The expression of tumor-associated antigens on the DBA/2 lymphoma L1210 and three L1210 sublines, each resistant to a different antileukemic agent [guanazole, methylglyoxal-bis(guanylhydrazone), and 4,4-diacetyldiphenylurea-bis(guanylhydrazone)] was investigated by the use of monoclonal hybridoma antibodies. Hybridomas were produced by the fusion of spleen cells from DBA/2 mice immunized with irradiated L1210 or L1210 subline cells and cells of a non-immunoglobulin-secreting BALB/c myeloma variant. Three clones producing antibodies reacting with L1210 or L1210 subline cells were used to study the antigenic expression of L1210 and L1210 subline cells. Monoclonal antibodies from anti-L1210 and anti-L1210 subline hybridomas exhibited a greater reactivity with L1210 subline cells than with L1210 cells in complement-dependent cytotoxicity, quantitative absorption, and membrane immunofluorescence experiments, thereby demonstrating a tumor-associated antigen shared by L1210 and the L1210 sublines and an increased expression of this antigen on subline cells. Cross-blocking tests of antibody binding demonstrated that monoclonal antibodies from anti-L1210 and anti-L1210 subline hybridomas recognized the same or very closely situated antigenic determinants on the tumor cell surface. Most syngeneic and allogeneic tumor cells used as controls failed to react with the anti-L1210 and anti-L1210 subline hybridoma antibodies. However, two syngeneic tumors, L5178Y and P388-D1, demonstrated a significant reaction with the monoclonal antibodies. In addition, several spontaneous mammary tumors from C3H/St and DBA/2Ha, both high-frequency mammary tumor strains, reacted in various degrees with anti-L1210 or anti-L1210 subline hybridoma antibodies in absorption tests and in immunofluorescence experiments. On the other hand, liver, kidney, and spleen from normal C3H/St mice, as well as mammary tumors from BALB/c and C3Hf, both low-frequency mammary tumor strains, did not demonstrate significant reactivity in similar experiments. Normal lactating mammary glands from high-frequency mammary tumor mouse strains reacted with the monoclonal antibodies, whereas lactating mammary glands from low-frequency mammary tumor mouse strains were negative by this method. Purified murine mammary tumor virus preparations reacted strongly with the monoclonal antibodies in solid-phase radioimmunoassays, whereas a purified murine leukemia virus preparation failed to do so in similar experiments. These results indicate that the tumor-associated antigen(s), differentially expressed on L1210 and L1210 subline cells, is related to an antigen which is associated with the murine mammary tumor virus.

Marrow transplantation is effective treatment for a number of haematological diseases in patients under the age of 50 who have an HLA-identical sibling donor. It is generally successful when used early in the treatment of aplastic anaemia. It is the only treatment that offers long-term disease-free survival for patients with acute leukaemia who have relapsed at least once, with 10-30 per cent apparent cures. Although still somewhat controversial, it appears also to be the treatment of choice for patients with acute non-lymphoblastic leukaemia in first chemotherapy induced remission and for those with chronic myelogenous leukaemia in the chronic phase since approximately 50-60 per cent of these patients are surviving after marrow transplantation in complete remission, apparently cured. Marrow grafting is the only effective treatment for many patients with inherited immunological-deficiency diseases and certain genetic storage diseases. It is being explored for the therapy of patients with lymphoma, Hodgkin's disease, multiple myeloma, small-cell lung cancer, testicular cancer, ovarian cancer and genetic disorders of haematopoiesis. Cures of congenital Fanconi anaemia, Blackfan-Diamond anaemia, osteopetrosis, and paroxysmal nocturnal haemoglobinuria have been achieved by marrow grafting. Genetic disorders associated with haemolytic anaemia and cyclic neutropenia have been cured by marrow grafting in animals. Target disorders for marrow transplantation in humans are thalassaemia major and sickle cell disease, and, indeed, a first successful transplant for treatment of thalassaemia major has recently been described (Thomas et al, 1982). Marrow transplantation has been limited by the fact that many patients do not have HLA-identical siblings and very few have monozygotic twins. The Seattle team has now explored the use of less well-matched family member donors in more than 80 patients with leukaemia. These donors share one HLA haplotype genetically with the patient and are phenotypically identical at two of the three major HLA loci on the other HLA haplotype (Clift et al, 1979). Overall, the post-transplant survival appears more a reflection of the type and stage of the leukaemia than of the marrow donor. Patients with leukaemia grafted in relapse have a projected survival of 20-30 per cent and those transplanted in remission of 50 per cent. A hybridoma producing monoclonal antibodies (McAb) NATF9.9 (F9) was obtained from fusion of murine myeloma X63 and splenocytes of AKR mice immunized with a single intravenous injection of 5 X 10(7) thymocytes of CBA mice. F9 McAb were cytotoxic for 80% thymocytes, 10% splenocytes, 20% lymph node cells, 85% cortical and 32% medullary thymocytes of CBA, C57BL/6, BALB/c, DBA/2 and SJL but not for the cells of C58 and AKR mice. F9 McAb reacted only with T cells and did not react with B cells and EL4 thymoma cells (Thy-1.2+, Lyt-1+2-3-). The proportion of F9+ cells accounts for about 40% among T lymphocytes of the lymph nodes and spleen as tested by flow-type cytometry. Lymph node cells treated with F9 McAb plus complement completely lost their reactivity with rat anti-Lyt-2 McAb and only partly (by 30%) with anti-Lyt-1 McAb. The reactivity pattern of F9 McAb attests to their specificity for Lyt-3.2 antigen.

