Previous reports have shown that the tyrosinase activities of melanosomes and soluble tyrosinase isolated from
melanoma
were diminished when these preparations were incubated with
3,4-dihydroxyphenylalanine
(dopa). It was concluded from these results that the tyrosinase activities of these preparations decreased when melanin was synthesized in these systems. The present investigation has revealed that melanin synthesized in vitro from dopa formed a complex with purified mushroom tyrosinase. The addition of melanin diminished the tyrosinase activity of the sample. These results show that the formation of the melanin-tyrosinase complex results in a decrease in the activity of the tyrosinase solution. The tyrosinase activity of the melanin-tyrosinase complex could not be increased by certain procedures which were previously found to enhance the tyrosinase activity of isolated melanosomes.

The
melanoma
of Sinclair swine exhibits several characteristics similar to human
melanoma
but demonstrates an unusually high incidence of spontaneous regression. A total of 66 finite cell lines derived from 21 swine melanotic lesions, both cutaneous and visceral, were studied in vitro over their life spans of up to 14 months. The growth characteristics of the cultures varied with the age of the swine from which the tumors were obtained. Cell cultures of tumors obtained from swine aged less than 2 months grew steadily in cluture with a population-doubling time of 120 to 180 hr until growth and division ceased after a maximum of 25 to 35 population doublings (6 to 8 passages). Cell cultures of tumors obtained from swine aged 3 months or older showed a biphasic growth pattern with an early slow growth rate (population-doubling time 120 to 160 hr), which shifted after 3 to 6 passages to a faster rate (80 to 110 hr population-doubling time) until termination of growth and division after a maximum of 75 to 85 population doublings (18 to 20 passages). The cultures were morphologically heterogeneous including cuboidal, spindle and dendritic cell types. Electron microscopy showed classic melanosomes only in the primary and passage 1 cultures although vesicular inclusions were numerous in later-passage cells. However, continued melanin synthesis was indicated by the spectroscopic characteristics of material obtained from medium of passage 8 cultures and by
DOPA
staining of cultures as advanced as passage 18.

Purified melanosomes isolated from subcutaneously growting Harding-Passey melanomas of NMRI-mice were labeled either in vitro with [14C]tyrosine or [14C]
DOPA
in the melanin portion, or in vivo in the melanin and protein portion following i. p. injection of [14C]tyrosine. Treatment of monolayer cultures of Harding-Passey
melanoma
cells (HPM-73 line) with such labeled melanosomes resulted in rapid uptake of label during the first 4 h which leveled off thereafter. A portion of the "incorporated" label could be removed by a 15 min chase with unlabeled melanosomes. Uptake of labeled melanosomes by HPM-73 cells was followed by increased cellular melanization which was not only due to melanin derived from incorporated melanosomes but primarily to newly formed melanin. Tyrosinase activity was elevated in melanosome-treated cells. Tyrosinase activity of control cells was significantly reduced following a 24 h exposure to actinomycin D or cycloheximide. On the other side, the same inhibitor treatment of melanosome-pretreated cells resulted in less inhibition of tyrosinase activity. The present findings suggest "melanophagic" properties of cultured
melanoma
cells resulting in enhanced melanogenesis after phagocytotic uptake of functionally active exogenous melanosomes.

Three cases of metastatic malignant lesions are presented. By conventional pathologic examination these lesions were diagnosed as undifferentiated carcinomas. They were studied using a chimpanzee antiserum specific for human

-associated antigens. In each case, cells from the tumor were reactive with the anti-

antiserum. The tumors were also studied using the

test. All three tumors were positive in the

test, suggesting the presence of tyrosinase within the tumor cells. The results suggest that serologic identification of

A study was made on the effect of Triton X-100 in different concentrations on the tyrosinase activity of two differently pigmented melanosome fractions of pigmented hamster
melanoma
, obtained by gradient centrifugation. Release of considerable tyrosinase activity was established spectrophotometrically in the supernatant of the treated melanosomes. The release of enzyme activity after treatment of the melanosome is probably due to a change in the permeability of the melanosome structures under the effect of the non-ionic detergent Trition X-100. A direct activating effect of Trition X-100 on the particulate enzyme should be excluded, since inhibition of
DOPA
-oxidase activity was established in the soluble tyrosinase fraction.

Alteration in

-oxidase activity of Harding-Passey

microsomal fraction tyrosinase, activation of tyrosinase after partial delipidization of the microsome, phospholipid composition of both native microsomes and the ones after delipidization have been studied. In the course of tumor growth (starting on the 14th day after tumor transplantation) the tyrosinase activity has been shown to decrease monotonously both in native microsomes and in the ones after delipidization. The phospholipid composition alters both in the process of tumor growth and after delipidization. Alteration in the tyrosinase activity in native microsomes as well as after their delipidization has been found to depend on the relative composition of certain phospholipids. In those cases a linear correlation between the enzyme activity and the phosphatydylethanolamine of phosphatydylcholine ratio has been found. This interrelationship may be accounted for by either simultenous presence of inhibitors and activators of the enzyme activity in the lipids or by changes in the membrane structure depending on the relative phospholipid composition.

cells were adapted to long-term growth in culture. The morphology of cultured
melanoma
cells ranged from dendritic to cuboidal, similar to that described for human
melanoma
cells. Doubling times of the swine

cells were also similar to those of human
melanoma
cells in vitro.

oxidase-positive cells were detected by light microscopy, and melanin and premelanosomes were detected by electron microscopy. Cell cultures could be propagated from progressing, partially regressed, and primary cutaneous lesions, as well as from visceral metastases. Thus, it appears that, under these cell culture conditions, Sinclair swine
melanoma
cells can be adapted to prolonged growth in vitro.
