Lincomycin-spectinomycin, while effective in eliminating Mycoplasma gallisepticum from hatching eggs when administered via the yolk sac, had a marked deleterious effect on hatch-ability when given by this route. On the other hand when lincomycin-spectinomycin was injected into the air cell there was a reduction in the level of experimental infection with an acceptable effect on hatchability.

An improved medium for the isolation of Mycoplasma species from contaminated cell cultures is reported. A modification of the mycoplasma detection method using the uridine/uracil uptake ratio method is described. Results obtained using this method with mycoplasma contaminated cell cultures and with contaminated cell cultures treated with the antibiotic Lincomycin are presented.

Eleven mycoplasma strains were isolated from the semen of 24 stallions. Eight of these strains were identified as Mycoplasma equigenitalium. Three strains which hydrolized arginine could not be identified. The growth inhibition test with immune sera against M. arginini and M. equirhinis was negative. Antibiotic sensitivity test showed that all strains were sensitive to four antibiotic of tetracycline group (oxytetracyclin, minocycline, transcycline and vibramycin). Lincomycin and gentamycin appeared to be the most active against all the strains. Comparative analysis of routine semen examination did not reveal any difference between ejaculates infected with mycoplasma and free of these organisms. However, the levels of certain biochemical components of the semen plasma (glycerylphosphorylcholine, ergothioneine, fructose and of the semen plasma (glycerylphosphorylcholine, ergothioneine, fructose and total protein) from mycoplasma-positive ejaculates were significantly lower than in the semen plasma from mycoplasma free ejaculates.

The susceptibility of 11 T-strains, 12 strains of Mycoplasma hominis, and a single strain of M. fermentans to 15 antimicrobial agents was determined by study of inhibition of metabolic activity in a broth dilution system. All three species were inhibited by tetracycline, chloramphenicol, streptomycin, gentamicin, and kanamycin, and were relatively resistant to cephalothin, cephaloridine, polymyxin, vancomycin, and ampicillin. Three antimicrobial agents had significant differential effects on these species. Erythromycin was more active against T-strains than against M. hominis or M. fermentans. Lincomycin, clindamycin, and nitrofurantoin had greater activity against M. hominis and M. fermentans than against T-strains. The activity of the drugs tested was generally uniform over a wide range of inocula. The effect of pH and the difference between minimal inhibiting and minimal mycoplasmacidal concentrations of the drugs tested were consistent with expectations based on the effects of these drugs on bacteria.

Mycoplasma fermentans strains reputedly from human infections or tissue culture cells were much more susceptible to azithromycin than to clarithromycin or erythromycin. Lincomycin, clindamycin and several tetracyclines also exhibited good mycoplasmastatic activity but mycoplasmacidal concentrations were substantially greater than the MICs. Ciprofloxacin was the most active of three fluoroquinolones tested and was mycoplasmacidal at concentrations close to the MIC. Tiamulin and mupirocin were also very active. Synergy with specific M. fermentans antiserum plus guinea-pig complement was not observed with any class of antibiotic although the number of viable mycoplasmas was markedly reduced by the combined immunological components. Marked differences in susceptibility to various aminoglycosides were observed. Human strains isolated in cell-free media up to 1967 were aminoglycoside susceptible (MIC range 0.5-25 mg/L) but recent human isolates and strains isolated from tissue culture cells often showed either single or multiple aminoglycoside resistance (MIC > 500 mg/L). Two aminoglycoside-susceptible strains developed resistance to streptomycin or neomycin (> 500 mg/L) within five passages in broth containing streptomycin or neomycin, respectively. Resistance to tobramycin, kanamycin or gentamicin emerged after seven, eight and 14 cycles of exposure to the respective antibiotic. Streptomycin resistance was associated with a five-fold increase in resistance to tobramycin. Neomycin-, kanamycin-, gentamicin- and tobramycin-resistant variants showed mutual cross-resistance but remained susceptible to streptomycin. Induced resistance persisted for at least 17 passages in aminoglycoside-free broth. The use of aminoglycosides in human medicine and the frequent inclusion of some of these drugs in tissue cell cultures to combat bacterial and mycoplasmal contamination might account for the aminoglycoside resistance of recent M. fermentans isolates.

Five groups of 20 commercial leghorn hens near peak production were challenged with Mycoplasma gallisepticum, and medicated with enrofloxacin in the drinking water at 7-11, 21-25, or 7-11 and 21-25 days postchallenge (PC), a combination of lincomycin/spectinomycin at 7-11 and 21-25 days PC, or left as untreated controls. Egg production records were maintained, and all eggs produced during the 63 days following challenge were incubated for 18 days and then cultured for M. gallisepticum. All groups experienced marked egg production drops beginning about 1 week PC; production returned to near normal levels after approximately 3 weeks. Under the conditions of this study water medication did not prevent egg production losses, probably because of the delay in the onset of medication, but medication may have resulted in a more rapid recovery of egg production. Enrofloxacin was highly effective in reducing the level of egg transmission of M. gallisepticum, although a small number of eggs (0-0.3%) in the treated groups were culture positive after medication was begun. Only 1 of 2463 live embryos was culture positive after the onset of treatment. In the unmedicated control group, 12.8% of all eggs were culture positive. Lincomycin/spectinomycin also reduced egg transmission (3.4% of eggs were culture positive after treatment was initiated), but not as effectively as enrofloxacin. Most of the egg transmission occurred before the medication could have had an effect. Both enrofloxacin and lincomycin/spectinomycin reduced the number of dead-in-shell embryos.
