A survey was carried out to characterise the bacterial and mycoplasma flora of a closed colony of marmosets, Callithrix jacchus. Bordetella bronchiseptica and Staphylococcus aureus were the only potential bacterial pathogens isolated from the colony; Candida albicans was also identified. S aureus and C albicans were the only organisms which were associated with macroscopic lesions or overt disease. Post mortem examination of the animals proved only reliable method of establishing the presence of some bacterial species in the colony, eg, B bronchiseptica and of obtaining a reliable estimate of the incidence of the various organisms within the colony. The marmosets were all infected with ureaplasmas in the upper respiratory tract. Mycoplasma salivarium was isolated from this site in several animals. There was no evidence to indicate that cross-infection had occurred between the animal technicians and the marmosets, except in one possible case of candidiasis.

New born guinea pigs of Hartley strain derived by hysterectomy were fed commercial pellets, cow's milk, egg yolk and vitamin mixture since 0 days of age, when they were kept at 31 +/- 1 degrees C. Out of 33 animals, 30 were reared for 40 days under aseptic state and they were transfered to a barrierred facility to establish an SPF colony free from Bordetella bronchiseptica, Streptococcus zooepidemicus (Animal C), Salmonella spp., Tyzzer's organisms, Mycoplasma spp., Reo 3 virus, Sendai virus, Eimeria spp., Chirodiscoides caviae and Gliriocola porcelli.

During a period from 1973 to 1978, 392 and 225 lots including 12,232 mouse and 8,044 rat individual sera, respectively, were examined for antibodies to murine hepatitis virus, Sendai virus, Bordetella bronchiseptica, Mycoplasma pulmonis, Tyzzer agents, Salmonella typhimurium and Corynebacterium kutscheri. Of mouse lots 94.5% and 39.3% from breeder and user colonies, respectively, were negative for all antibodies examined as well as 31.6% and 17.2% of rat breeder and user colonies, respectively. Among positive lots from mouse users, high positivity rates were seen with Senai virus (47.6%), M. pulmonis (19.0%), and murine hepatitis virus (JHM : 18.2%, MHV : 31.0%), while the rates were high in rat user lots with Sendai virus (24.4%), B. bronchiseptica (39.3%) M. pulmonis (12.5%), murine coronaviruses (JHM : 19.0%, MHV-2 : 28.0%) and tyzzer agents (MSK : 19.6%, RT : 17.9%). These pathogenes with high positivities should be monitored indispensably as a quality control of laboratory mice and rats.

Four 60-day-old pigs were experimentally infected with Bordetella bronchiseptica at a state in which the animals were clinicylly normal. A single pig only, killed on the 180th day, presented a slight deflection of septum nasi. Morphologically, there were severe dystrophic and necrotic changes in the mucosa of the conchae, the nasal cartilages as well as in the bones. The pathogen was reisolated from two of the pigs. Clinical and morphological investigations were carried out also with spontaneously affected animals, isolating Pasteurella multocida, Pseudomonas aeruginosa, and Mycoplasma organisms, but not Bordetella bronchiseptic. Morphologically, there were in 37 of the pigs severe dystrophic and necrotic lesions in the nasal mucosa, cartilages, and bones. Basophilic inclusion bodies were demonstrated in the epithelial cells of the tubulous glands. It is concluded that a combined method of prophylaxis and treatment would be most effective, and of the drugs borgal has proved likewise most promising.

The progeny of 9 SPF gilts fed a balanced ration (Table I) was used in an inoculation experiment with field strains of Bordetella bronchiseptica isolated in herds suffering atrophic rhinitis. Acute rhinitis was produced within a week after intranasal inoculation of B. bronchiseptica into 1 to 11-day-old piglets. Coughing occurred in some of the exposed pigs, but signs of pneumonia did not develop. A few pigs were killed at intervals of 1 to 3 weeks after exposure. These pigs all showed histological lesions in the turbinate and B. bronchiseptica was recovered from various parts of the respiratory tract. Pigs killed 3 weeks after inoculation showed advanced turbinate atrophy (Tables II and III). Among inoculated litter mates reared to slaughter weight, only one developed clinical signs (slight) of atrophic rhinitis, and a tendency towards an elimination of the B. bronchiseptica infection from the accessible part of the nasal cavity was noticed during the growth period. By examination of nasal swabs collected when the pigs were 10 to 13 weeks old, Mycoplasma flocculare was isolated as well from pigs inoculated with B. bronchiseptica as from the control pigs. The growth rate of the experimental pigs was high and no difference in feed consumption or feed conversion occurred between the exposed pigs and the control pigs. By post mortem examination of the snouts from the pigs slaughtered at approx. 85 kg live weight, severe atrophic rhinitis was not found. Approximately one third (32%) of the exposed pigs showed slight atrophic rhinitis lesions (Table IV). The results are discussed and it is concluded that the isolated B. bronchiseptica strains are pathogenic in young pigs and able to induce turbinate atrophy 2 to 3 weeks after inoculation. Turbinate atrophy induced in pigs a few weeks old, may apparently restore completely or almost completely during the growth period. Under the provided experimental conditions, infection with B. bronchiseptica did not result in the development of a lasting, growth-retarding atrophic rhinitis.