A mouse monoclonal antibody (IgM) was obtained by cell hybridization between X63-Ag8.653 myeloma cells and spleen cells from a BALB mouse that was immunized with GRSL leukemia cells of the GR strain. This antibody identified a unique fetal antigen, which is expressed exclusively on embryonic thymocytes of all strains tested. Therefore, the antigen defined was named fetal thymus antigen-1, FT-1. The proportion of FT-1+ fetal thymocytes detected by immunofluorescence assay sharply decreases as gestation time increases, and finally they disappear from the thymus. On the other hand, Thy-1+ cells increase in inverse proportion. The immunofluorescence studies and absorption tests showed that FT-1 antigen is not detectable on brain, liver, kidney, or lymphoid tissue cells of adult mice. However, it is expressed on some leukemia cells of various mouse strains, which demonstrated that this is the first example of an oncofetal antigen of a mouse leukemia. The molecular weight of FT-1 antigen on leukemia cells was estimated to be 130,000 by means of biosynthetic labeling with [3H]galactose and [35S]methionine. The two-dimensional gel electrophoresis pattern of FT-1 antigen shows a family of glycoproteins with extensive charge heterogeneity. It was also shown that the FT-1 antigen molecule carries the receptor for DBA lectin.

Tumor-associated antigens of DBA/2 lymphoma L1210 and three immunologic drug-resistant L1210 sublines were studied by monoclonal antibodies produced by fusing non-Ig secreting BALB/c myeloma cells with spleen cell of DBA/2 mice immunized with L1210 subline cells that are syngeneic with respect of DBA/2 mice. After the cell fusion, two stable clones of antibody-producing hybridomas were selected for further analysis of the antibody specificity by complement-dependent cytotoxicity and quantitative absorption tests. These clones produced a cytotoxic antibody(s) of IgG2 class recognizing a TAA(s) that was expressed strongly on cells of the immunizing subline as well as on cells of their L1210 sublines, but weakly on the parental L1210 cells. Control normal syngeneic mouse tissues and tumors of syngeneic and allogeneic origins, including those that carry known murine leukemia virus-associated antigens, showed no significant reaction with the antibody, whereas two DBA/2 tumors (i.e., L5178Y and p388-D1) showed weak reactivity. Since all three L1210 sublines were more immunogenic than the parental L1210 cells in eliciting host's tumor graft rejection and contained more TAA than the parent line, the data suggest that the monoclonal antibody obtained may be defining the TAA that is involved in tumor rejection.

