The antitumor activity of tallysomycins A and B was determined in five experimental tumor systems in mice. Tallysomycins A and B were highly active against B16 melanoma, sarcoma 180 ascites tumor and Lewis lung carcinoma, and moderately active against P388 leukemia but were without effect on lymphoid leukemia L1210. The antitumor activity of tallysomycin A was 2 to 3 times that of tallysomycin B and 3 to 17 times that of bleomycin. Tallysomycin A was about 1.5 and 4 times more toxic for mice than tallysomycin B and bleomycin, respectively, in terms of subacute LD50 values.

Testing of delayed hypersensitivity responses to recall antigens, newly encountered antigens and tumor antigens has contributed to the understanding of several immunologic factors in human neoplasia. Patients with Hodgkin's disease tend to have depressed responses to both newly encountered and recall antigens. Patients with solid tumors are more likely to be deficient only in the response to newly encountered antigens. In patients who have intact response to recall antigens, reactivity to antigen preparations from tumor and control tissue may be studied. Tumor-associated or organ-associated antigens have been demonstrated by delayed hypersensitivity responses in leukemia, Burkitt's lymphoma, malignant melanoma and carcinoma of the lung, breast, cervix uteri and intestine. Approaches to a definition of the specificity of these reactions are described. The results with these tumor antigen tests correlate strongly with the clinical course. This is a promising technique for monitoring immunotherapy. The results from tests with recall and newly encountered antigens also correlate with the clinical status and perhaps with prognosis. Various possible interpretations of these changes are discussed. Further work should be directed toward an exact definition of immunologic defects in patients with cancer and toward the use of this understanding for a rational program of immunotherapy.

Usnic acid, a lichen antibiotic, showed low-level activity in the Lewis lung carcinoma test system. In an effort to produce new agents of potential use in the treatment of lung cancer, derivatives of the natural product were synthesized and evaluated with a cytotoxicity assay. Structure--activity analysis of the cytotoxicity data indicated the importance of the lipophilicity and the beta-triketone moiety of usnic acid on cytotoxicity. No significant increases in survival of test animals over controls were shown by any of the synthetic compounds in the P388 leukemia or the Lewis lung carcinoma test systems.

Red blood cell deformability is a major determinant of capillary blood flow. In mice with L1210 leukemia and with Lewis lung carcinoma, red blood cell deformability was significantly decreased during tumor growth. In mice with L1210 leukemia, deformability was significantly decreased by day 5 after transplantation and progressively decreased through day 9. Terminally, red blood cell deformability returned to normal or above normal values. In mice with Lewis lung carcinoma, significant decreases in deformability were noted 21--28 days after transplantation and persisted throughout the remainder of the tumor course. Impaired capillary blood flow, secondary to abnormal red blood cell deformability, is therefore associated with advanced cancer.

1,4-Dihydroxy-5,8-bis(((2[(2-hydroxyethyl)amino]ethyl)amino))-9,-10-anthracenedione dihydrochloride (CL 232315; NSC 301739D), a representative of a new chemical class of compounds with antineoplastic properties, has been evaluated for antitumor activity in experimental mouse tumor systems. The compound produced significant increases in life span (ILS) and long-term survivors when tested against the P388 and L1210 leukemias as well as the solid neoplasms, B16 melanoma and Colon Tumor 26. The optimal treatment regimens resulted in a 173 to greater than 200% ILS with 20 to 80% 60-day survivors in mice with P388 leukemia, A 205% ILS with 55% 60-day survivors in mice with L1210 leukemia, and an ILS of greater than 300% with 80% 90-day survivors in mice with B16 melanoma. In contrast to Adriamycin, CL 232315 was active against the i.v. implanted L1210 leukemia and demonstrated moderate activity against P388/Adria, a subline of P388 resistant to Adriamycin. The compound was ineffective when tested against the Lewis lung carcinoma and the Ridgway osteogenic sarcoma. CL 232315 was active i.p., s.c., and i.v., but p.o. activity was not demonstrated. Schedule dependency was not observed when the compound was administered once daily for nine days, once every four days, or as a single dose.

Of 2,141 patients with scleroderma who were seen at the Mayo Clinic, Rochester, Minn, between 1959 and 1975, 78 had 87 internal malignancies. The relative frequency of the types of cancers was similar to that for the general population. The increased numbers of patients with breast and uterine carcinomas were consistent with the female preponderance in scleroderma. Contrary to previous reports, carcinoma of the lung was not the most frequent malignancy associated with scleroderma. The lymphoma-leukemia malignancies were second only to breast carcinoma in frequency, comprising about 17% of the malignancies. Both conditions developed within a three-year period in 68% (45/66) of our patients affected with systemic scleroderma, and this subgroup comprised a high mortality group. This study is not an epidemiologic one in the strictest sense, but it is a review of the experience of the Mayo Clinic with patients who have had both scleroderma and an internal malignancy.

