Combating heart disease is one of the challenging problems of biomedical science today. Towards this goal an indigenous preparation 'Trikatu' (a herbal combination containing
Piper
longum (fruit),
Piper
nigrum (fruit) and Zingiber officinale (rhizome) dry powder) was fed to normal and cholesterol fed male Rattus norvegicus to ascertain its efficacy as a hypolipidaemic agent. Its effects on body weight, blood and tissue (aortic, cardiac and hepatic) lipids--total, free and esterified cholesterol, low density lipoprotein(LDL) and high density lipoprotein(HDL) cholesterol, triglycerides and phospholipids--and the atherogenic index were measured. It was found that 'Trikatu' by virtue of its ability to reduce triglycerides and LDL cholesterol and to increase HDL cholesterol can reduce the risk of
hyperlipidaemia
and atherosclerosis. Hence 'Trikatu' can be used as a potent hypolipidaemic agent and it can reduce the atherosclerosis associated with a high fat diet.

The present study was conducted to develop new inhibitors of pancreatic lipase and alpha-glucosidase from Chinese dietary herbs. Sixty-three dietary herbs from 39 taxonomic families were selected and extracted with aqueous ethanol or water. The extracts were then tested with in vitro enzyme assays for their ability to inhibit pancreatic lipase and alpha-glucosidase activities. Orlistat and acarbose were used as two positive controls. longum and Morus alba showed strong pancreatic lipase inhibitory effects with IC50 at (28.00 +/- 5.51), (5.24 +/- 0.51), (14.76 +/- 2.58), (4.78 +/- 0.58), (3.41 +/- 0.67) mg x L(-1), respectively. These extracts also showed potent alpha-glucosidase inhibitory activities with IC50 at (1.98 +/- 0.13), (0. 18 + 0.007), (0.71 +/- 0.08), (0.077 +/- 0.005), (0.089 +/- 0.006) g x L(-1), respectively. The results provide useful information for developing new drugs or natural health products for

A novel thermostable form of piperlonguminine (GB-N) was extracted from medicinal plant

longum in efforts to explore the bioactive components underlying the mechanism of
Piper
longum in reducing plasma lipids. In vitro, HepG2 cells were employed to investigate the effects of GB-N on regulating cellular total cholesterol and low-density lipoprotein (LDL) receptor (LDLR) mRNA abundance, while high-fat and high-cholesterol diet-induced hyperlipidemic Wistar rats were used to investigate in vivo effects of GB-N. Cellular total cholesterol assay showed that GB-N dose-dependently reduced cellular total cholesterol in HepG2 cells in the presence and absence of elevated plasma cholesterol levels by 25% and 32%, respectively. Reverse transcription PCR assay showed that LDLR mRNA abundance was up-regulated dose-dependently by 142% via GB-N treatment in HepG2 cells. Animal experiment revealed that GB-N dose-dependently reduced serum total cholesterol by 26%, triglyceride by 47%, LDL cholesterol by 30%, while increased serum high-density lipoprotein (HDL) cholesterol by 524% in diet-induced hyperlipidemic Wistar rats. In conclusion, the results suggest the potential therapeutic uses of GB-N in the prevention and treatment of
hyperlipidemia
and related diseases.

The effects of effective part group on
hyperlipidemia
in animal were studied. The SD rats, hamsters and Kunming mouse were divided into blank group, model group. The positive control group and test group were fed with normal diet, blank and other groups were fed with high fat diet (mouse only a single intraperitoneal injection of egg yolk ). The corresponding concentration of solvent, simvastatin, effective part group of emulsion were given gavage once daily. The animal serum total cholesterol (TC) , triglyceride (TG) , low density lipoprotein (LDL) , high density lipoprotein (HDL) and liver TC, TG contents were determined to observe the effects of the effective fractions on blood lipid regulating function. Comparing with control group, the animial
