The cytotoxicity including complement dependent cytotoxicity (CDC) and antibody-dependent cell-mediated cytotoxicity (ADCC) of two kinds of monoclonal antibodies (Mo-Ab), namely, MSN-1 which was obtained by means of an immunization procedure with intact SNG-II cells from human endometrial adenocarcinoma and HMST-1 obtained from hybridoma fused lymphocytes from lymph node with murine myeloma cells were studied by the 51Cr liberation test in vitro. 1) Against SNG-II, MSN-1 and HMST-1 showed significant CDC activity at 38.4 +/- 4.2% and 41.9 +/- 4.8% respectively, when a guinea pig complement was used. However, no significant CDC, action on SKG-IIIb was induced by MSN-1 or HMST-1. 2) Mo-Ab:MSN-1 and HMST-1 showed no significant liberation of 51Cr in relation to SKG-IIIb and SNG-II when using peripheral blood lymphocyte (PBL) separated by centrifugation gradient as the effector cells, and then the cytotoxicity of PBL alone against K-562 cell, SNG-II and SKG-IIIb was 40%, 9% and 2% respectively. This suggested that neither Mo-Ab had any ADCC activity and that SKG-IIIb and SNG-II cells were NK resistant cell-lines.

A new monoclonal antibody, 1C5, was produced by fusion of spleen cells obtained from mice immunized with CAC-1, a human cell line of cervical adenocarcinoma of the uterus, and NS-1 myeloma cell. The objectives of this study were to obtain moAb that can be used for routine histology and cytology, and to examine the histogenesis of cervical adenocarcinoma. 1. 1C5 reacted with 88% of cervical adenocarcinoma of the uterus, but did not react with cervical squamous cell carcinoma of the uterus and other squamous cell carcinoma. However, 1C5 reacted with some adenocarcinomas, such as endometrial carcinoma of the uterus and ovarial carcinoma. 2. The staining pattern by 1C5 was different, in cervical adenocarcinoma from that in endometrial carcinoma of the uterus, and also different in the endocervical type from that in the endometrioid type of cervical adenocarcinoma. Therefore, 1C5 is useful in distinguishing between two types of adenocarcinoma of the uterus. 3. 1C5 did not react with normal squamous cells or normal columnar cells of the uterine cervix, or with normal endometrial cells of the uterus. However, the columnar cells in a limited area of the squamocolumnar junction were strongly stained with 1C5. 4. 1C5 reacted with ethanol-fixed, and routine formalin-fixed and paraffin-embedded tissue. Thus, 1C5 may be used for clinical diagnosis. 5. 1C5 was found to be IgG1. 6. The molecular weight of the 1C5-defined antigen was 26,000 daltons, and the epitope of the 1C5-defined antigen was carbohydrate moiety. 7. Spleen cells from BALB/c mouse immunized with the human endometrial adenocarcinoma cell line (ISHIKAWA) were fused with mouse myeloma cell line (NS-1) in the presence of polyethylene glycol (Mr 1000). One monoclonal antibody, MCA-97 (IgM subclass), which showed reactivity with the ISHIKAWA-cell line, was obtained by a limiting dilution technique. In a cellular enzyme-linked immunospecific assay, the MCA-97 antibody reacted with all of 7 adenocarcinoma cell lines tested. In immunoperoxidase testing of formalin-fixed paraffin-embedded sections, the MCA-97 antibody reacted with most endometrial adenocarcinomas and endometrioid adenocarcinomas of the ovary, but did not react with squamous cell carcinomas or ovarian serous adenocarcinomas. In addition, it reacted with the glandular epithelium of normal tissues, such as proliferative endometrium, fallopian tube, uterine cervix and gastrointestinal tract. The reversed passive hemagglutination method demonstrated the antigen defined by MCA-97 in the sera of 5 out of 11 patients with endometrial carcinomas, and 3 out of 4 of those with ovarian endometrioid adenocarcinomas. It was not demonstrable in sera of most normal female volunteers, or any patients with cervical squamous cell carcinomas or ovarian serous adenocarcinomas. Thus, MCA-97 is of potential clinical application in diagnostic serology and pathology.

We present a 61 year old Para 4 woman who presented with stage II Infiltrating lobular carcinoma of the breast after modified radical mastectomy. She was treated with Tamoxifen for seven years. She was diagnosed with multiple myeloma during year seven post mastectomy because of wrist pain. She was treated with melphalan, prednisone and allopurinol which she tolerated well and the pain in the wrist improved. Tamoxifen was also stopped. Ten months later she presented with vaginal bleeding and was diagnosed with a poorly differentiated endometrial adenocarcinoma at hysteroscopic suction curettage and had an abdominal hysterectomy. Two years later the patient succumbed to metastatic endometrial cancer.
