Isolated rat heart was subjected to Langendorff perfusion for 10 minutes of total global ischemia, and reperfusion for 15 minutes. The content of myocardial intracellular calcium, content of oxygen free radical induced lipid peroxide mitabolite-malondialdehyde (MDA), content of Lactic dehydrogenase (LDH) in coronary effluent, and the change of myocardial morphology were studied. The effects of flunarizine(FNZ) and/or vitamin C(Vit C) on the above parameters were observed. The results showed that, at 15 minutes of reperfusion, the contents of myocardial intracellular calcium, of MDA in myocaredium and of LDH in coronary effluent in the FNZ + Vit C group, Vit C group and FNZ group were all lower than those in the control group, and the change of morphology in the three groups was also slighter than that in the control group. When FNZ and Vit C were administered together, the effect was more marked and equivalent to the sum total of the effects of the two drugs. These suggest that both FNZ and Vit C reduce reperfusion injury through suppression of myocardium calcium overload and oxygen free radical damage.

Tumor necrosis factor-alpha (TNF-alpha) level, tissue-typed plasminogen activator(t-PA) activity and PA inhibitor (PAI) activity were determined in three groups: (1) 25 NIDDM patients with silent myocardial ischemia (SMI) or silent cerebral ischemia (SCI); (2) 18NIDDM patients without SMI or SCI; (3) 20 age-matched normal controls. Diagnosis of SMI or SCI was based on the finding of ischemic evidence by SPECT of myocardiotomograph or cerebrotomograph. All patients ECG and blood pressure were normal, and they had no history of clinical symptoms and signs of MI or CI. The result showed that the TNF-alpha level and PAI activity in the ischemia group were the highest and the t-PA activity in the ischemia group was the lowest, as compared with those in the other two groups respectively. It suggests that in NIDDM patients who have high TNF-alpha, high PAI activity, low t-PA, and even no symptoms and signs of MI or CI, anticoagulant therapy might be useful to prevent the progression of diabetic macroangiopathies.

Langendorff perfusion isolated rat heart was subjected to total global ischemia (coronary flow rate is equal to zero) for 10 minutes and reperfusion for 15 minutes. The heart rate (HR), left ventricular developed pressure (LVDP), coronary flow rate (CFR), electrocardiogram (ECG) and the effects of calcium antagonist-flunarizine (FNZ) and/or oxygen free radical scavenger--vitamine C on the above parameters were observed. The results showed that FNZ dilated coronary vessel (P < 0.01) and had a slight negative chronotropic effect, but it had no effect on LVP. Vitamine C did not affect HR, LVP and CFR. The recovery of the product of HR and LVDP-Rate Pressure Product (RPP) in the FNZ + Vit. C group, Vit. C group and FNZ group was significantly higher than that in the control group (P < 0.05) at ten minutes reperfusion. All the results suggest that FNZ and Vit. C may improve the recovery of heart function after reperfusion.

To examine the effect of ischemia/reperfusion in cardiac surgery with cardiopulmonary bypass (CPB) on nitric oxide (NO) levels of peripheral blood, venous levels of NO2- and NO3- were measured at multiple time-points before, during, and after CPB by means of high-pressure liquid chromatograph to reflect NO concentration indirectly in 20 patients. The results showed there was a linear regression between the decreased trend of NO release at the period of peri-CPB is related to the endothelial cell activation caused by ischemia/reperfusion at the same time. Therefore it is necessary to explore the regulation and action of NO release during systemic inflammatory response procedure in CPB.

This article reports the apoptosis and necrosis of neurons following ischemia-reperfusion in the brain of rats. The focal ischemia model was established by occluding the middle cerebral artery for 2 h and reperfusing for 0.5-48 h. The neuronal changes were investigated by HE staining, TUNEL and agarose gel-electrophoresis. The foci of ischemia were in the preoptic area, striatum and cortex. At 0.5 h of reperfusion, there were quite a few apoptotic neurons in the preoptic ischemic focus, but only a few scattered apoptotic cells were seen in the striatum and cortex. During 3-6 h of reperfusion, the number of apoptotic cells increased and the necrotic cells appeared. The number of apoptotic cells reached a peak at 12-24 h and the in morphology varied, and they were mainly located in the inner boundary of infarct. At 48 h, the apoptotic cells decreased; and the necrotic cells increased. Agarose gel-electrophoresis showed the dense smear and ladder pattern of DNA fragments, the apoptosis and necrosis of neurons coexisted, and apoptosis contributed to the enlargement of the ischemic infarct.

