and elastase, go from peritoneum to the pleural cavities and never from thorax to the peritoneum. The intraperitoneal injections of collagenase or of collagenase and

can also cause polyseritis in the rat; but more often they provoke heavy hemorrhagic lesions of the wall of the abdomen and the diaphragm. Perforations of the diaphragm were observed in 8 rats of 32, with in 6, a intrathoracic
hernia
of the liver or the stomach. After the intrapleural injection of collagenase or of collagenase and
trypsin
, hemorrhagic lesions were seen in the thorax, but not in the abdomen. These facts are a new proof for the transdiaphragmatic propagation of the proteasic solutions injected in the peritoneum.

An experimental model of disc herniation in tail discs of rats is described. Constant result on nucleus
hernia
and intervertebral narrowing were obtained by an easy manipulation on numerous rats. Intradiscal injection of aprotinin produced a widening of the disc height. Trypsin, collagenase, chymopapain, and hyaluronidase induced a narrowing of disc height;
trypsin
induced macroscopic necrosis of the soft surrounding tissues; and collagenase had a destructive effect on nucleus pulposus, annulus fibrosus, and even on end-plates. Chymopapain and hyaluronidase acted mainly on nucleus pulposus. Hyaluronidase could be of interest as a nucleolytic drug and needs further studies on optimal dosage and lack of side effects in the surrounding tissues before injecting it into human discs.

Fibroblasts were derived from dermis and scar of a 47-year-old white man with a recurrent incisional
hernia
as a result of fractured ribs. The scar was thin and stretched, suggesting a defect in the maturation of granulation tissue. After surgical repair, biopsy specimens of discarded scar and skin were used to generate fibroblast cell lines. Fibroblasts maintained in medium containing 10% fetal bovine serum and antibiotic were studied between their third and eighth passage. By phase contrast microscopy, no structural differences were obvious, but it was noted that to pass scar fibroblasts, a more aggressive
trypsin
regimen was required. Immunohistologic and Western blot analysis of patient scar fibroblasts showed (1) more a smooth muscle actin within stress fibers, (2) increased expression of the vitronectin integrin receptor alpha(v) (CD 51), and (3) reduced expression of the collagen integrin receptor alpha2 (CD 49b). The expression of vinculin from focal adhesions or a tubulin from microtubules was the same among cell lines. Contractions of scar and dermal fibroblast-populated collagen lattice were compared. At 24 hours, contractions were 69 percent with newborn fibroblasts (normal); 68 percent for patient dermal fibroblasts; and only 48 percent for patient scar fibroblasts. The retarded contraction of scar fibroblast-populated collagen lattice was significant (p > or = 0.002). Myosin ATPase activity, critical for lattice contraction, and cell migration were equivalent among all cell lines. A plausible mechanism for the retardation of scar lattice contraction is disruption of fibroblasts and collagen interactions, for which the attachment of cells to collagen is altered. It is proposed that either the decrease in the expression of collagen integrin receptor alpha2 (CD 49b), an increase in the expression of the vitronectin receptor alpha(v) (CD 51), or a combination of both is responsible for disruption of collagen fibroblast interactions.

It has been found that
trypsin
and alpha-chymotrypsin inhibitors isolated from Ascaris suum act embryotoxically and teratogenically on White Leghorn chicken embryos. Mortality rate for the chicken embryos on day 15 of incubation was 45.0 +/- 3.5% after injection of trypsin inhibitor and 44.0 +/- 3.5% after adminstration alpha-chymotrypsin inhibitor. Gross examination of surviving embryos and their dissection revealed pathological changes (abdominal dropsy, umbilical
hernia
, subcutaneous oedema, hemoperitoneum, hemopericardium), symptoms indicating retardation in growth (lack of down, retarded ossification of long bones, decreased mean body weight) as well as malformations (schistocelia, micrognathia, cyclopia, crossed beak, cranial deformities) after injection of inhibitors from Ascaris. The highest incidence of embryos with pathological changes and malformations was found after administration of alpha-chymotrypsin inhibitor. The most commonly occurring abnormality was schistocelia (21.4 +/- 3.88%). Growth malformations were not found in the control groups. The
trypsin
and alpha-chymotrypsin inhibitors present in Ascaris homogenate have a significant disturbing effect on the development of the chicken embryo.

A 2-month-old entire female domestic short-hair cat (Felis domestica) with no history of trauma was presented for assessment of a swelling on the left thoracic wall. Palpation revealed a large, painless, reducible swelling between the tenth and eleventh ribs on the left side. Radiograph demonstrated dorsal displacement of the abdominal viscera through the tenth intercostal space. An abdominal ultrasound examination confirmed the displacement of stomach and spleen through tenth intercostal space. Surgical correction of the herniated contents was undertaken via intercostal celiotomy. An acellular dermal matrix scaffold, prepared from deceased donor caprine-skin upon treatment with 0.25%

in 4 mol/L NaCl for 8 hours followed by 2% sodium dodecyl sulfate for 48 hours, was used to repair a 3 cm wide intercostal defect present between the tenth and eleventh ribs.
