A new modification of our detergent technique for the preparation of nuclei for flow cytometric DNA analysis is described. The attainment of low coefficients of variation of the peaks and of quantitative staining of nuclei from different tissues was a problem with the original method. This was solved in the new modification by trypsinization of the unfixed nuclei. The nuclei were stabilized by spermine. A simple procedure for long-term storage of samples at -80 degrees C was integrated into the method. The fluorescence of the nuclei was stable for at least 3 hours after staining. Light exposure protection of the samples was essential. No cell loss was caused by storage or staining. The method was successfully applied on samples including: (a) Normal tissues--human lymphocytes, granulocytes and spleen. Mouse lymphocytes, bone marrow, spleen, liver, kidney and thymus. (b) Human neoplasms--lung cancer, breast cancer, lymphoma, leukemia, bladder cancer and cancer of the oral cavity. (c) Human tumors in nude mice--breast cancer, lung cancer, melanoma and colon cancer. (d) Mouse ascites tumors--JB-1, L 1210, Ehrlich and P 383. It therefore seems well suited as a routine clinical procedure.

Changes in natural killer (NK) cell, antibody-dependent cell-mediated cytotoxicity (ADCC) and interferon activities after administration of Nocardia rubra cell wall skeleton (N-CWS) to a group of subjects with high risk of lung cancer (retired workers of the Okunojima poison gas factory) were studied. No difference in NK cell activity against K-562 derived from chronic myelogeneous leukemia was observed among normal controls, non N-CWS-injected workers and N-CWS-injected workers. None of the normals or N-CWS-injected retired workers showed low NK cell activity (less than 10% cytotoxicity), but among the non N-CWS-injected workers 6.3% showed reduced activity. Following injection of N-CWS, NK cell activity against K-562 and T-24 derived from bladder cancer was significantly elevated by the second week and then gradually reverted to the preinjection level. When a second injection was administered at the twelfth week, a similar elevation was noted. Further, a significant increase was noted in ADCC activity two weeks after N-CWS injection. There was a significant elevation of interferon at the second week after N-CWS injection. It was considered that N-CWS affects the NK cell activity enhancement by serum interferon.

This study evaluated cause-specific mortality among 3,161 men who were employed in the aerospace division of a rubber manufacturing company. Compared to other production workers at the plant, aerospace workers in deicer and fuel cell manufacturing jobs experienced a 60% excess of deaths from lung cancer. Deicer and fuel cell workers who were under 65 years of age had lung cancer rates that were approximately twice those of other rubber workers of comparable age. Aerospace division employees also had elevated rates of bladder cancer, leukemia, lymphoma, and multiple myeloma. However, detailed analyses suggested that, with the exception of lung cancer, these cancer excesses were not likely to be attributable to employment in the aerospace division.

Cancer incidence was studied among 2,448 male rubber workers employed for 18 months or more with at least eight years of observation time. A suggested increased risk of bladder cancer, lymphoma, and leukemia was found in the footwear department; the number of observed to expected cases was 4/2.81, 5/2.26, and 4/1.76, respectively. An unexpected increased risk of pancreatic cancer was found for the maintenance-workshop department, the observed to expected numbers being 6/0.90.

The mortality experience of 10,322 men employed in woodworking industries was compared with that of 406,798 nonwoodworkers. All subjects were enrolled in an American Cancer Society study and followed prospectively from 1959 through 1972. Age-adjusted rates of death from all causes and from all cancers were not higher in the woodworker group, but excess rates were observed for cancers of the lung, stomach, and bladder, as well as nonmelanoma skin cancer and possibly leukemia. Woodworkers experienced significantly decreased rates of colon-rectum cancer and coronary heart disease. The elevated cancer rates could not be explained by cigarette smoking habits. If anything, there is evidence to suggest a possible interaction between employment in woodworking trades and heavy cigarette smoking, in increasing the risk of lung and bladder cancer.

Although it is over 30 years since an excess of bladder cancer was first identified in British rubber workers, the fear has persisted that this hazard could still be affecting men working in the industry today. Furthermore, suspicions have also arisen that other and hitherto unsuspected excesses of cancer might be occurring. For these reasons 33 815 men, who first started work in the industry between 1 January 1946 and 31 December 1960, have been followed up to 31 December 1975 to ascertain the number of deaths attributable to malignant disease and to compare these with the expected number calculated from the published mortality rates applicable to the male population of England and Wales and Scotland. The findings confirm the absence of any excess mortality from bladder cancer among men entering the industry after 1 January 1951 (the presumed bladder carcinogens were withdrawn from production processes in July 1949), but they confirm also a statistically significant excess of both lung and stomach cancer mortality. A small excess of oesophageal cancer was also observed in both the tyre and general rubber goods manufacturing sectors. American reports of an excess of leukaemia among rubber workers receive only limited support from the present study, where a small numerical excess of deaths from leukaemia is not statistically significant. A special feature of the study is the adoption of an analytical method that permits taking into account the long latent period of induction of occupational cancer.

