G-box (CCACGTGG) like sequences are present in a variety of plant promoters and in many cases they have been demonstrated to be required for maximal expression of the corresponding gene. A nuclear protein, GBF, interacts specifically with the G-box motif of several RBCS and CAB promoters. Here we describe the isolation of a cDNA from Arabidopsis thaliana that encodes a protein, designated GBF-1, with DNA binding properties similar to GBF. GBF-1 is characterized by a basic/leucine zipper motif which is strikingly similar to the wheat protein identified as
HBP
-1. GBF-1 also interacts with an oligonucleotide derived from the wheat histone 3 promoter containing the binding site (hexamer, TGACGT) for
HBP
-1. This DNA element also contains a G-box-like motif, modification of which results in loss in binding of GBF-1.

A phenobarbital-inducible rat hepatic microsomal UDP-glucuronosyltransferase (UDPGT) that catalyzes the glucuronidation of 4-hydroxybiphenyl (4-HBP) has been purified to homogeneity. This UDPGT has an apparent subunit molecular weight of 52,500, as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The 4-
HBP
UDPGT was shown to catalyze the glucuronidation of 4-
HBP
, 4-methylumbelliferone, and p-nitrophenol but did not react with testosterone, androsterone, morphine, chloramphenicol, 4-hydroxycoumarin, or 7-methoxycoumarin. The apparent Km of 4-
HBP
UDPGT for 4-
HBP
was determined to be 0.26 mM and for UDPGA was 1.0 mM. Upon treatment with endoglycosidase H, the 4-
HBP
UDPGT underwent about a 2000-dalton decrease in subunit molecular weight, suggesting that this protein is N-glycosylated. Additionally, this protein demonstrated immunoreactivity with antibodies raised in rabbit against rat 17 beta-hydroxysteroid and 3 alpha-hydroxysteroid UDPGTs. This work describes the purification and characterization of a 4-
HBP
UDPGT from rat liver microsomes and, furthermore, provides evidence that suggests that this UDPGT is different from another UDPGT previously shown to react with 4-
HBP
and chloramphenicol.

Wheat transcription factors
HBP
-1a and
HBP
-1b bind to the hexamer motif, ACGTCA, of wheat histone gene promoters.
HBP
-1b also binds to the hexamer motif in the promoter of the 35S RNA gene of cauliflower mosaic virus, whereas
HBP
-1a does not. A cDNA clone encoding
HBP
-1b was isolated on the basis of its binding specificity to the hexamer motif. The deduced amino acid sequence indicates that
HBP
-1b, like
HBP
-1a, belongs to a leucine zipper class of transcription factors. Mutational analyses of the
HBP
-1a and -1b encoded cDNAs revealed that truncated polypeptides containing the leucine zipper and basic regions are sufficient for DNA binding.
HBP
-1a and -1b form homodimers, as expected from earlier studies on this class of transcription factors, but did not form heterodimers. Although the hexamer motif or its homologs exist in several plant genes,
HBP
-1a and -1b exhibited the highest binding affinity to the hexamer motif in the histone promoters, suggesting that both DNA binding proteins are involved in transcriptional regulation of wheat histone genes.

We identified two novel DNA-binding proteins, ssDBP-1 and ssDBP-2, in wheat germ nuclear extract that interact with the proximal sequences of the promoter regions of the wheat histone H3 and H4 genes. Mobility shift and methylation interference assays have demonstrated that these factors specifically bind to the single-strand DNA which partially overlaps the hexamer and octamer cis-elements of the H3 promoter. Both proteins are distinguishable from
HBP
-1a and
HBP
-1b which specifically bind to the H3 hexamer sequence. These ssDNA-binding proteins are supposed to regulate the transcription of the wheat histone genes.

Hypertension
has been reported to occur in 50 to 75 percent of subjects with autosomal dominant polycystic kidney disease (ADPKD) prior to the onset of marked renal insufficiency but concurrent with cystic deformation of the renal parenchyma. The present study was undertaken to examine whether the renal structural abnormalities are greater in hypertensive (
HBP
) versus normotensive (NBP) male and female patients with ADPKD who were matched within gender groups for age, body surface area, serum creatinine concentration (males
HBP
1.2 +/- 0.02 vs. NBP 1.1 +/- 0.03 mg/dl. NS: females
HBP
0.9 +/- 0.03 vs. NBP 0.9 +/- 0.02 mg/dl, NS) and creatinine clearance (males
HBP
100 +/- 3 vs. NBP 108 +/- 3 ml/min/1.73 m2, NS: females
HBP
97 +/- 3 vs. NBP 96 +/- 2 ml/min/1.73 m2, NS). Renal volume was significantly greater in the
HBP
compared to the NBP group (males
HBP
624 +/- 47 vs. NBP 390 +/- 43 cm3, P less than 0.0005; females
HBP
446 +/- 32 vs. NBP 338 +/- 24 cm3, P less than 0.002). Since increased renal volume is due to increased cysts, the results indicate that the early high incidence of
hypertension
in ADPKD correlates with the renal structural abnormalities in this disorder.