Vascular endothelial growth factor(VEGF) is a highly specific mitogen promoting the formation of blood vessels in embryogenesis and wound healing. It is also a potent inducer of vascular permeability. It is a member of the cystine knot growth factor superfamily. A detailed structural and functional characterization of the interactions between VEGF and its receptors is a prerequisite for the design of molecule antagonists. These structural characterizations and biological properties make VEGF an important research object in the fields of neovascularization of ischemia tissues, prognosis of cancers, tumor metastasis, and the gene therapy.

Male SD rats(n = 29) were subjected to 2 hours' middle cerebral artery occlusion and were killed at various times of reperfusion (0.5-48 hours). The histological features of neuronal changes, the macrophagic response and the relationship between them were investigated. The neurons underwent acute changes(shrinking or swollen), and the pyknotic apoptotic neurons were occasionally seen within 3 hours. The number of apoptotic neurons increased with recirculation time. The scattered red neurons and ghost cells were first found at 6 hours of reperfusion. Many apoptotic neurons, apoptotic bodies were detected in the penumbra of ischemic core at 48 hours. Lectin histochemistry with GSI-B4 was performed to observe the macrophage. The results showed the active microglia and macrophage from meninges presented at 12 hours. The macrophages from parenchyma vessels appeared at 24 hours. Reactive microglia and extrinsic macrophage aggregated into the band of macrophage which phagocytized the apoptotic neurons. The results indicate that, firstly, it is important to prevent the neuronal apoptosis and reduce the volume of infarct in stroke patients; secondly, irreversible neuronal lesions simultaneously stimulate microglial activation and the presence of extrinsic macrophages from meninges was earlier than the appearance of the macrophages from parenchyma blood-monocytes. The authors also suggest that the data on the neuronal changes over time after ischemia and reperfusion be useful reference materials for selecting the most timely medication to treat cerebral infarction.

The objective of this study was to assess the prophylactic and therapeutic effects of Yi Qi Tong Mai Oral Liquid (YQTMOL) on acute myocardial ischemia. The models of ischemia were made, in which the anterior coronary arteries of anesthetized dogs were ligated. Four groups of dogs were examined and compared. We determined the extent of their myocardial ischemia and infarction by means of epicardial mapping and N-BT staining. The results showed that in the treatment groups, the increase of sigma-ST was inhibited 30-180 minutes after the oral administration of YQTMOL(compared with control group, P < 0.01), and the sigma-ST was decreased (P < 0.05). Furthermore, in the group dosed with YQTMOL (10 g/kg), the ratios of "area of infarction/heart weight" and "area of infarction/ventricle weight" both decreased (compared with control group, P < 0.05). These findings indicated that YQTMOL reduced the area of myocardial ischemia and the degree of myocardial infarction in dogs, so it could be effective for the prevention and treatment of acute myocardial ischemia.

In this study, four metal porphyrins (tetraphenylporphyrinesulfonatoiron, tetraphenylporphyrinesulfon-atocopper, tetraphenylporphyrinesulfonatomanganese, and tetraphenyl porrphyrinesulfonatocobalt) were used as dual function mimicry of superoxide dismutase (SOD) and catalase (CAT). The first function, scavenging O2.-, has been proved by using riboflavine-methionine photoreduction method in the concentration range of 10(-5)-10(-6) mol/L. The second function, scavenging H2O2, has been demonstrated by means of spectrophotometry, and the percentage of decomposing H2O2 increases with the concentration of the imitating compounds. Metal porphyrins, SOD and CAT were measured by the liver homogenate technique of mice and the results showed that they had obvious action of decreasing the lipid peroxidation. Tetraphenylporphyrinesulfonatomanganese was measured by isolated ischemia reperfusion heart of rats. The results indicated that the Mn-TPPS4 could reduce the damages of active oxygen and had evidently protective effects on isolated ischemia-reperfusion mocardium of rats.
