Absolute capacity for IL2 production by human peripheral blood mononuclear cells (PBMC) was studied using 12-O-tetradecanoylphorbol 13-acetate (TPA) and
calcium
ionophore A23187, which act synergistically, to induce lymphokine synthesis. Culture parameters were optimized for the TPA/A23187 stimulation such that maximal IL2 titers were produced with a high degree of reproducibility. Thus, using a synthetic medium, TPA/A23187 at 20/50ng/ml respectively, and a cell concentration of 2.5 X 10(6)/ml, IL2 titers in the cultures increased linearly over a period of 96h, reaching values at least 15-fold higher than with lectin stimulation. This allowed for determinations of IL2 synthetic capacity in individual blood samples. Large fluctuations in normal IL2 production (range 1775-10654 BRMP U/ml at 48h) were observed among 23 normal persons. A statistically significant lower IL2 productive capacity was observed in the age group above 40 as compared to those under 40. The lower rates of IL2 synthesis in a group of patients with
Hodgkin's disease
was seen only among those who had undergone immunosuppressive therapy; newly diagnosed cases fell within the normal range.

We have obtained new insights into the behavior of a class of excitable systems when a stimulus, or parameter, is slowly tuned through a threshold value. Such systems do not accommodate no matter how slowly a stimulus ramp is applied, and the stimulus value at onset of repetitive activity shows a curious, nonmonotonic dependence on ramp speed. (Jakobsson, E. and R. Guttman. Biophys. J. 1980. 31:293-298.) demonstrated this for squid axon and for the
Hodgkin
-Huxley (HH) model. Furthermore, they showed theoretically that for moderately slow ramps the threshold increases as the ramp speed decreases, but for much slower ramp speeds threshold decreases as the ramp speed decreases. This latter feature was found surprising and it was suggested that the HH model, and squid axon in low
calcium
, exhibits reverse accommodation. We have found that reverse accommodation reflects the influence of persistent random fluctuations, and is a feature of all such excitable systems. We have derived an analytic condition which yields an approximation for threshold in the case of a slow ramp when the effect of fluctuations are negligible. This condition predicts, and numerical calculations confirm, that the onset of oscillations occurs beyond the critical stimulus value which is predicted by treating the stimulus intensity as a static parameter, i.e., the dynamic aspect of a ramp leads to a delay in the onset. The condition further demonstrates a memory effect, i.e., firing threshold is dependent on the initial state of the system. For very slow ramps then, fluctuations diminish both the delay and memory effects. We characterize the class of excitable systems for which these behaviors are expected, and we illustrate the phenomena for the HH model and for a model of cAMP-receptor dynamics in Dictyostelium discoideum.

Properties of the delayed outward current (IK) in ventricular myocytes of the guinea-pig were studied using the whole cell clamp method. The experiments were performed under conditions in which IK was enhanced by application of isoproterenol while the

current was eliminated by
Ca2+
-removal and by the addition of Cd2+. The reversal potential (Erev) of IK, determined from the current tails, was about 10 mV less negative than the K+ equilibrium potential. This was estimated by examining the reversal potential of the inward rectifier K+ current in Ba2+-containing solution, or from the Nernst equation. The Erev--log[K+]o relationship had a slope of 49 mV per tenfold change in [K+]o. In Na+-free solution, Erev became more negative. Thus, although the major charge carriers in IK are K+ ions, Na+ ions may also contribute in part to this current. The PNa/PK ratio in IK, calculated by applying a Goldman-
Hodgkin
-Katz relation to the reversal potential, was 0.016. The activation of IK during depolarization showed a sigmoidal time course at the onset, while the time course of the current tails was monoexponential at voltages more negative than-50 mV, but biexponential at more positive voltages. These observations can be explained by the conductance equation of the
Hodgkin
-Huxley type in which the kinetic variable is raised to the second power. These and other features of IK observed in the ventricular cells are discussed in comparison to the properties of similar current systems reported in other cardiac preparations.

A 35-year-old white male with rheumatoid arthritis who had developed hypercalcemia, hypercalciuria, and nephrolithiasis was found to be abnormally sensitive to vitamin D as a result of lack of regulation of circulating 1,25-dihydroxyvitamin D (1,25-(OH)2D). An increase in daily intake of vitamin D from 10 micrograms (400 units) per day to 50 micrograms (2000 units) per day produced an abnormal elevation in serum 1,25-(OH)2D, hypercalcemia, and hypercalciuria which were corrected by prednisone. Serum 25-hydroxyvitamin D initially was abnormally low, and increased with vitamin D to values which were in the low normal range. There were significant positive correlations between serum 1,25-(OH)2D (p less than .05) and serum
calcium
and between serum 1,25-(OH)2D and urinary
calcium
(p less than .05). Serum immunoreactive parathyroid hormone, initially in the lower range of normal, decreased further during hypercalcemia. A radiograph of the chest, gallium scan, and serum angiotensin-converting enzyme activity were normal. No granulomas or evidence of lymphoma were found in biopsies of the liver and of several lymph nodes. It is concluded that the abnormal
calcium
metabolism in this patient resulted from increased circulating 1,25-(OH)2D and that the defect in vitamin D metabolism was not related to sarcoidosis, other granulomatous disease,
Hodgkin's disease
, or lymphoma. The relationship, if any, of the abnormal metabolism of vitamin D and
calcium
to rheumatoid arthritis remains to be established.

