Over 90 per cent of the thymus cells from each of twenty-six donors were T lymphocytes, identified by E-rosetting and less than 3 per cent of the cells were B lymphocytes identified by EAC-rosetting. With advancing age, the proportion of T lymphocytes decreased while that of B lymphocytes increased. The degree of (3H)thymidine incorporation of thymus cells was inversely proportional to the age of the thymus-cell donor. The PHA or PWM- induced blastogenic response of thymus cells gradually increased with advancing age when the response was expressed as the stimulation index. However, the actual rate of (3H)thymidine incorporation in all three groups was rather similar when cells were cultured with mitogens. The difference in stimulation index was due to the variation in incorporation rate in cultures without stimulants. The PHA response was approximately four-fold higher than that of PWM response. Thymus cell response to allogeneic lyphocytes, on the other hand, had no correlation with the age of thymus donor. The most surprising result in the present study was that the thymus cells from each of ten donors, aged 1-14 years, were incapable of responding to all four different recall antigens. Peripheral blood lymphocytes from nine to ten randomly selected age-matched children responded very well to one or more antigens.

Using the EAC rosette test 176 lowland black white (lbw) and lowland red white (lrw), 0-130 day-old calves were examined to determine the number of lymphocytes B. The calves came from 2 farms with different industrial breeding technology. Farm A was characterized by a considerable mortality of the animals due to diseases typical for the neonatal period, whereas in farm B no deaths were noted. As the result of examinations it has been found that calves from farm A showed an appreciably higher incidence of lymphopenia with concomitant higher maximal numbers of EAC-rosettes-forming-lymphocytes.

Rosetting properties (E, EAh, EAox, EAC rosettes) and presence of surface immunoglobulins (SIg) were examined on peripheral blood lymphocytes from 30 breast cancer patients immediately prior to therapy and 4 weeks thereafter. Therapy consisted of limited radical surgery followed by combined X-ray and telecobalt radiotherapy. The results were compared to patients who had received the same treatment 1 year ago (n = 13), 2 years ago (n = 13) and 3 to 10 years ago (n = 20). All irradiated patients exhibited a considerable leuko- and lymphopenia with a particular decrease of E and EAh rosettes, and a concommittant relative increase of EAox and EAC rosettes. SIg positive cells showed no significantly different percentages before and after therapy although in absolute counts they were similarly reduced as the other subpopulations after radiotherapy. The possible prognostic influence of radiation induced lymphopenia is discussed without coming to clear conclusions.

The immune functions in patients with mammary, pulmonary, or head and neck tumors were investigated after irradiation. The treatment caused an initial lymphopenia and longlasting depression in the lymphocyte proliferative responses to PHA, Con A and PPD. The percentages and the ratio of E and EAC rosette forming cells remained unchanged.

The cytotoxic functions of highly purified blood lymphocytes from patients with breast cancer were studied before and after radiotherapy. Addition of PHA or of rabbit antibodies to target cells (chicken erythrocytes) were chosen as two means of inducing lymphocyte cytotoxicity in vitro. The proportion of T and non-T-lymphocytes was determined by means of E and EAC rosette tests. The antibody-induced cytotoxocity of lymphocytes decreased following radiotherapy while that mediated by PHA remained unchanged. There was some reduction in the percentage of EAC rosette-forming cells. These results, as well as our earlier observations, suggest that the decrease in the peripheral blood of the proportion of lymphocytes with receptors for activated complement is responsible for changes in the antibody-mediated lymphocyte cytotoxicity.

The proportion of T and B lymphocytes in the peripheral blood was determined in patients with either mammary cancer or with various pelvic malignancies. In cancer patients studied prior to irradiation the level of cells forming either E-rosettes or EAC'-rosettes was similar to that found among healthy controls. Radiation therapy resulted in a striking lymphopenia. The level of cells with T-cell markers was diminished to a greater extent than the level of cells with B-cell markers. The relative proportion of T-cells forming high affinity E-rosettes was not reduced following radiation, so that it can be concluded that radiation affects predominantly the subpopulation of T-cells which do not form high affinity E-rosettes. Irradiation of the pelvic area resulted in a more rapid reduction of the level of T lymphocytes than irradiation of the mediastinum, although the final relative proportions of the cells were similar in both groups of patients. The results of the present study suggest that the reduction of the level of T lymphocytes following irradiation results from its effect on the lymphocytes in the major blood vessels, and that radiation of the thymus is not a prerequisite for this phenomenon.

Lymphocytes from 34 patients with active and with stable multiple sclerosis (MS) were examined by rosetting techniques. Patients with active disease had a relative lymphopenia (35 +/- 9%). When tested with erythrocytes (E) from one out of three sheep used, patients with active MS had a decrease in E-rosette forming cells (E-RFC) compared to patients with stable disease and with healthy controls. Other tests (active E-, EAET-, EA-, and EAC-RFC) did not disclose any differences between patients and controls. The results suggest that the origin of the E may be of importance in E-RFC tests.

Cell-mediated immunity (CMI) was evaluated in 11 patients with idiopathic membranous nephropathy (MN), 50 patients suffering from chronic proliferative glomerulonephritis (CGN) without renal insufficiency and 24 healthy controls. The following parameters were measured: delayed skin reactivity to purified protein derivative (PPD), circulating lymphocytes, lymphocyte cell-surface markers (En and EAC rosettes) and functional markers (mitogenic responses to Con A and PHA). The MN patients with nephrotic syndrome (NS) had less mean induration of skin reactivity and a smaller proportion reacting to the PPD antigen as compared with the control subjects. In contrast, the intensity of skin reactivity and the frequency of negative reactions in MN patients in remission and CGN were similar to those of the control subjects. During the nephrotic stage of MN the proportion of T lymphocytes decreased with simultaneous increase of the proportion of B lymphocytes. It was also found that the MN patients with NS showed impaired lymphocyte reactivity with lower Con A and PHA responses compared to the normal controls. Conversely, the mean mitogenic responses to the antigens in patients with MN in remission and CGN were similar to those of the control subjects. Thus, the majority of MN patients with NS demonstrated an impaired response in a CMI assay system. The possible significance of these phenomena in the pathophysiology of MN is discussed.

Petroleum ether extract from H. spinosa root exhibited antitumor activity in Ehrlich ascites carcinoma and Sarcoma-180 bearing mice Processed extract suppressed significantly the tumor fluid volume at the end of 3 weeks experiment. It decreased about 50% of packed cell volume and increased life span of EAC/S-180 bearing mice in a day dependent manner. Red blood cell count, hemoglobin content and white blood cell count were more or less normal after processed extract treatment of the tumor bearing mice. In tumor control animals, neutrophils increased (273.7% in EAC and 263.4% in S-180 bearing mice respectively with respect to normal mice) whereas lymphocytes decreased (60.0% in EAC and 56.5% in S-180 bearing mice respectively with respect to normal mice). It also inhibited the rapid increase of body weight of tumor bearing mice.
