A hitherto unknown family of six-coordinate vanadyl porphyrins of the sterically crowded, nonplanar 2,3,7,8,12,13,17,18-octaethyl-5,10,15,20-tetranitroporphyrin incorporating axial ligand L [where L is pyridine, tetrahydrofuran (THF), or
methanol
(MeOH)] has been isolated as VO(tn-OEP)(L) in the solid phase for the first time and also structurally characterized. The presence of four electron-withdrawing, bulky nitro groups at the meso positions of vanadyl octaethylporphyrins severely distorts the porphyrin macrocycles and significantly enhances the affinity for the axial ligands, where even weak sigma-donating ligands, such as MeOH, bind strongly enough to be isolable in the solid phase and that too under the offset effects of the macrocyclic distortions. Thus, the axial ligand affinity is influenced by both the electronic and conformational effect, which cannot be separated completely in this series. The solid-state magnetic measurements and their typical electron paramagnetic resonance (EPR) spectrum show the presence of a single, unpaired electron, consistent with V(IV) formulation. The VO stretching frequency for VO(tn-OEP) occurs as a sharp, strong peak at 1008 cm(-1), which is consistent with five-coordinate vanadyl porphyrins, while VO(tn-OEP)(L) displays a strong band at lower wavenumbers. The downshift in nu(VO) upon axial coordination increases with increasing donor strength of the axial ligands; for pyridine, the downshift is 30 cm(-1), while for THF and MeOH, the downshifts are nearly 18 cm(-1). X-ray structure determinations authenticate axial coordination in a purely saddle-distorted porphyrin macrocycle for all of the complexes reported here in which V-Np distances are significantly shorter, while the porphyrin cores have been expanded on axial ligand coordination. As a result, vanadium atoms are more inplane than in a five-coordinate species. The binding of L does not change the spin or metal oxidation states (V(IV), d(1)-system) of the complexes; therefore, the changes observed are truly the reflections of axial ligand coordination. Electrochemical data obtained from cyclic voltammetric studies reveal that the complexes are much easier to reduce (by approximately 1200 mV) but more difficult to oxidize (by approximately 500 mV) as compared to nearly planar VO(OEP). The complexes undergo two one-electron oxidations due to pi-cation radical and dication formation and three one-electron reductions. The first two reductions are because of pi-anion radical and dianion formation, while the third quasi-reversible reduction is assigned to a metal-centered process (V(IV) --> V(III)). These results can be useful for identifying the interaction of the vanadyl porphyrins with the biological targets in their reported involvement in potent insulinomimetic activity and in anti-
HIV
agents.

HIV
-infected patients on antiretroviral therapies were cut into about 1 mm length segments and drugs were extracted by first shaking the samples with
methanol
in a 37 degrees C water bath overnight (>14 h), followed by methyl tert-butyl ether/ethyl acetate (1:1) extraction under weak alkaline conditions. The extracted lopinavir and ritonavir were separated by reversed-phase chromatography and detected by tandem mass spectrometry in electrospray positive ionization mode with multiple reaction monitoring (MRM), while efavirenz was monitored in negative ionization MRM mode. This method was validated from 0.01 to 4.0 ng/mg hair for ritonavir and 0.05-20 ng/mg hair for lopinavir and efavirenz by using 2 mg of a human hair sample. The interday and intraday assay precision (coefficients of variation, CV) for spiked quality control (QC) samples at low, medium and high concentrations were within 15% and accuracy ranged from 89% to 110%. Assay reproducibility was also demonstrated by analysis of incurred hair QC samples (CV <14%). No significant matrix ionization suppression was observed. This developed method allowed for the monitoring of these target medications in the hair samples of
HIV
-infected women on antiretroviral therapy in an observational study using small amounts of hair.

