We established a model of orthotopic transplantation of human hepato-carcinoma. The tissues from human hepatoma LTNM4 were surgically implanted into the livers of 32 BALB/c nu/nu nude mice. These led to the development of tum or masses in the livers of 31 nude mice; the rate of success was about 96.8% (31/32). Furthermore, this model was applied to the studies on the tumor inhibitory action of DHAQ-PBCA-NS, a hepatic targeting preparation for anticarcinoma. Satisfactory effect was attained. The result has proved the applicability of this animal model to researches on hepatic targeting preparations against carcinoma.

Hydroxyapatite (HA) bioceramics is a biomaterial used in orthopedic area. We observed its anticancer effect in experimental hepatoma-22 (H22) in mice. BALB/c mice implanted with H22 were grouped into I, II, III and IV. Intratumoral injections were given as follows: (I) HA 0.1 g and ADM 0.1 mg n = 28; (II) HA 0.1 g n = 16; (III) ADM 0.1 mg n = 16; (IV) untreated n = 20. The tumor regression rates of the groups I, II and III were 65.9%, 41.9% and 51.1%, respectively. The mean survivals of groups I, II, III and IV were 32, 22, 26 and 18 days. Six mice of group I were alive more than 50 days before being killed, in which 4 tumor samples showed that no residual tumor cells could be seen under microscope. The possible mechanism of HA anti-cancer action is: (1) the dissolved matter of HA, glycoprotein and microfibril around the tumor cells may disturb the metabolism of cancer; (2) fibrocalcific process restricts the tumor growth; (3) immunoenhancement action (4) as a drug-release system HA is a hopeful agent of intratumoral injection for treatment of cancer.

An ultrastructural comparison between 15 cases of hepatocellular carcinoma (HCC) and 5 cases of human embryonic liver (HEL) was performed. There were similarities. Both of them are polygonal in cell shape with a central nucleus and are abundant in RER and mitochondria; both have laminating form of RER and formation of bile capillary and perisinusoid space. These are important evidences for diagnosis of a tumor originating from hepatic cells. On the other hand, obvious differences were present between HCC and HEL. Cancer cells possess certain characteristics; they are pleomorphic and heterogeneitic; the differentiation of their ultrastructural components is not synchronic, and they are invasive in growth. These enable us to suspect the malignancy of a tumor. In addition, single death cancer cells with pyknotic nucleus were seen in HCC; some of them were phagocytozed by adjacent cells. We conside these are apoptosis even no formation of apoptoic bodies was found. In the majority of HCC cases, lymphocyte-mediated cancer cell lyses by both T-lymphocytes and large granular lymphocytes (nature killer cell) were found.

The aim of this study was to evaluate oral glucose tolerance test(OGTT)in the assessment of reserved function of liver for predicting the tolerability of patients to hepatectomy and hence provided a criteria for selecting the candidates for undergoing hepatectomy, since the majority of hepatocellular carcinoma (HCC) patients were associated with posthepatitis cirrhosis. The preoperative and postoperative OGTT and liver biopsy for pathological investigation were carried out in 62 cases of hepatecomized patients and 49 cases of unresected patients for comparison. The results revealed that the patients whose preoperative OGTT curve was of P type recovered uneventfully after hepatectomy, but those whose curve was of L type of tolerated poorly to hepatectomy and were liable to postoperative hepatic failure and complications. The severity of cirrbosis in those poor risk patients fell to C III or C IV histological degree. 29 patients with intermediate feature of OGTT curve between P type and L type, i.e. I type underwent regional vascular occlusion at hepatic hilus as hepatectomy, and infusion of Danshen extract solution before vascular occlusion to prevent hepatocytes from reperfusion injury. Of them, 20 recovered uneventfully, 8 suffered from complications such as ascites and/or juandice, and 1 died within 1 month after operation. The followup study showed that the survival time of patients with P type OGTT curve was longer than that of I type, and the latter was longer than that of L type. The pattern of OGTT curve could change from preoperative P type to postoperative L type, depending on the severity of vascular interruption of liver and the ischemic injury to hepatocytic mass in operation.

The levels of serum acidic isoferritin (SAIF) in 48 patients with hepatocellular carcinoma (HCC), 30 patients with hepatitis, 28 patients with liver cirrihosis, and 33 healthy subjects were measured by using enzyme-linked immunosorbent assay (ELISA--double-determinant). The result revealed that the SAIF values of HCC, HP and LC did not show normal distribution. The median values were 440 micrograms/L, 21 micrograms/L, 120 micrograms/L and, in normal subjects, 66 micrograms/L respectively. There were statistically significant differences between HCC and the control groups. The sensitivity of SAIF to HCC was 85.46%, with the cut-off point being 250 micrograms/L. SAIF was not correlated to AFP in HCC cases. The values of SAIF had no relationship with the volume of the tumors and the clinical stages. SAIF may be an available and useful serum marker and be beneficial to the diagnosis of early-staged HCC.

