Immunity to malarial infection may be transferred with immune lymphocytes. This study was designed to determine which lymphocyte type is responsible for the adoptive transfer of immunity to malarial infection. In one set of experiments, the ability of immune T and B lymphocytes, separated by passage through nylon-wool columns, to transfer immunity to infection was determined. In another experiment, the effect of killing T lymphocytes with anti-theta serum on the transfer of immunity was determined. The effect on the ability of immune lymphocyte suspensions to transfer immunity after B lymphocytes were removed from such suspensions by centrifugation on Ficoll-Hypaque gradients, after they had formed rosettes with sensitized, complement-coated sheep erythrocytes, was also determined. The ability of lymphocyte suspensions to adoptively transfer resistance to malarial infection was greatly impaired by the removal from the suspensions of differentiated B-type lymphocytes. Our results indicate that it is the differentiated B cell, most probably an antibody-producing cell, which lacks both theta antigen and the complement receptor that is responsible for conferring immunity to malaria.

Characterization and functional aspects of squirrel monkey peripheral blood mononuclear cells (PBMC), and mainly T cells, are described in the present paper; this should enable the study of cellular immune responses in an experimental model for malaria. PBMC were obtained from Ficoll-Hypaque gradient separation and fractionated into T cells and non-T cells by means of E-rosetting techniques and adherence to plastic dishes. PBMC subset phenotypes were characterized by means of monoclonal antibodies (mAb) directed against human leukocyte differentiation antigens (Ag), fluoresceinated lectins, anti-surface Ig (squirrel-monkey-specific) antibodies (Ab) and latex bead ingestion assays. PBMC functions were assayed through lymphoblastic transformation tests (LTT) in the presence of either numerous mitogenic, comitogenic and anti-mitogenic lectins or anti-human leukocyte differentiation Ag mAb. We sought to standardize reference values for lymphocyte phenotypes and functions in normal squirrel monkeys (prior to experimental infection). We also present evidence that splenectomy (generally rendered necessary for experimental human malaria infection) performed six months prior to the present investigation did not modify PBMC numbers and functions in the tested animals.

Sporozoites of the rodent malaria, Plasmodium berghei, were obtained from infected Anopheles stephensi by grinding mosquitoes, prepurifying the material in a discontinuous Hypaque gradient and further purifying by means of continuous free-flow electrophoresis. Bacteria, debris, mitochondria, mitoplasts, and other contaminants were removed in the electric field. The isolated sporozoites were morphologically intact and were positive in indirect immunofluorescence assay. They were infective to mice prior to and following free-flow electrophoretic separation. The surface of the sporozoites exhibited a polysaccharide-rich layer. The predominant surface protein labelled after surface iodination had a molecular weight between 42,000 and 46,000 daltons.
