30 female Wistar rats weighing between 70 and 90 g were fed for 8 months with 0,188% N-[4-(5-nitro-2-furyl)-2-thiazolyl] Formamide (FANFT). After this period a papillary tumor of the urinary bladder was demonstrable in each animal. Histological examination always revealed a transitional cell carcinoma with prevailing medium tumor stages; no distant metastases were found. The grade of malignancy was classified as medium and high. No significant statistical relationship could be ascertained between tumor stage and grade or between grade and weight, but a correlation was established between tumor stage and weight. After 12 months, 36/60 kidneys were found to be normal, whereas in 20/60 dysplasias and in 2/60 transitional cell carcinomas of the renal pelvis were observed; secondary hydronephrosis due to bladder tumors occured 4 times. Four sublines of a murine N-[4-(5-nitro-2-furyl)-2-thiazolyl]-foramide (FANFT)-induced transitional cell carcinoma (MBT-2) possessing spontaneous metastatic ability were isolated via in vivo/in vitro serial selection of metastatic lung lesions. Subcutaneous inoculation of the parent cell line (MBT-2) produced primary tumors when injected into C3H mice. These primary tumors rarely metastasize. A subline designated L3F1 was established from 1 MBT-2 pulmonary metastatic tumor. Further in vivo/in vitro selections established three additional sublines designated L3F2, L3F3 and L3F4. Serial selection resulted in MBT-2 sublines of greater metastatic potential in terms of both incidence of metastasis and the number of metastatic tumors per lung. The parent line differed from the four sublines in metastatic potential, in vitro cell morphology, and in vitro growth parameters. The L3F2 subline was examined for the time of onset of metastasis by removal of the primary tumor. Metastasis of the subcutaneously transplanted tumor occurred between 14 and 21 days after injection of the L3F2 subline. The L3F2 primary tumors and lung metastases were morphologically characterized by light and electron microscopy.

The prophylactic effect of maltose tetrapalmitate (MTP), a newly developed non specific immunoadjuvant in preventing or delaying bladder cancer induction by N-[4(5-nitro-2-furyl)-2-thiazolyl] formamide (FANFT) or in reducing the growth rate of the induced tumor was compared to other well-known immunoadjuvants (BCG, C. parvum, levamisole and pyran copolymer). The evaluation of the prophylactic role of immunoadjuvants demonstrated that MTP, levamisole and C. parvum were the most effective in prolonging animal survival. MTP was found superior to either of them in reducing the tumor size. The development of lung metastasis was lower in the group receiving MTP or C. parvum. Next, MTP was studied for its therapeutic effect against primary FANFT induced tumor. The subcutaneous (s.c.) and oral routes of MTP administration and their combination with intravesical route were tried. Combinations of intravesical with s.c. or oral MTP were most effective in reducing tumor size, obtaining lower metastases along with greater mononuclear lymphocyte infiltration in the primary tumor.