A novel nitrosourea derivative, methyl-6-[[[(2-chloroethyl)nitrosoamino]carbonyl]-amino]-6-deoxy-alpha-D-glucopyranoside (MCNU), is a water-soluble compound in which a methoxyl group is attached to the C-1 position and an N-(2-chloroethyl)-N-nitrosoureido group is attached to the C-6 position of the glucose moiety. MCNU exhibited a marked life-prolongation or growth-inhibitory effect against mouse L1210 leukemia, adenocarcinoma 755, Nakahara-Fukuoka sarcoma, Lewis lung carcinoma, and B16 melanoma. Ip, oral, or iv administration of MCNU was markedly effective against L1210 leukemia, and the therapeutic ratio by ip administration was larger than that of chlorozotocin or CCNU. The life-prolongation effect of MCNU against established Lewis lung carcinoma was similar to that of methyl-CCNU. The bone marrow toxicity of MCNU was less than that of CCNU but considerably more than that of chlorozotocin.

A new antitumor antibiotic, named auromomycin, was isolated from the culture broth of Streptomyces macromomyceticus, a macromomycin-producing strain. The antibiotic was recovered from the culture filtrate by salting out with ammonium sulfate and further purified by successive application of ion-exchange chromatography on Amberlite IRA-93 (Cl form) and DEAE-Sephadex (OH form), Gel filtration on Sephadex G-50 and hydrophobic chromatography on Octyl-Sepharose CL-4B. The antibiotic is an acidic polypeptide with a molecular weitht of 12,500 and an isoelectric point of pH 5.4 and consists of 16 different amino acids. It has characteristic absorption maxima at 273 nm and 357 nm in the ultraviolet spectrum and two minima at 280 nm and 350 nm in the optical rotatory dispersion spectrum. Auromomycin exhibits antibacterial activity not only against Gram-positive bacteria, but also Gram-negative bacteria. Antitumor activities of auromomycin were revealed against EHRLICH ascites carcinoma, ascites sarcoma 180, L1210 leukemia and LEWIS lung carcinoma. Auromomycin was found to be converted into macromomycin by adsorption chromatography on Amberlite XAD.

Macromomycin (MCR) is a polypeptide antimuor antibiotic isolated from the culture broth of Streptomyces macromomyceticus. Antitumor activities of MCR were examined against three different tumor system, i.e., EHRLICH ascites carcinoma, L1210 leukemia and LEWIS lung carcinoma. Daily intraperitoneal treatment with MCR for 5 days showed a strong inhibition against EHRLICH ascites carcinoma. Both single and repeated intraperitoneal injections of MCR were effective over a wide dose range against intraperitoneally inoculated L1210 leukemia and MCR intravenously administered was also active against intravenously inoculated L1210 leukemia. Daily local subcutaneous injections of MCR produced the prolongation of life span of mice to which LEWIS lung carcinoma was subcutaneously inoculated with some cured mice, but daily intraperitoneal injections of MCR showed no activity. Single intravenous administration of MCR inhibited early LEWIS lung carcinoma, but not advanced LEWIS lung carcinoma. The combination of MCR with aracytidine, or cyclophosphamide showed a synergistic activity against L1210 leukemia. MCR was not inactivated by treatment with serum, although neocarzinostatin was markedly inactivated by the same treatment.

The chemotherapeutic activity of analogs of the anthracycline antibiotic, nogalamycin, was investigated in the P388 and L1210 leukemias and the B16 melanoma in mice. Among the compounds tested, 7-con-O-methylnogarol was found to have superior activity. Depending on the route and schedule of administration, increases in lifespan (ILS) in excess of 100% were observed in all three tumor systems. Additional testing of 7-con-O-methylnogarol demonstrated significant activity in the murine colon 26 and colon 38 tumors and the CD8F1 mammary tumor. 7-Con-O-methylnogarol was not significantly effective against murine Lewis lung carcinoma, although ILSs of 38% and 29% were achieved in two experiments. Activity was observed against ip inoculated P388 leukemia after ip, sc, oral, and iv drug administration. 7-Con-O-methylnogarol was also highly active (ILS greater than or equal to 120%) after ip drug administration to mice with iv inoculated P388 leukemia. Significant ILS values resulted from a variety of schedules of administration against ip inoculated P388 leukemia and B16 melanoma. Experiments in which the time of single-dose administration was varied prior to the time of ip P388 leukemia inoculation showed that residual drug or bioactive drug-related materials remained in mice for 8 hours after 50 mg/kg administered ip and for 48 hours after 200 mg/kg administered sc.
