Interleukin-2 receptor
(IL-2R) is expressed on activated lymphocyte after stimulation with antigen or interleukin-2 (IL-2), meanwhile soluble form of the receptor is released. Using enzyme-linked immunosorbent assay, serum IL-2R levels were determined in 34 healthy controls, 61 patients with systemic lupus erythematosus (SLE) and 32 patients with rheumatoid arthritis (RA), yielding mean +/- SD values of 355 +/- 89, 807 +/- 453 and 567 +/- 210 U/ml respectively. In both SLE and RA patients, the active disease group had more markedly elevated serum IL-2R levels compared with those of the inactive group. lymphopenia
, renal disorders, decreased C3 level and increased anti-DNA, thus make it a good parameter to monitor disease activity in SLE.

Recombinant human erythropoietin (r-HuEPO) is recognized to be effective in the treatment of anemia in patients on chronic dialysis. However, studies on the influence of r-HuEPO on the immune system are currently limited and inconsistent. In order to clarify the alteration of T and B lymphocyte subpopulations in patients on CAPD following administration of r-HuEPO, the changes in the expression of HLA-DR,
IL2R
and CD4/CD8 ratio in the peripheral blood of CAPD patients were evaluated using flow cytometry. In addition, the production of immunoglobulins in peripheral lymphocytes by enzyme immunoassays in 30 CAPD outpatients with anemia, who were treated with r-HuEPO in Tokai University Hospital, was also studied. The dose of r-HuEPO was 6,000 IU in 13 patients in group I and 9,000 IU in 17 patients in group II. The r-HuEPO was given subcutaneously once a week for up to 9 weeks. The level of hematocrit increase significantly following treatment with r-HuEPO. The numbers of lymphocytes and their CD4/CD8 ratios in peripheral blood showed no significant changes after administration of r-HuEPO. lymphocytes decreased
and normalized after administration of r-HuEPO. In comparison with healthy controls, basal formation of IgG, IgA and IgM was decreased significantly in PBMC from patients on CAPD. Following treatment with r-HuEPO, the production of IgG, IgA and IgM in PBMC from CAPD patients did not show any significant changes. In conclusion, this study suggested that the administration of r-HuEPO altered T lymphocyte function and also corrected anemia in CAPD patients.

and neutrophilia with increased numbers of mature neutrophils on post-injection Days 1 and 2, but these values returned to normal levels by post-injection Day 3.
Interleukin-2 receptor
(IL-2R) expression by peripheral blood mononuclear cells (PBMC) was evaluated by flow cytometry using a monoclonal antibody specific for bovine IL-2R alpha. Treatment with DEX significantly decreased expression of IL-2R alpha in Concanavalin A (Con A)-activated PBMC on Day 1 (P < 0.02) and on Day 2 (P < 0.1). On Day 3, expression of IL-2R alpha by PBMC was similar in control and DEX-treated calves. This decrease in IL-2R alpha expression correlated with decreased proliferative responses of PBMC to the T-cell mitogens, phytohemagglutinin (PHA) and Con A. Following in vitro treatment with recombinant human (rhu) interleukin-12 (IL-12) Con A-induced proliferative responses of PBMC tended to be higher in both groups. However, the rhu IL-12 induced increase of Con A activated proliferative responses were significantly greater in DEX-treated calves than in control calves. IL-2R alpha expression by PBMC was found to be less in calves transported 800 km in a truck as compared to that in PBMC from controls. These data suggest that stress-induced immunosuppression in calves may involve decreased IL-2R alpha expression and decreased IL-12 production. Serum chemistry results indicated a trend toward higher creatine kinase (CK) levels in DEX-treated calves. This may be due to the lysis of corticosteroid sensitive lymphocytes.

The effect of intravenous immunoglobulin (IVIG) on various laboratory values was measured immediately before and after completion of serial monthly infusions of IVIG (2 g/kg) or an equal volume of placebo over 3-12 months, in 46 patients with neuromuscular diseases participating in controlled trials. Hematological, lymphocyte subpopulation, and chemistry values were analyzed and compared. After IVIG, but not placebo, a 34% reduction in lymphocytes was noted in 44/46 patients with a selective reduction of the T cells, but not the B or
IL2R
-positive cells. Counts returned to baseline within 30 days. Creatine kinase levels decreased by 23% and sedimentation rate increased by 275% after IVIG infusion. A nondilutional, artifactual, hyponatremia and hypomagnesemia was noted with IVIG but not placebo. We conclude that IVIG affects a variety of serum chemistry and hematological values either directly or artifactually by interfering with the laboratory method used for the assays. Transient

is consistently seen, and may play a role in the immunomodulating effect of IVIG.

The objective of this investigation was to study (1) the numbers of leukocytes, (2) the proportions of lymphocytes expressing CD4, CD8, WC1, B or
IL2R
and (3) neutrophil phagocytosis and oxidative burst activity in blood around parturition in three groups of dairy cows fed different levels of a total mixed ration during the last eight weeks before calving. All cows were fed ad libitum during the first eight weeks of lactation. Serum concentration of the acute phase protein serum amyloid A (SAA), the milk somatic cell count (SCC) and disease incidence were also recorded. Special emphasis was given to the weeks just before and just after calving as dairy cows are known to be immune suppressed during this period. Dry period diet had only minor effects on leukocyte numbers, and did not influence neutrophil phagocytosis and oxidative burst. In addition, no effect was observed on disease incidence or SAA concentrations. However, an increase in the proportion of B-lymphocytes and a decrease in the proportion of WC1+ T lymphocytes were observed after calving in cows fed high or low energy rations during the dry period, but not in cows fed a medium energy ration. The weeks just before and after parturition were characterised by neutrophilia, eosinopenia,
lymphopenia
and monocytosis, but time had no effect on neutrophil phagocytosis and oxidative burst. The proportions of CD4+, CD8+, B+ and IL-2R+ lymphocytes increased in early lactation relative to the mid dry period. In addition, the concentration of SAA increased dramatically at calving. The results emphasise the need for more studies to clarify the complex interactions between nutrition and immunity during the peripartum period in dairy cattle.