During the changeover from the eighth to the ninth revision of the "International Classification of Diseases: Manual of the International Statistical Classification of Diseases, Injuries, and Causes of Death" (ICD), there were several major alterations of coding for the rubrics Ischemic Heart Disease (IHD) and
Hypertension
(
HBP
). As expected, these changes caused major discontinuities for IHD and HPB. These discontinuities were not, however, uniform over sex-race groups. When examined by component ICD codes, the discontinuities were found to vary in both magnitude and direction among the groups. In addition to discontinuity, there was a change in the rate of decline for IHD and
HBP
after the changeover. This rate of decline varied as well by sex-race group. In general, the decline among blacks was slower than among whites. Earlier studies that assessed IHD mortality have used different groupings of ICD codes to obviate the discontinuity, and researchers have observed a similar differential decline. These results should be viewed with caution because of the potential impact of differential coding on sex-race groups. As preparations are made for ICD-10, special attention should be given to the preservation of epidemiologic continuity to provide better assessment of trends in population subgroups.

In 10 patients with adult respiratory distress syndrome, we studied the effects on respiratory system mechanics of two levels of positive end-expiratory pressure (PEEP), best PEEP (BP) and half of this value (
HBP
), using a respiratory inductive plethysmograph (RIP) combined with a super syringe. We found the following. 1) Inflation compliance of pressure-volume (PV) curves did not change significantly. 2) End-expiratory volume increased with
HBP
and further with BP (278 +/- 186 and 464 +/- 313 ml, respectively, P less than 0.01). This increase was positively correlated with inflation compliance for
HBP
and BP (r = 0.794, P less than 0.01 and r = 0.876, P less than 0.01, respectively). 3) No dynamic hyper-inflation was detected on mechanical ventilation at zero end-expiratory pressure (ZEEP), and the time constant of the respiratory system was in the normal range (0.79 +/- 0.21 s). 4) Hysteresis of PVrip curves, which were corrected for gas exchange, decreased significantly with PEEP (P less than 0.05). We conclude that PEEP does not change inflation PV curve but induces an increase in intrathoracic volume whose magnitude is related to compliance and PEEP level. The reduction of hysteresis with PEEP suggests less gas trapping and thus a functional improvement.

A cDNA clone isolated from a lambda gt11 expression library of teratocarcinoma OTT6050 specifies for a glycoprotein with a molecular weight of about 44,000. The new glycoprotein was termed heparin binding protein-44 (HBP-44), since it was absorbed to a heparin-agarose column and was eluted from it by a buffer containing 1.5 M NaCl.
HBP
-44 mRNA was intensely expressed in PYS-2 parietal endoderm cells and in the kidney, and the RNA level increased about 10-fold during differentiation of F9 embryonal carcinoma cells to parietal endoderm cells. From the cDNA sequence,
HBP
-44 was concluded to be rich in charged amino acids, and large segments of the protein appeared to form alpha-helixes. The protein was considered to be anchored to the membrane by a cluster of hydrophobic amino acids present in the N-terminal region. Indeed, the N-terminal sequence of
HBP
-44 was homologous to asialoglycoprotein receptor, which is anchored to the membrane by the N-terminal region. Furthermore, a portion of the N-terminal region of
HBP
-44 was homologous to the leucine zipper domain. Except for the N-terminal region,
HBP
-44 had over-all homology with structural proteins such as myosin heavy chain. We propose that
HBP
-44 is extruded from plasma membranes and interacts with heparin and related molecules and that it is involved in the interactions of plasma membranes with basement membranes.

A majority of histone genes are expressed in the S phase during the cell cycle. Using the gene expression system of transformed sunflower cells into which wheat histone H3 gene was introduced by the Ti-plasmid gene transfer technique, we determined three cis-acting control sequences (hexameric, octameric, and nonameric motifs) which seemed to confer the S-phase-specific transcription of wheat histone genes. Furthermore, as candidates for regulatory transcription factors, three nuclear DNA-binding proteins
HBP
-1a,
HBP
-1b, and
HBP
-2 that interact with the hexameric and nonameric motifs were identified. The structural analysis of the cDNA of
HBP
-1a revealed that a nuclear protein has the leucine-zipper structure and a DNA-binding motif. The hexameric motif in the H3 gene was also seen in cauliflower mosaic virus 35S (CaMV 35S) promoter and shown to function as a regulatory element of this promoter. The wheat
HBP
-1b can interact with the hexameric motif of the CaMV 35S promoter. Much attention has been paid to the significance of the hexameric sequences within the H3 and CaMV 35S promoters and the DNA-binding proteins
HBP
-1a and
HBP
-1b.

This study examined the association between aerobic, caloric exercise expenditure and life status (living vs deceased) as well as the prevalence rates of
hypertension
(
HBP
) and cardiovascular disease (CVD) in former male athletes (ATH) and non-athletes (N-ATH). The initial survey for this study was done in 1952. Follow-up surveys of respondents were done in 1960, 1968, 1976, and 1984. The present study used all subjects who responded fully to activity and health questions in 1976 and who were reported as either dead or alive (not lost to follow-up) in 1984. A total of 348 subjects (185 ATH, 163 N-ATH) were assessed and caloric expenditure groups were established by kilocalories (kcal) of aerobic exercise per week; 0 kcal (group 1), 1-399 kcal (group 2), 400-899 kcal (group 3), 900-1499 kcal (group 4), 1500-2499 kcal (group 5), and 2500+ kcal (group 6). Only activity considered to be aerobic was used in the establishment of the aerobic categories. Death rate was highest in groups 1 and 2. Subjects in group 1 tended to be the oldest. Year of birth (age) (P less than 0.001) and CVD (P less than 0.05) as reported in 1976 were significantly related to mortality between 1976 and 1984. College athletic status and 1976 exercise level were not significantly related to mortality. Prevalence of CVD and
HBP
was highest in groups 1 and 6, suggesting a moderate amount of aerobic activity as optimal.