Eight hundred and fifty-four piglets which died or were euthanized due to pneumonia or rhinitis atrophicans, were investigated during the period of 1986-1990. Of the animals, 569 showed bronchopneumonia, 218 had pleuritis, pericarditis and peritonitis, 165 had rhinitis atrophicans, 58 pleuropneumonia, and 9 animals had fibrinous pneumonia. Pasteurella multocida, Haemophilus parasuis, Bordetella bronchiseptica, Actinobacillus pleuropneumoniae and Pasteurella haemolytica were isolated in 59.1%, 29.5%, 27.8%, 3.7%, and 2.3% cases of bronchopneumonia respectively. Samples from pigs with pleuritis or rhinitis atrophicans showed Pasteurella multocida in 63.8 and 68.5%, Bordetella bronchiseptica in 28.4 and 39.4%, streptococci in 28.9 and 3.9%, Haemophilus parasuis in 25.2% and 20.6%, Actinobacillus pleuropneumoniae in 5.1 and 5.5%, and Pasteurella haemolytica in 3.2 and 3.0%, respectively Actinobacillus pleuropneumoniae was found in 51 of 58 cases of pleuropneumonia and in 5 of 9 cases of fibrinous pneumonia; 55.6% and 44.4% respectively of those forms of pneumonia were positive for Pasteurella multocida. In the agar diffusion test, 36.8-82.6% of bacterial isolates showed resistance to streptomycin, 7.7-45.5% to sulfamethoxazole-trimethoprim, 5.7-44.6% to tetracycline, 0.2-32.8% to ampicillin, 0.0-16.3% to lincospectin, 2.0-81.2% to furazolidone, 0.4-4.5% to chloramphenicol, 1.3-78.1% to penicillin and 0-0.3% to enrofloxacin.

A novel method to reduce contamination of the bronchoscope during microbial sampling of the lower airways of foals was evaluated. Methylene blue (MB) was used as a nasopharyngeal dye marker to assess the relative contamination from the upper airways of bronchoalveolar lavage (BAL) specimens obtained by standard bronchoscopy (SB) and a "guarded" bronchoscopic method (GB). For GB, a clear sterile cellulose sheath was fitted over the bronchoscope in an effort to protect the endoscope tip and channel from contamination. Methylene blue was detected visually in seven of eight BAL samples from foals following SB, but in none of the samples recovered by GB (p less than 0.001). Significantly less MB was detected in BAL by spectrophotometry in the GB group as well (p less than 0.02). The GB was next employed to study the microbial flora in the lower airways of healthy weaned foals (n = 30). Bacteria were isolated from 29 of 30 (97%) BAL samples, and in moderate or large numbers from 26 of 30 (87%) of the foals. Potential pathogens, including Bordetella bronchiseptica, Streptococcus zooepidemicus, Staphylococcus aureus, Mycoplasma felis and Streptococcus pneumoniae, were cultured from the lower airways of foals. In conclusion, the bronchoscope and bronchoalveolar lavage specimens were readily contaminated by a dye marker placed in the nasopharynx of foals, and the degree of contamination was significantly reduced by sheathing the endoscope. This contamination during bronchoscopy may obscure the interpretation of isolates from BAL specimens from foals, which may possess a bacterial flora in the lower airways without cytological evidence of inflammation.

An enzyme-linked immunosorbent assay (ELISA) for detection of Bordetella avium infection in turkey poults was developed. One-week-old poults challenged intratracheally with 10(12) colony-forming units of B. avium had detectable titers (greater than or equal to 11), with an average of 13.6% positive samples when the birds were 6 to 11 weeks old. The method was sensitive enough to detect maternal antibodies to B. avium in poults up to 3 weeks of age. The same poults challenged at 1 week of age had 100% tracheal infection up to 3 weeks of age, which dropped to 0% by 6 weeks. The method resulted in no false-positive samples (titer = 0) from birds not infected with B. avium and tested weekly between 4 and 11 weeks of age. Antibodies in turkey flocks infected with Newcastle disease virus, hemorrhagic enteritis virus, and Mycoplasma meleagridis, and birds infected with Escherichia coli had no apparent cross-reactivity to the B. avium antigens used in the ELISA. The percentages of B. avium-positive serum samples collected from different turkey flocks did not significantly differ (P greater than 0.05) when samples were tested by the developed ELISA at different times, an indication of the reproducibility of the method.

A survey was conducted to characterize domestic and exotic bird populations, estimate seroprevalence to selected disease agents, and describe health management practices on 62 premises containing "backyard" flocks located within one mile of 22 commercial California meat-turkey flocks participating in National Animal Health Monitoring System (NAHMS). Chickens were present on 56 backyard premises and turkeys on seven. Antibodies were identified against Mycoplasma gallisepticum, M. synoviae, M. meleagridis, Salmonella pullorum, Newcastle disease virus, avian encephalomyelitis virus, Bordetella avium, hemorrhagic enteritis virus, infectious bronchitis virus, and infectious bursal disease virus in 367 blood samples from 32 backyard premises. Twenty-two owners of backyard premises said they restricted visitor contact with their birds, and two required visitors to wear rubber boots and use boot disinfectant. Owners of seven premises used biologics and/or pharmaceutics for disease prevention. One family member worked on a commercial turkey ranch, but no other contact between owners, relatives, or employees and commercial poultry was reported.
