Spleen cells from BALB/c mouse immunized with the human endometrial adenocarcinoma cell line (ISHIKAWA) were fused with mouse myeloma cell line (NS-1) in the presence of polyethylene glycol (Mr 1000). One monoclonal antibody, MCA-97 (IgM subclass), which showed reactivity with the ISHIKAWA-cell line, was obtained by a limiting dilution technique. In a cellular enzyme-linked immunospecific assay, the MCA-97 antibody reacted with all of 7 adenocarcinoma cell lines tested. In immunoperoxidase testing of formalin-fixed paraffin-embedded sections, the MCA-97 antibody reacted with most endometrial adenocarcinomas and endometrioid adenocarcinomas of the ovary, but did not react with squamous cell carcinomas or ovarian serous adenocarcinomas. In addition, it reacted with the glandular epithelium of normal tissues, such as proliferative endometrium, fallopian tube, uterine cervix and gastrointestinal tract. The reversed passive hemagglutination method demonstrated the antigen defined by MCA-97 in the sera of 5 out of 11 patients with endometrial carcinomas, and 3 out of 4 of those with ovarian endometrioid adenocarcinomas. It was not demonstrable in sera of most normal female volunteers, or any patients with cervical squamous cell carcinomas or ovarian serous adenocarcinomas. Thus, MCA-97 is of potential clinical application in diagnostic serology and pathology.

In order to analyze renal abnormalities in mice with polycystic kidney disease (PKD), we produced a series of monoclonal antibodies reactive with the murine kidney by hybridizing P3U1 myeloma cells with spleen cells from DBA/2 mice immunized with the kidney of adult-type PKD mice, DBA/2FG-pcy. One clone, D28, reacted specifically with the basement membrane of the proximal tubules of DBA/2 mice and DBA/2FG-pcy mice. It did not react with other parts of the murine kidney nor with other tissues such as the skin, ovary, fallopian tube, testis, lung and small intestine. While other components such as collagen IV, laminin and the core protein of proteoglycan could be found, the D28 epitope could not be found in the basement membrane of renal cysts formed in adult-type (DBA/2FG-pcy) and infant-type (C57BL/6J-cpk) PKD mice. The D28 epitope did not, however, disappear from the basement membrane of proximal tubules in other types of renal abnormalities. These results suggest that the formation of renal cysts in the proximal tubules is associated with an alteration to the proximal tubule-specific structure of the basement membrane. The D28 monoclonal antibody should prove a useful tool with which to analyze basement membrane-associated abnormalities in genetic PKD.

Multiple myeloma SET domain (MMSET) has been shown to be overexpressed in many different cancer tissues. Our study was to investigate the expression of MMSET in serous ovarian carcinoma and to evaluate its clinical significance in patients with serous ovarian carcinoma. Immunohistochemistry was performed to determine the expression of MMSET in 132 serous ovarian carcinoma, 32 normal ovarian and fallopian tube specimens. The high expression of MMSET was observed in 49.2% (65/132) in patients with serous ovarian carcinoma. MMSET high expression correlated with the advanced extent of serous ovarian carcinoma and poor outcome. MMSET may serve as a new molecular marker to predict the prognosis of serous ovarian carcinoma in the clinic.