Several Louisiana parishes (counties) using the Mississippi River for their source of public drinking water have the highest mortality rates (1950-69) in the United States for several cancers. Therefore, a case-control mortality study on cancer of the liver, brain, pancreas, bladder, kidney, prostate, rectum, colon, esophagus, stomach, non-Hodgkin's lymphoma, multiple myeloma, leukemia, Hodgkin's disease, lung; breast and malignant melanoma, from 1960 to 1975 in South Louisiana parishes grouped for similarities in industrial characteristics, having approximately equal exposure of the population to surface and groundwater, was conducted. Noncancer deaths were randomly selected as controls and matched to the case death on age, race, sex, and year and parish group of death. Water source at death was assigned based on the residence at death and described as surface or ground and chlorinated or nonchlorinated. A significantly increased risk for surface, chlorinated water use was noted for rectal cancer. No risk could be demonstrated for colon cancer. The risk noted for bladder cancer by other investigators is not substantiated. Brain cancer risk appears to be associated with chlorinated groundwater, but this may be industrial confounding. Breast cancer demonstrated a slight, but significant, risk associated with surface chlorinated water. This risk, however, might be due to confounding of rural life style, early childbearing and large families with nonchlorinated water found in these settings. Chlorination risk for kidney cancer was not significant. No risk was observed in association with surface water for other cancers of the gastrointestinal or urinary tract. Multiple myeloma was significantly associated with a risk from ground water.

This report describes mortality occurring between 1940 and mid-1978 among 29,087 men and women employed in a rubber plant for at least two years. Mortality patterns for the period July 1, 1974, to July 1, 1978, were compared to previously published findings for January 1, 1940, through June 30, 1974. Expected numbers of deaths were based on U.S. general population mortality data. There were excess deaths from bladder cancer and leukemia among white male union members during both follow-up periods. During recent follow-up of white male union members employed for at least five years, there were excesses in deaths from three additional cancers: esophageal cancer (11 observed/4.8 expected), biliary and liver cancer (6 observed/3.3 expected) and lymphoma and multiple myeloma (14 observed/5.8 expected). Evidence from other studies of rubber workers suggests that observed excesses in deaths from bladder cancer and leukemia are related to work-place exposures. The present findings suggest that occupational exposures etiologically relevant to these diseases may not have been reduced in recent years or that sufficient time has not elapsed for such reductions to result in decreased mortality. Further investigation is required to clarify the contribution of occupational factors to observed excesses in deaths form cancers of the esophagus and the biliary passages and liver and from lymphoma and multiple myeloma.

Studies of the cancer risk among rubber workers published since 1968 are reviewed. The total material examined comprised 80,000 workers and 4000 cancer deaths. An increased risk of stomach cancer, lung cancer, bladder cancer, and leukemia was found. The increased risk of cancer at a specific anatomic site was correlated with the mode of exposure specific for the working area or division. On the other hand, no clear exposure-risk relationship could be demonstrated. It is likely that there has been an increased risk of cancer since the elimination of alpha- and beta-naphthylamine by 1950. However, the etiologic fraction is less than that due to prevalent exposures increasing the risk of common cancers at specific anatomic sites.

Interstitial deletions of the short arm of chromosome 9 are associated with glioma, acute lymphoblastic leukemia, melanoma, mesothelioma, lung cancer, and bladder cancer. The distal breakpoints of the deletions (in relation to the centromere) in 14 glioma and leukemia cell lines have been mapped within the 400 kb IFN gene cluster located at band 9p21. To obtain information about the mechanism of these deletions, we have isolated and analyzed the nucleotide sequences at the breakpoint junctions in two glioma-derived cell lines. The A1235 cell line has a complex rearrangement of chromosome 9, including a deletion and an inversion that results in two breakpoint junctions. Both breakpoints of the distal inversion junction occurred within AT-rich regions. In the A172 cell line, a tandem heptamer repeat was found on either side of the deletion breakpoint junction. The distal breakpoint occurred 5' of IFNA2; the 256 bp sequenced from the proximal side of the breakpoint revealed 95% homology to long interspersed nuclear elements. One- and two-base-pair overlaps were observed at these junctions. The possible role of sequence overlaps, and repetitive sequences, in the rearrangement is discussed.
