Mammalian type-C viruses contain a major internal polypeptide of about 30,000 daltons that is characterized by both intraspecies and interspecies antigenic reactivities. The interspecies reactivity of the 30,000-dalton polypeptide is an immunological marker of the mammalian type-C viruses, since even with this sensitive assay other mammalian viruses with RNA-dependent DNA polymerase activity did not contain the type-C interspecies antigen.

virus (GaLV). SiSVgp70 related antigens (AG) and immune complexes (IC) are detected both in leukemic and in nonleukemic sera. In a further analysis of these data, the prognostic significance of SiSVgp70 related AG and IC in leukemic patients was examined. The data show that the presence of SiSVgp70 related AG and IC indicates an unfavorable clinical course and a shorter survival time in acute leukemias (AL) and in chronic myelogenous leukemia in blast crisis (CML-BC). Survival data of 56 of 64 patients tested were analyzed (38 patients with AL and 18 patients with CML-BC). Patients with AL whose sera were positive for SiSVgp70 related AG and IC had a median survival time of 9.5 months after diagnosis versus 16 months for patients negative for such AG and IC. This difference in survival time was more pronounced for patients with acute nonlymphocytic leukemia (ANLL) (6.5 versus 19 months). The difference in survival between SiSVgp70 related AG- and IC-negative and positive groups as tested by life table analysis (log-rank test) is significant (P less than 0.05). Patients with AL of the AG- and/or IC-positive group had fewer complete remissions. Patients who had no remissions belong to the AG- and/or IC-positive group (P = 0.06). Patients with CML-BC whose sera were positive for SiSVgp70 related AG and/or IC had a median survival time of 2 months after diagnosis versus 7 months for patients with sera negative for such AG and IC. As tested by log-rank test, survival curves between the two groups are significantly different (P less than 0.05). These findings suggest that SiSVgp70 related AG and IC may play an important role in the course of acute leukemia and can provide useful prognostic information.

Digestion of the carbohydrate part of gp70 antigens by glycosidase treatment abrogated the precipitation mediated by IgM antibodies, whereas that mediated by IgG antibodies was not markedly affected. Data suggest that antibodies detected in human plasma samples may have oncovirus specificity, but in considerable part of cases they can be of heterophil nature.

The biological and antigenic properties of HEL-12 virus have been compared with gibbon

lymphosarcoma virus (GALV) and simian sarcoma and simian sarcoma-associated viruses, SiSV and SSAV, respectively, HEL-12 virus did not transform human or marmoset fibroblasts but rescued SiSV focus-forming activity from non-productively transformed marmoset cells (HF/SiSV-NP). Like SSAV and GALV, HEL-12 virus induced syncytia with XC cells. In addition, HEL-12 cells which did not produce virus but which contained HEL-12 proviral DNA, rescued SiSV from HF/SiSV-NP cells in co-cultivation experiments. Results of neutralization and serum cytotoxicity tests utilizing SiSV rescued by HEL-12 [SiSV-(HEL-12)] indicated that HEL-12 virus envelope proteins are very closely related to those of SSAV but readily distinguished from those of GALV. Tissues obtained at necropsy from a 7 year old male gibbon

were analyzed by electron microscopic, immunological and enzymological techniques to determine the comparative localization of tumor cells and virus throughout the body. In general, the different assays correlated well; the reverse transcriptase (RT) assay and p30 radioimmunoassay (RIA) being the most sensitive, although the RT assay was able to detect activity in one tissue scored negative by p30 RIA. Tissues were infiltrated with tumor cells to varying degrees which correlated well with the level of virus markers in most cases with the exception of the liver and kidney. In these 2 organs there was marked infiltration of free virus and tumor cells but there was no evidence of virus infection or production by these cells.
