Ten liquid media were compared under standard conditions for their ability to support the growth of Legionella pneumophila. Modified gonococcal-ferric cysteine broth (without sodium chloride) supplemented with 1% yeast extract yielded the best overall growth of the one strain of L. pneumophila examined. Growth rates were independent of pH changes which occurred during incubation. The growth rates of 10 different strains of L.pneumophila were compared in this medium. There appeared to be little difference in the growth rates of strains passaged frequently or infrequently, or between environmental and clinical isolates. Moderate aeration resulted in a faster growth rate and in approximately a 1 log10 higher final cell concentration as compared to a static broth culture. These experiments demonstrate that there are moderate to marked differences among the various media described in the literature and that no liquid medium yet developed supports rapid growth of L. pneumophila incubated without shaking.

The culture conditions of Afipia felis, A. broomeae, A. clevelandensis and three unnamed Afipia genospecies were investigated on BCY agar supplemented with different substances known as growth factors of Legionella spp. and, furthermore, with sodium chloride and other salts. The organisms were found to be susceptible to a certain degree to byproducts of the autoclaving which are scavenged by activated by charcoal. Growth was weakly enhanced by ferric pyrophosphate, cystein.HCl, and alpha-ketoglutarate. These substances are no obligatory growth factors. The optimal pH value was about 6.8. Afipia spp. showed a strong susceptibility to NaCl and other salts. They possess phosphatase, phosphoamidase, phosphodiesterase, a weak sulfatase, glycine aminopeptidase, and L-lysine aminopeptidase. The strains differed with regard to other proteases and aminopeptidases. The decimal reduction times of A. felis at 55 degrees C and 60 degrees C were 11 min, < 1 min, respectively.

The effects of various concentrations of sodium chloride solutions (0.1%-3%) and different temperatures (4, 10, 20, 30 and 37 degrees C) on survival of Legionella pneumophila were investigated. It was found that at temperatures between 4 degrees C and 20 degrees C, Legionella organisms survived in salt solutions up to 3% NaCl. Only the combination of high temperatures, i.e. 30 degrees C and 37 degrees C, with NaCl concentrations over 1.5%, reduced cell numbers significantly. It was interesting to note that the addition of small amounts of NaCl (0.1%-0.5%) enhanced survival of Leg. pneumophila, suggesting a protective effect of NaCl. In order to obtain information about conditions encountered in the environment, the survival experiments were repeated in sterile sea water from the Baltic Sea and the North Sea. The marked bacterial die-off, especially at higher temperatures, was not observed in natural sea water. All these results indicate that Leg. pneumophila can survive in the marine environment.

Invasive aspergillosis of the lungs and the central nervous system and Legionella pneumophilia serotype 1 infection of the lungs were diagnosed in a 22-month old child during inpatient induction treatment for T-lymphoblastic leukemia. Environmental investigations i.e. samples from the hospital water system did not reveal any Legionella. The patient may have been exposed to waterborne pathogens despite terminal water filtration due to a technical device to release residual tap water from the hose after showering. A sodium chloride nose spray was found to be contaminated with the A. fumigatus isolate of the patient.
