The clinical and pathological findings in 19 ferrets (Mustela putorius furo) with malignant lymphoma are reviewed. Peripubescent ferrets had rapidly progressive stage IV high grade immunoblastic or small non-cleaved cell lymphoma. Adult ferrets had stage II or IV low grade diffuse small lymphocytic (DSL) lymphoma, stage IV high grade small non-cleaved cell lymphoma, or stage IV high grade immunoblastic polymorphous (IBP) lymphoma. Three ferrets had concurrent IBP and DSL lymphoma involving different organs. The IBP admixture of immunoblasts, large atypical lymphocytes, Reed-Sternberg-like cells, lymphoblasts and small lymphocytes has been associated with certain retrovirally associated lymphomas and nodal hyperplasias in man, non-human primates and cats. Aleutian disease, a parvovirus-induced lymphoproliferative disease, also involves clinical and histological features similar to certain lymphomas in ferrets. Seven ferrets tested were seronegative for feline leukaemia virus antigen. Only one of eight ferrets was positive for Aleutian parvovirus antibody. The clinical and pathological findings are suggestive of a viral aetiology for certain lymphomas in ferrets.

Cluster outbreaks of lymphoma and leukemia have been associated with viral infections in many species including humans, cattle, and cats. This study describes epidemiological, clinical, and pathological features of cluster outbreaks of lymphoma in multiferret households and examines and compares the Aleutian disease virus (ADV) and feline leukemia virus (FeLV) status of cases, ferrets at risk, and controls. Three ferret groups with 21 cases of histologically diagnosed lymphoma (12.6% cumulative incidence) and their cohabitants (n = 35) were examined and compared with three control groups (n = 52) of cohabitating ferrets without lymphoma. A familial distribution was observed in one group but most cases were not consanguinous. Ferrets greater than 3 years of age developed chronic disease in two of the groups and 2-year-old adults had acute disease in the remaining group. Lymphocytosis, splenomegaly, and lymphadenopathy were prominent features. Histologically, predominantly small noncleaved cell and polymorphous lymphoid lesions were observed. All of the ferrets with lymphoma that were tested for ADV and FeLV using serology or PCR were negative. The rate of ADV antibody among cases or ferrets at risk was not significantly different from controls. None of the cluster ferrets were seropositive for FeLV p27 antigen using a monoclonal ELISA. Infection with a novel ferret virus is suspected, but an etiological agent has not yet been identified.

Three cases of juvenile mediastinal lymphoma developed in a laboratory colony of ferrets. Two ferrets became acutely moribund, and one was found dead with no preceding signs of illness. Splenomegaly, hepatomegaly, and a large thoracic mass were the primary features in each case. All three ferrets had multiorgan metastasis of the tumor. Two ferrets were tested for feline leukemia virus and Aleutian disease virus with negative results.

Studies of reactions in animals with hereditary diseases (Sapphire minks highly sensitive to Aleutian disease virus, ADV; CBA mice with 60-70% incidence of tumors; AKR mice with 90% incidence of leukemia) showed that serum DNAse activity in these animals dropped after injection of a foreign heterogeneous DNA and remained decreased during 72 h. By contrast, serum DNAse activity considerably and persistently increased after injection of DNA in Standard minks resistant to ADV, C57BI/6J mice with 1% tumor incidence, and random-bred albino mice. Presumably the capacity of standard minks to react to a foreign heterogeneous DNA by increase of DNAse activity ensures their resistance to DNA-containing ADV, while incapacity of Sapphire minks to respond to DNA by DNAse activity induction makes them sensitive to ADV. A similar relationship between the capacity to react to DNA by changes in serum DNAse activity and capacity to inherit a disease was detected in mouse strains.

The therapeutic effects of melatonin against viral infections, with emphasis on the Venezuelan equine encephalomyelitis (VEE), are reviewed. Melatonin has been shown to prevent paralysis and death in mice infected with the encephalomyocarditis virus and to decrease viremia. Melatonin also postpones the onset of the disease produced by Semliki Forest virus inoculation and reduces the mortality of West Nile virus-infected mice stressed by either isolation or dexamethasone injection. An increase in the host resistance to the virus via a peripheral immunostimulatory activity is considered responsible for these effects. It has also been demonstrated that melatonin protects some strains of mink against Aleutian disease, and prevents the reduction of B- and T-cells as well as Th1 cytokine secretion in mice infected with leukemia retrovirus. In VEE-infected mice, melatonin postpones the onset of the disease and death for several days and reduces the mortality rate. This protective effect seems to be due to the increase in the production of interleukin-1beta (IL-1beta), as 100% of the infected mice treated with melatonin die when IL-1beta is blocked with antimurine IL-1beta antibodies. Although melatonin administration raises serum levels of tumor necrosis factor-alpha (TNF-alpha) and interferon-gamma (IFN-gamma), the mortality observed in neutralization experiments with the corresponding anticytokine antibodies, suggests that neither TNF-alpha nor IFN-gamma are essential for the protective effect of melatonin on murine VEE virus infection. Melatonin treatment also enhances the efficiency of immunization against the VEE virus. Reactive oxygen species have been implicated in the dissemination of this virus, and their deleterious effects may be diminished by melatonin. This indole inhibits nitric oxide synthetase activity and it is a potent scavenger of nitric oxide, which also plays an important role in the spread of the VEE virus. In conclusion, the immunomodulatory, antioxidant, and neuroprotective effects of melatonin suggest that this indole must be considered as an additional therapeutic alternative to fight viral diseases.

This paper offers an overview of important veterinary viral diseases of mammals stemming from aberrant immune response. Diseases reviewed comprise those due to lentiviruses of equine infectious anaemia, visna/maedi and caprine arthritis encephalitis and feline immunodeficiency. Diseases caused by viruses of feline infectious peritonitis, feline leukaemia, canine distemper and aquatic counterparts, Aleutian disease and malignant catarrhal fever. We also consider prospects of immunoprophylaxis for the diseases and briefly other control measures. It should be realised that the outlook for effective vaccines for many of the diseases is remote. This paper describes the current status of vaccine research and the difficulties encountered during their development.
