Using a three-panel runway task, the effects of NIK-247 on impairment of working memory produced by scopolamine, hippocampal lesions, and cerebral ischemia were investigated in rats; these effects were compared with those of the well-known cholinesterase inhibitors, tetrahydroaminoacridine (THA) and physostigmine. Intraperitoneal injection of scopolamine (0.56 mg/kg) significantly increased the number of errors (pushes made on the two incorrect panels of the three-panel gates located at four choice points). NIK-247 (3.2-18 mg/kg PO), THA (1-10 mg/kg PO), and physostigmine (0.1 and 0.32 mg/kg IP) dose-dependently reduced the increase in errors induced by scopolamine. NIK-247 (32 mg/kg) was also effective in reducing the increase in errors produced by lesions of the dorsal hippocampus. A 5-min period of cerebral ischemia markedly increased the number of errors. NIK-247 (3.2 and 10 mg/kg), given immediately after blood flow recirculation and again 20 min before the runway test carried out 24 h after ischemia, significantly reduced the increase in errors expected to occur after ischemia. Tetrahydroaminoacridine (3.2 mg/kg) and physostigmine (0.1 mg/kg) similarly reversed the increased errors in ischemic rats. These results suggest that NIK-247 alleviates the impairment of working memory produced by scopolamine, hippocampal lesions, and cerebral ischemia, possibly through activation of the central cholinergic system.

The aim of this paper was to study the anatomical relationship between the piriformis muscle and the sciatic nerve with regard to the possibility of neurological deficit after THA. The incidence of anatomical variation of both structures is 15-30% in the literature. The authors studied 91 cadavers and found an atypical relationship in 19 cases (20.9%). In this study individual variations were found with the following frequency: The sciatic nerve exits below the piriformis muscle in 79.1% of the cases. The sciatic nerve separates into two divisions above the piriformis, one branch passing through the muscle, the other below it (14.3%). An unsplit nerve passes through the piriformis muscle in 2.2%. The nerve separates into two divisions above the piriformis, one branch exiting above the muscle and passing along its dorsal aspect, the second exiting distally below the muscle in 4.4%. The most common reasons for sciatic nerve injury in surgery of the hip joint are direct injuries, ischemia of the nerve tissue, compression or excessive distraction of the nerve, compression by bone cement, thermal damage during cement polymerization, injury during THA dislocation, compression by hematoma, bone prominence or an implanted acetabular component. According to the presented anatomical study, overstretching of the nerve itself or its branches in the area of the pelvitrochanteric muscles after their release from their origin can be another mechanism. Such overstretching can appear in the presence of some of the aforementioned anatomical variants.

18F-labeled fluorodeoxyglucose positron emission tomography (18F-FDG PET) is the most sensitive tool for studying brain metabolism in vivo. We investigated the image patterns of 18F-FDG PET during reperfusion injury and correlated changes of whole brain blood flow utilizing a rat myocardial ischemia/reperfusion injury (MIRI) model. The results assessed by echocardiography indicated resultant cardiac dysfunction after ischemia-reperfusion in the rat heart. It was found that the average standardized uptake value (SUVaverage) of the whole brain was significantly decreased in model rats, and the glucose uptake of different brain regions including accumbens core/shell (Acb), left caudate putamen (LCPu), hippocampus (HIP), left hypothalamus (LHYP), olfactory (OLF), superior colliculus (SC), right midbrain (RMID), ventral tegmental area (VTA), inferior colliculus (IC) and left thalamus whole (LTHA) was significantly decreased in MIRI rats whereas no significant difference was found in the SUVaverage of amygdala (AMY), right CPu, RHYP, right HYP, left MID, right THA, pons and medulla oblongata (MO). These 18F-FDG PET data provide a reliable identification method for brain metabolic changes in rats with MIRI.