Monoclonal antibodies reactive with NIH/3T3 cell surface antigens were obtained from hybridomas of murine myeloma cells fused to spleen cells of rats immunized with NIH/3T3 cell plasma membranes. Four of the antibodies, of forty that have been studied, appeared to react with allospecific antigenic determinants: they bound to NIH/3T3 cells but not to BALB/ 3T3 cells. Each of these four antibodies immunoprecipitated a glycoprotein of about 80,000 daltons that migrated to an isoelectric point of about pH 5.0. Polypeptides of identical molecular weight and isoelectric points, and yielding the same proteolytic cleavage fragments, were present in BALB/3T3 cells, but were not antigenically reactive. The 80,000-dalton glycoprotein was a major constituent of the plasma membrane. It was a predominant lactoperoxidase iodinated component of intact NIH/3T3 cells, and saturation binding of 125I-labeled antibody indicated that there were about 10(6) antigenic sites/cell. Studies of the distribution of the immunoreactive glycoprotein among different strains of mice confirmed the polymorphic expression of the determinant: Spleen cells of BALB/c, DBA/1, DBA/2, and CBA mice did not bind anti-80,000-dalton glycoprotein monoclonal antibodies, whereas spleen cells of a large number of other strains of mice were positive for antibody-binding. The antigenic reactivity varied markedly among different cell lines and was greatest with the NIH/3T3 mouse embryo fibroblast, G8-1 Swiss Webster myoblast, and IC-21 SV40-transformed C57BL/6 mouse peritoneal macrophage. The properties of the 80,000-dalton glycoprotein characterized this molecule as a new cell surface differentiation alloantigen of murine mesenchymal cells.

The results presented in this communication show very clearly that the mice with IghCa haplotype can produce anti-TNP antibodies from which a fraction share the idiotypic determinants expressed on M460 myeloma protein. Nevertheless the gene(s) which encode(s) this specificity, is present in the repertoire of strains of mice which have not this haplotype. As in DBA/2 mice this silent clone(s) can be stimulated by an appropriate immunological manipulation and their progeny even can be frozen in hybridomas. The expression of 460 Id positive anti-TNP antibody forming cells is regulated by T cells in BALB/c mice. These T cells are specific for 460 Id and share the idiotypic determinants of anti-460 Id antibodies since they were not found in BALB/c mice with an Ab3 type of response.

A mouse of monoclonal cell line (L1) was produced by fusing the mouse myeloma P3X63/Ag8 with CD2F1 spleen cells immunized with a highly immunogenic subline of L1210 leukaemia (L1210/DTIC). A very few positive clones (1%) were isolated and one of these was chosen for detailed study. The monoclonal antibody L1 is an IgM immunoglobulin strongly reacting in a complement-dependent cytotoxicity assay against L1210/Cr leukaemia and its more or less immunogenic sublines. The specificity of the L1 antibody against L1210 leukaemia was studied by extensive screening with normal adult and foetal tissues, lymphoid tissues from several independent strains and a panel of the most common experimental tumours, to all of which it was unreactive. Attempts at immunotherapy were carried out in DBA/2 mice challenged with L1210 leukaemia and treated with L1 (ascites) and complement. Although the in vitro cytotoxic titre of ascites fluid from mice bearing hybridoma was very high (10(-7)), no therapeutic effect was obtained in vivo.

Two hybrid clones from a fusion of C57BL/6 anti-DBA/2 spleen cells and the myeloma line Sp2/0 secrete antibodies reactive with a product of the murine major histocompatibility complex (MHC). The two antibodies are provisionally designated S13.11 and S13.29. Both react in rabbit-complement-mediated cytotoxicity with spleen cells of H-2d, H-2f, H-2r, and H-2p strains. In addition, both antibodies hemagglutinate red blood cells from these strains. S13.11 is also cytotoxic for H-2a, H-2k, H-2u, and H-2v spleen cells but does not hemagglutinate red blood cells from mice bearing these haplotypes. With the exception of H-2v, this strain pattern mimics the public specificity H-28. Quantitative absorption of S13.11 shows that H-2d cells are twice as efficient as H-2k cells in their ability to remove the S13.11 antibody. S13.29 reacts weakly in cytotoxicity with H-2k spleen cells and does not react with cells from H-2u or H-2v. Blocking studies indicate that S13.11 and S13.29 react with the same or a closely related molecule on the cell surface.

Monoclonal antibodies (MAbs) were generated against recombinant human insulin-like growth factor 1 (IGF-1) by fusion of NS-1 myeloma cells with spleen cells from BALB/c X DBA mice immunised with recombinant IGF-1 and synthetic peptide sequences derived from the published amino acid sequence of IGF-1. MAbs were produced that recognised four distinct epitopes including two defined segments of the C and D domains. All MAbs were IgM mouse immunoglobulins. These results indicate the feasibility of producing MAbs to highly conserved proteins.
