratios (Aqueous humor:Plasma) of
lactate dehydrogenase
(
LDH
) and phosphoglucose isomerase (PGI). Furthermore, subretinal fluid examination demonstrated concentrations of
LDH
and PGI higher than aqueous humor. While the level of carcinoembryonic antigen (CEA) in the plasma was normal (less than 2.5 ng/ml) following pneumonectomy, it was 121 ng/ml in the subretinal fluid. This would suggest that a choroidal lesion alone, in the absence of a clinically detectable primary tumor, is insufficient to elevate the plasma CEA.
