The inhibition of de novo nucleotide, serine, and
methionine
biosynthesis in mammalian cells treated with antifolates has been attributed generally to a reduction in the levels of tetrahydrofolate cofactors. In L1210
leukemia
cells grown in tritiated folic acid (1 microM), most of the endogenous radiolabeled folates were present as formyl-substituted tetrahydrofolates (60-73%, including 10- and 5-formyl and 5,10-methenyl tetrahydrofolate), with lower levels of tetrahydrofolate (including 5,10-methylene tetrahydrofolate), 5-methyl tetrahydrofolate, and non-metabolized folic acid. Trimetrexate (1 microM) caused an elevation of dihydrofolate levels within 5 min following drug addition, from approximately 1 to 20% of the total folates. Whereas total reduced folates were preserved, losses in the levels of individual forms ranged from minor changes in the formyl tetrahydrofolates (approx. 10% decrease), to significant losses in the levels of tetrahydrofolate (approx. 60%) and 5-methyl tetrahydrofolate (95%). Under these conditions, the incorporations of [3H]deoxyuridine into TMP and [14C]glycine into purines or of [14C]formate into biosynthetic products were inhibited (69-95%). The majority (59-100%) of the endogenous radiolabeled folates in L1210 cells grown in various concentrations (0.2 to 3 microM) of [3H]folic acid was bound to soluble intracellular proteins when cell-free extracts were fractionated by rapid gel filtration or charcoal adsorption. Total intracellular folate levels increased in proportion to the changes in medium folic acid concentration; however, cofactor binding was saturable. At low concentrations, below that which supported maximal growth (less than 0.75 microM), all of the intracellular folates were protein-bound; only when maximal growth was achieved, could unbound folates be detected. Incubation with trimetrexate (1 or 10 microM), methotrexate (10 microM), or calcium leuvovorin (50 microM) did not alter significantly the levels of total and protein-bound [3H]folates in cells grown in 1 microM [3H]folic acid. Under all conditions, formyl tetrahydrofolates were the major intracellular derivatives; however, these forms were poorly represented in the bound fraction. Conversely, all of the other intracellular folate forms were completely bound. Tetrahydrofolate was the predominant protein-bound derivative in control cells; in antifolate-treated cells, both bound tetrahydrofolate and 5-methyl tetrahydrofolate were largely replaced by protein-bound dihydrofolate. This interconversion in drug-treated cells was independent of (i) sustained levels of [3H]formyl tetrahydrofolates, or (ii) high extracellular concentrations of unlabeled calcium leucovorin (50 microM). Hence, protein-bound tetrahydrofolates must not only be substrates for enzyme mediated reactions (i.e. TMP synthesis) but also must slowly equilibrate with unbound cofactor. Experiments utilizing tunicamycin, endoglycosidase H and glycopeptidase F revealed that p130 and p40 exhibited properties characteristic of glycoproteins. Labeled myeloperoxidase polypeptides were immunoprecipitated using a monospecific rabbit antibody and were identified and quantitated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis/fluorography and liquid scintillation counting. , and membrane-bound COX was solubilized and quantified by SDS/PAGE. Immunoprecipitated 35S-labelled COX from cells induced to differentiate into monocytic or granulocytic lineage were clearly detected on the autoradiograms as a protein of approx. 70 kDa molecular size, whereas only a very faint COX band was detected in untreated HL-60 cells. During both monocytic and granulocytic differentiation, COX activity (measured by the conversion of exogenous arachidonic acid into prostaglandin E2) was dramatically increased. In addition, thromboxane synthesis was preferentially enhanced during monocytic differentiation. HL-60 cells, induced to differentiate into the monocytic or granulocytic lineage, provide a useful tool to investigate the cellular mechanisms involved in regulation of the synthesis of individual prostanoid-metabolizing enzymes.

virus type I (HTLV-I) were reported previously. One recombinant VV, WR-proenv1, synthesized the authentic env protein. Both this altered HA signal peptide and the internalized env signal peptide functioned as insertion signals for the endoplasmic reticulum. Although about half of the nascent chimeric protein was cleaved at the carboxyl terminus of the internalized env signal peptide to produce the authentic env protein, the other half was cleaved at the carboxyl terminus of the altered HA signal peptide alone to synthesize the chimeric protein. These events led to a less efficient transport of the env protein produced by WR-env17 from the rough endoplasmic reticulum to the Golgi apparatus than that of the authentic env protein synthesized by WR-proenv1. The efficiency of the processing and transport of the env protein affected the immunogenicity of these two recombinant VVs.

In addition, 10% of the mice receiving 7 mg PTT.119 on days 2 and 9 were free of L1210 leukemic grafts when autopsied at the end of the 70-day observation period. The average life span of AKR mice with Ridgway osteogenic sarcoma grafts was significantly increased from 36-40 days to greater than 79 days following one or two s.c. injections of 5, 7, or 12.5 mg/kg PTT.119. Administration of PTT.119 at 14 or 14 and 21 days after tumor graft not only induced regression of palpable tumors but resulted in the absence of grafts in 60%-70% of the mice in several of the treated groups on autopsy at 180 days. In contrast, spontaneous mammary tumors were less susceptible to PTT.119; an ILS of only 15%-38% was observed in C3H/StRos mice, which eventually succumbed to tumor. Nevertheless, the total regression of initial tumors and the absence of further tumor incidence (greater than 180 days) was confirmed by autopsy in 5%-10% of the C3H/StRos mice receiving multiple i.p. injections of 5 or 7.5 mg/kg PTT.119. Human T-cell
leukemia
virus types I (HTLV-I) and II (HTLV-II) have two nonstructural trans-acting regulatory genes, tax and rex, located in the 3' region of the viral genome. The tax gene product (HTLV-I p40tax and HTLV-II p37tax) is the transcriptional activator of the viral long terminal repeat. The rex gene encodes two protein products, p27rex/p21rex and p26rex/p24rex in HTLV-I and HTLV-II, respectively. Rex acts posttranscriptionally to facilitate accumulation of full-length gag/pol and singly spliced env mRNA in the cytoplasm of HTLV-infected cells. Previous studies showed that the first ATG of the rex gene is critical for Rex production and function. The importance of the internal ATGs to Rex function is not known. However, in vitro mutagenesis of the HTLV-I rex gene has provided indirect evidence which suggests that p21rex, and by analogy HTLV-II p24rex, results from initiation at an internal AUG of the tax/rex mRNA. By using an infectious molecular clone of HTLV-II, we investigated the importance of the internal ATGs of the rex gene on Rex protein production and function. Our results indicate that p24rex of HTLV-II is not initiated at an internal AUG and that the internal