The quantification of the
HIV
integrase inhibitor raltegravir in blood plasma is described using solid phase extraction (SPE) coupled with an accurate high-performance liquid chromatography assay with ultraviolet (UV) detection. The method was validated over the range of 20-10,000 ng/mL using simple sample preparation and chromatography. The SPE method was optimized to be selective and highly efficient. The buffer's ionic strength and pH were optimized for retaining RAL and the internal standard on the column, the percentage of
methanol
was optimized in the cleaning step to remove unwanted plasma contaminants, and the type and amount of acid was optimized for complete elution of the compounds. This method has no interference with other potentially co-administered antiretrovirals or common drugs. Average recoveries for the extraction method were consistently high: 90% for raltegravir and 90% for the internal standard diazepam. This method was found to be accurate and precise. Within day (n=6) and between day (n=18) accuracies ranged from 97.5 to 104.4%. Within-day (n=6) and between-day (n=18) precision ranged from 1.4 to 3.8%, and from 2.4 to 7.9%, respectively. This is the first published method to use simple UV technology and reliable SPE extraction methodology for the quantification of raltegravir in human plasma. This method can be easily implemented in most bioanalytical laboratories.

Nucleoside reverse transcriptase inhibitors (NRTIs) need to enter cells to act against the
HIV
-1. Human organic cation transporters (hOCT1-3) are expressed and active in CD4+ T cells, the main target of
HIV
-1, and have been associated with antiviral uptake in different tissues. In this study, we examined whether NRTIs interact and are substrates of hOCT in cells stably expressing these transporters. Using [(3)H]N-methyl-4-phenylpyridinium, we found a high-affinity interaction among abacavir [[(1S,4R)-4-[2-amino-6-(cyclopropylamino)purin-9-yl]-cyclopent-2-enyl]
methanol
sulfate] (ABC); <0.08 nM], azidothymidine [3'-azido-3'-deoxythymidine (AZT); <0.4 nM], tenofovir disoproxil fumarate (<1.0 nM), and emtricitabine (<2.5 nM) and hOCTs. Using a wide range of concentrations of lamivudine [(-)-beta-L-2',3'-dideoxy-3'-thiacyitidine (3TC)], we determined two different binding sites for hOCTs: a high-affinity site (K(d1) = 12.3-15.4 pM) and a low-affinity site (K(d2) = 1.9-3.4 mM). Measuring direct uptake of [(3)H]3TC and inhibition with hOCT substrates, we identified 3TC as a novel substrate for hOCT1, 2, and 3, with hOCT1 as the most efficient transporter (K(m) = 1.25 +/- 0.1 mM; V(max) = 10.40 +/- 0.32 nmol/mg protein/min; V(max)/K(m) = 8.32 +/- 0.40 microl/mg protein/min). In drug-drug interaction experiments, we analyzed cis-inhibition of [(3)H]3TC uptake by ABC and AZT and found that 40 to 50% was inhibited at low concentrations of the drugs (K(i) = 22-500 pM). These data reveal that NRTIs experience a high-affinity interaction with hOCTs, suggesting a putative role for these drugs as modulators of hOCT activity. Finally, 3TC is a novel substrate for hOCTs and the inhibition of its uptake at low concentrations of ABC and AZT could have implications for the pharmacokinetics of 3TC.

In regions with high prevalence of

and malaria, co-infection of both diseases is common; hence, there is a high possibility of concurrent administration of antiretroviral and antimalarial drugs. This study describes a new ion-pair reversed-phase high-performance liquid chromatographic (HPLC) method for simultaneous determinations of ritonavir (RTV), quinine (QN), and its major metabolite, 3-hydroxyquinine (3-HQN), in human plasma. Following a simple extraction with diethyl-ether under alkaline conditions, chromatographic separation was achieved on a 5-mum particle size C-18 column (200 mm x 4.6mm I.D.) using a mobile phase consisting of

:acetonitrile:0.02 M potassium dihydrogen phosphate (15:10:75) containing 75 mmol/L perchloric acid (pH 2.8). Retention times for RTV, 3-HQN, QN and the internal standard were 2.8, 4.0, 7.0 and 12 min, respectively. The limits of detection and validated lower limits of quantitation were 10 and 12.5 ng/ml for RTV while the corresponding values were 5 and 70 ng/ml for both QN and 3-HQN, respectively. The new HPLC method is simple, rapid, selective, reproducible and cost-effective. As demonstrated in three volunteers, it will facilitate the conducting of simultaneous therapeutic monitoring of quinine and ritonavir in patients concurrently receiving both drugs.

The cloned PTD-NPY fusion gene was identified by PCR and restriction enzyme digestion and sequenced. The exact recombinant plasmid was linearized by Sac I and integrated by electrotransformation into the genome of Pichia pastoris GS115 cells.