This study screened 32 cases of hepatocellular carcinoma (HCC) from Chengdu detecting HBV DNA. HBsAg and p53 mutations by using Southern blot hybridization, immunohistochemistry and polymerase chain reaction/restriction enzyme digest methods, respectively. The results revealed all the cases had been infected by HBV; the frequency of HBV DNA integration into HCC cell was 72%, and the positive staining for HBsAg 96.3%(26/27). Of the 32 cases, 8 showed nuclear staining of p53 protein (25%), no mutation of p53 gene at all in 27 nontumorours liver tissues was identified. The resluts suggest the inactivation of p53 function may play a significant role in the genesis of HBV-associated HCC; however, the largely negative p53 mutation results in the current study conversely indicate that hepatocarcinogenesis may even involve other comprehensive mechanisms. Further studies are worth doing to evaluate the possible contribution of HBV to the p53 mutation in HCC.

To investigate the role p15 gene plays in the pathogenesis of human primary hepatocarcinoma, 35 human hepatocarcinomas, 35 cases of adjacent non-cancerous liver cirrhosis and the blood cells of 10 normal human were analyzed for somatic mutation in p15 gene with PCR-SSCP. One case of adjacent non-cancerous liver cirrhosis showed abnormal migration single strand. In the hepatocarcinomas and in the other cases of adjacent non-cancerous liver cirrhosis, no mutation was found. Cloning and sequencing of the amplified abnormal migration single strand DNA revealed that it contained a wild type exon 2 of p15 gene in 345 bp length. The results indicate that the inaction of p15 gene by point mutation is a very uncommon event in human hepatocarcinoma.

Hepatitis B and C viruses (HBV, HCV) are closely related with hepatocellular carcinoma(HCC). The association between the new discovered GB-virus C (GBV-C) and HCC has not yet been known. In this study, 124 HCC patients were detected for the prevalence of GBV-C RNA by the one-step nested reverse transcription polymerase chain reaction (RT-PCR) and followed by hybridization using GBV-C probes located at 3'-untranslated region (3'-UTR) from its reported genomes. The results showed that 33 of 124 (26.6%) HCC cases were GBV-C RNA positive, including 12 cases positive HBsAg and anti-HCV, and 3 for cases negative HBsAg and anti-HCV. The clinical background of the patients with HBsAg and/or anti-HCV who were also positive for GBV-C RNA did not differ from the background of those who were negative for GBV-C RNA except the ratio of blood transfusion history. In conclusion, GBV-C has a high prevalence in Chinese HCC patients. Even though no sufficient data supports the causality of GBV-C on hepatocarcinogenesis, further researches aimed at whether GBV-C infection aggravates the incidence of HCC are warranted.

In the present study, the relationship between PKB signaling and reactive oxygen species (ROS) during the course of exogenous and endogenous ROS or antioxidants regulating human 7721 hepatoma cell proliferation was studied. To change endogenous ROS levels, 7721 cells were transfected with human manganese superoxide dismutase (MnSOD) construct containing sense or antisense MnSOD cDNA. Low level of exogenous ROS H2O2(1-10 mumol/L) significantly stimulated PKB activity and c-fos/c-jun expression and cell growth, which could be abolished by antioxidant danshensu (40 mg/L). It was observed that overexpression of MnSOD inhibited 7721 cell growth by inhibiting PKB activity and c-fos/c-jun expression; the PKB activity and c-fos/c-jun expression, however, were stimulated by down-regulated MnSOD expression. In addition, PKB-7721 cells (transfected with sense PKB cDNA) promoted c-fos/c-jun expression by stimulating PKB activity. These results suggest that the redox state stimulated hepatoma cell growth through PKB pathway, which modulates AP-1 expression.

A novel transcript of human HPO (HPO-205) encoding a 205 amino acid ORF corresponding to a translated product of 23 kD different from the previous HPO that lacks the N-terminal 80 amino acids, was isolated from human fetal liver cDNA library by 5'RACE methods. The dose-dependent stimulation of DNA synthesis of HepG2 hepatoma cells by HPO-205 demonstrated its similar biological activity with HPO in vitro. The level of MAPK (mitogen-activated protein kinase)phosphorylation was further examined by Western blot analysis and the result revealed that HPO-205 protein might have stronger activity on stimulating hepatic cell proliferation than that of HPO. The similar result was observed by FACS technique. Therefore, our results suggest that the HPO-205 has activity to stimulate proliferation of liver derived cells, and this activity may be stronger than HPO.
