We characterized retrovirus-induced changes in PC-12 cell function and neuronal differentiation. PC-12 cells were infected with a neurotropic retrovirus (temperature-sensitive Moloney murine
leukemia
virus, mutant BA-1). We isolated a cell clone from this infected culture that displayed altered response to nerve growth factor; increased
choline acetyltransferase
activity; and decreased basal and nerve growth factor-stimulated acetylcholinesterase activity. In addition, Kirsten murine sarcoma virus infection of and subsequent expression of the v-ras oncogene in PC-12 cells induced neurite extension, enhanced
choline acetyltransferase
activity, and limited the growth potential of the infected cells.

The neurotransmitter acetylcholine is synthesized by
choline acetyltransferase
(
ChAT
) and transported into synaptic vesicles by the vesicular acetylcholine transporter (VAChT). Recently it has been reported that the entire coding region of VAChT mRNA is located in the first intron of the
ChAT
gene. In this study,
ChAT
and VAChT mRNAs were analysed in cultured sympathetic neurones. Cholinergic differentiation factor/
leukaemia
inhibitory factor and ciliary neurotrophic factor induced strong expression of
ChAT
and VAChT mRNAs in parallel. RT-PCR analysis of
ChAT
mRNAs revealed that five types of
ChAT
transcripts which differed in the 5' non coding regions were increased. RT-PCR analysis of VAChT mRNA indicated that the cytokines induced only VAChT mRNA species which did not contain the R-exon, and not those containing the R-exon. The results indicate that
ChAT
and VAChT expressions are coordinately but differentially regulated in cultured sympathetic neurones.

Transection of the fimbria-fornix leads to retrograde degeneration of axotomized septal cholinergic neurons as manifested by loss of
choline acetyltransferase
and low-affinity nerve growth factor receptor (p75NGFR) immunoreactivity. Nerve growth factor administered into cerebral ventricles at the time of axotomy can prevent these changes, while ciliary neurotrophic factor can prevent the loss of p75NGFR immunostaining.

inhibitory factor, like ciliary neurotrophic factor, prevents the loss of p75NGFR-stained medial septal neurons after fimbria-fornix axotomy, without maintaining
choline acetyltransferase
expression in these neurons. In addition, p75NGFR-immunostained neurons had significantly smaller mean diameter after axotomy in
leukaemia
inhibitory factor- and ciliary neurotrophic factor-treated animals as compared with either nerve growth factor-treated or unlesioned animals. These findings suggest that both
leukaemia
inhibitory factor and ciliary neurotrophic factor can prevent the axotomy-induced cell death of septal cholinergic neurons, but that, in contrast to nerve growth factor, these growth factors do not maintain the expression of
choline acetyltransferase
or the normal neuronal size of these injured neurons.

This paper describes the discovery and total synthesis of bioactive marine natural products conducted in our laboratory. Clavulone, chlorovulone, bromovulone, and iodovulone are antitumor marine prostanoids isolated from the Okinawan soft coral Clavularia viridis. The synthesis of clavulone and chlorovulone was achieved from chiral 4-hydroxy-2-cyclopentenone. Marine prostanoid punaglandins 3 and 4 were synthesized via similar methodology. The chemical structures of punaglandins 3 and 4 were revised by these syntheses. Dollaberane-type diterpenoid stolonidiol and claenone were isolated from Okinawan soft coral Clavularia sp. Stolonidiol showed potent

The absolute configuration of kalihinol A was determined by applying the CD exciton chiral method. Synthesis of kalihinene X, a kalihinane-type diterpenoid, was achieved. This synthesis involves the regioselective coupling reaction of carbanion of alkyl sulfone with epoxyalcohol and construction of cis-decalin by an intramolecular Diels-Alder reaction.
