(HD) patients and paired age- and sex-matched controls were tested for serum antibodies to
Epstein
-Barr virus (EBV), six strains of influenza virus and Simian sarcoma virus/Simian sarcoma-associated virus [SSV(SSAV)] antigens. HD sera showed significantly elevated titers against EBV and both increased incidence and mean titer of antibodies to the envelope glycoprotein of SSV(SSAV), whereas testing against the influenza viruses revealed no differences between HD and controls. A focus reduction assay demonstrated a low incidence in HD and controls of sera with neutralizing effects against SSV(SSAV) and the "baboon" C-type virus component of the human HL-23 virus complex, which seemed coupled to HL-A type W19.

The families had: malignant melanoma;

-Barr virus-associated malignancies and a birth defect syndrome; non-
Hodgkin lymphoma
and diverse carcinomas;
Hodgkin's lymphoma
and adenocarcinomas; and acute myelogenous leukemia. In addition to the Philadelphia chromosome in chronic myelogenous leukemia patients, karyotypic abnormalities, especially breaks and fragments, were found in 29% of cancer family members, but were inconsistent and usually attributable to radiotherapy. Mean sister chromatid exchange values were normal in chronic myelogenous leukemia, but low (by t-test) in tumor patients and their blood relatives in cancer-prone families. In tumor patients, mean sister chromatid exchange levels fell as age increased. After adjusting for this age effect, no significant differences remained among groups. In patients at high risk of cancer (because they have chronic myelogenous leukemia or a strong family history of cancer), spontaneous sister chromatid exchange rates were not a marker of cancer risk.

. The lymphoid depletion variant was accompanied by high antibody titers to the virus and had a worse prognosis than the nodular-sclerotic variant with lower antibody titers but better prognosis. The level of antibody in
lymphogranulomatosis
was stable. No relationship between the antibody level and the stage and results of the therapy given was found. In sera from patients with reticulosarcoma and lymphosarcoma no significant rise of antibody titers to EBV was found as compared with a group of normal donors, while in acute leukosis there was a tend for a decline in antibody titers.

In the literature EBV is considered a possible etiologic factor for
Hodgkin's disease
(HD). We investigated 187 cases of HD for the presence of
Epstein
Barr virus using the polymerase chain reaction (PCR) technique on formalin fixed, paraffin embedded lymph node tissue to clarify the clinical importance of the incidence of this genome. The 187 cases included all subtypes. 66 cases (35.2%) were positive for EBV DNA. The statistical analysis of follow-up data from 130 patients revealed no influence of EBV DNA on survival time. In our investigation detection of EBV DNA by PCR showed no prognostic relevance for patients with HD.

Hodgkin's disease
(HD) provide little information about the EBV-status in the lymphocyte predominant subtype (nodular paragranuloma) which is due to the small number of cases investigated. Therefore we studied 99 typical cases of nodular paragranuloma for the presence of EBV-DNA using the polymerase chain reaction (PCR) technique. Genomic DNA was amplifiable in 71 cases; 29 cases (= 40%) were positive for EBV-DNA (EBNA-1). In situ hybridization revealed EBV-RNA (EBER-1) in L&H cells and in a few lymphocytes. PCR results were correlated to clinical follow-up data and did not show any statistically significant relationship between EBV positivity and survival of the patients.

Ig-gene rearrangement studies revealed clonal populations, however, no translocations of the c-myc oncogene could be detected. The lymphomas developing with high frequency in SIV-induced immunodeficiency resemble a major subtype of human EBV-associated AIDS lymphomas. This animal model can therefore be used to further elucidate interactions of HIV and EBV in AIDS-related lymphomagenesis.