Human granulocyte-macrophage colony-stimulating factor (GM-CSF) was purified from 3 liters of serum-free conditioned medium of the
Hodgkin
's tumor cell line L428 KSA. The conditioned medium contained a high specific activity of 2.5 X 10(5) units of total colony-stimulating factor per mg protein. Colony-stimulating factor activity was determined by colony formation by human fetal liver cells or mouse bone marrow cells. The latter bioassay discriminated colony-stimulating factor 1, a subclass specific for monocyte/macrophage production, and G-CSF, specific for granulopoiesis, from GM-CSF. The starting material contained predominantly GM-CSF with CSF-1 and G-CSF constituting 10% and 12%, respectively, of the total activity. A seven-stage purification scheme was employed. The first stage involved concentration by batch chromatography on
calcium
phosphate gel. Subsequent stages involved gel filtration on Ultrogel AcA44, affinity chromatography on concanavalin A-Sepharose, batch chromatography on
calcium
phosphate gel and high-performance liquid chromatography on C1 reversed-phase (TSK TMS-250), gel permeation and C8 reversed-phase columns. The purified material showed a single disperse band, having an Mr of 30,000, by silver staining on sodium dodecyl sulfate polyacrylamide gel electrophoresis. An amino-terminal sequence of 20 amino acids was determined in a gas-phase sequencer with 500 ng of purified material. The sequence was identical to that predicted from the cDNA sequence. It was active on human fetal liver cells with half-maximum colony formation at 1 X 10(-12) M, but was not active on mouse bone narrow cells.

's lymphomas were treated with combinations of cyclophosphamide, vincristine sulfate, prednisone, and doxorubicin with and without 2 weekly doses of oral methotrexate in "intermediate" doses, followed by
calcium
leucovorin rescue. The addition of methotrexate did not increase the complete remission rate, the remission duration, or the survival (P value = 1.0, 0.74, and 0.78, respectively) in patients who did not have previous chemotherapy treatment. In previously treated patients, the complete remission rate was somewhat higher and the remission duration and survival were longer among those patients treated with the methotrexate containing program; however these differences were not statistically significant (P values = 0.88, 0.81, and 0.46, respectively). There was substantial morbidity and mortality during treatment with both treatment arms, among patients aged more than 60 years.

-Huxley type equations: a Ca inward current, a time-independent K outward current ("rectifier" current), and a time dependent slowly activated K outward current ("delayed rectifier" current). The maximum upstroke velocity of CaAP was compared with the Ca inward current. It turned out that the discrepancy between Vmax and maximum Ca current can be tremendous. A non-linearity between Vmax and gCa has been described. Furthermore, a twice peaking of the
calcium
current during CaAP cannot be excluded. The existence of outward currents in nearly the same size as the Ca current even during the upstroke of CaAP seriously question the use of CaAP as a measure of

We investigated the ionic interactions responsible for the characteristic nonrepetitive spike activity of amacrine cells. First we measured 4 pharmacologically separable ionic components: a voltage-gated, transient inward sodium current, a voltage-gated, sustained inward
calcium
current, a
calcium
-gated, sustained outward potassium current, and a voltage-gated, transient outward potassium current. The measurements provided the time course and magnitudes of the underlying conductances as functions of voltage. Each current was simulated following conventional
Hodgkin
-Huxley theory. A composite of the simulated currents was analytically reassembled to generate an approximation of the voltage response to a current step. By artificially varying the magnitude and kinetics of the different conductances in the simulation, we determined the range of values that supported the nonrepetitive spike-like response. Amacrine cells tend to remain refractory following an initial spike because (1) the entire activation range for potassium is located at positive potentials with respect to sodium inactivation, so sodium inactivation is never fully extinguished, and (2) the fully activated sodium conductance is of insufficient magnitude to subsequently reach threshold, given this residual inactivation. Shifting the sodium inactivation range by 10 mV, or increasing sodium conductance by 5 times, leads to a more repetitive form of activity. Changes in the magnitude, time course, or activation range of the potassium conductance cannot alter these conditions.

According to the available evidence it seems probable that the humoral hypercalcemia in this patient resulted from production of 1,25-dihydroxyvitamin D3 in the tumor.
